The following is a modified procedure: To a vigorously stirred solution of Br2 (24 gm) in petroleum ether (100 ml, b.p. 40-60 0C), PBr3 (48 gm) was added. After EPO <DP n="30"/>stirring for a few minutes at room temperature, the resulting PBrs was washed several times with petroleum ether by decantation and dried in vacuo. 4-hydroxy- 2,6-pyridinedicarboxylic acid (10 gm) was added to the same reaction vessel and, after thorough mixing, the temperature of the bath was raised to 9O0C and maintained at that temperature for 3 hrs. The cooled mixture was stirred with CHCl3 (75 ml) and Filtered through a Whatman filter paper. Absolute ethanol (200 ml) was added to the filtrate in small portions and the solution was concentrated in vacuo, untill it looked a little bit viscous. Immediately after, when it was still hot, while evaporating much of the ethanol in rotary-evaporator at 60° C to make it concentrated, small pieces of ice were added slowly into it untill white fluffy Intermediate 1 started separating out. The crystallization of Intermediate 1 from the mother liquor was even better and faster when the concentrated solution, after adding few pieces of ice, was seeded with 10 mg of Intermediate 1 prepared before. For the completion of this process it was left in the freezer at 0 0C for one night and next day morning it was filtered off under vacuum. LTD.; WO2006/80022; (2006); (A2) English View in Reaxys 2 :In a first step, PBr3 (F.W.=270.7 g/mol, d=2.852 g/ml, 0.1788 mol=48.4 g or 17 ml) was added to a vigorously stirred solution of bromine (0.1476 mol=23.6 g or 7.68 ml) in petroleum ether (50 ml). After stirring the mixture at room temperature for one hour, PBr5 was obtained in pure form by removing excess petroleum ether by decantation after repeated washings. After drying in vacuo, chelidamic acid (54.6 mmol=10 g) was added and the resulting solid mixture was heated to 90° C. for 3-4 hours. After cooling down to room temperature, 20 ml of chloroform was added, and the mixture was stirred for 30 minutes and then filtered (1H-NMR (CDCl3): δ 8.38 [2H, s]).
