Co-Immunoprecipitation and Pull-down Techniques

Page 1

Two technologies that detect proteins and protein interactions are introduced in this infographic: Co-Immunoprecipitation (Co-IP) and Pull-Down Assays. Co-IP is a useful in vitro method to evaluate proteins involved in the protein complex and the protein binding affinity.

Co-IP enables the capture and purification of the primary target and other macromolecules involved in the interactions. The pull-down assay can detect a physical interaction between two or more proteins and identifies previously unknown PPIs. In a pull-down assay, a bait protein is tagged and immobilized to affinity resin. When a sample incubates with the bait proteins, proteins binding to the bait protein will be captured and "pulled down".

The principles and characteristics of co-immunoprecipitation and pull-down techniques are compared.
