2.3 General procedure for ω-transaminase catalyzed reactions

General procedure: Sodium phosphate reaction buffer (50mM, pH 7.5) containing 0.1mM PLP was mixed with the ketone substrate (10 or 50mM) and the appropriate amino donor (0.1–0.5M). The pH was adjusted to 7.5 with phosphoric acid before adding the enzymes. The reactions were started by adding 50μl of cell-free extract (CFE, contains approximately 2mg of protein) and then shaken at 30°C for 24h. 2:Step 3-(-)-amphetamine

Step 3-(-)-amphetamine A suspension of the sodium acetate, 5percent palladium on Barium sulfate and racemic norchloroephedrine hydrochloride in 186 mL glacial acetic acid and 9.7 mL deionized water was hydrogenated at 50 psi for 72 h. The reaction was then filtered through celite and the celite washed with water. The majority of the acetic acid was removed in vacuo and the pH adjusted to 10 with 10percent sodium hydroxide solution. After extracting three times with ethyl ether, the combined organics were dried over anhydrous sodium sulfate. The ether was then removed by fractional distillation at ambient pressure, followed by fractional distillation at 1 mm Hg to yield 11.0 g (74percent) of (-)-amphetamine at 99percent purity by Gas Chromatography.
