IMPORTANCE OF AI IN THE INDIAN API INDUSTRY

Artificial intelligence (AI) represents a potent tool that leverages human like knowledge to address complex challenges efficiently. Significant progress in AI technology and machine learning offers a transformative opportunity in pharmaceutical research, including drug discovery, formulation, and testing. By employing AI algorithms to analyze vast biological datasets such as genomics and proteomics, scientists can pinpoint diseaserelated targets and forecast their interactions with potential drug candidates. This streamlined approach enhances the efficiency and precision of drug discovery, boosting the likelihood of successful drug approvals. Additionally, AI can cut down on development costs by optimizing research and development workflows. Machine learning algorithms aid in experimental design and forecast the pharmacokinetics and toxicity of drug candidates, enabling the prioritization and refinement of lead compounds while minimizing the need for extensive and expensive animal testing.
AI algorithms analyzing realworld patient data can facilitate personalized medicine approaches, resulting in more effective treatments and better patient compliance. This comprehensive overview explores AI's broad applications in drug discovery, drug delivery system designs, process optimization, testing, and pharmacokinetics/pharmacodyna mics (PK/PD) studies. In January 2024, Siemens Healthineers and the Indian Institute of Science (IISc) jointly established the Siemens HealthineersComputational Data Sciences (CDS) Collaborative Laboratory for AI in Precision Medicine at IISc Bangalore.
The CDS collaborative laboratory was dedicated to developing opensource AIbased tools that would automate the segmentation of pathological findings in neuroimaging data, specifically concentrating on diagnosing neurological diseases accurately and analyzing their
clinical impact at a population level. Further, in December 2023, Peptris Technologies, headquartered in Bangalore, India, secured USD 1 million in funding during a pre seed investment round led by Speciale Invest. This investment represents a significant milestone for the company as it aims to enhance its initiatives in AIbased solutions for drug discovery and development, particularly concentrating on creating new treatments for challenging medical targets in areas like oncology, inflammation, and rare diseases.
The Active Pharmaceutical Ingredient (API) is a critical sector within the pharmaceutical industry, representing approximately 35% of the market (Invest India). API refers to the biologically active substance in a drug that produces the desired medical effect. According to the World Health Organization, an API is any substance or combination of substances used in a final pharmaceutical product (FPP) intended to provide pharmacological activity or to directly affect the diagnosis, cure, mitigation, treatment, or prevention of disease, or to promptly restore, correct, or modify physiological functions in humans.
India ranks as the thirdlargest producer of APIs globally, accounting for an 8% share of the industry. Over 500 different APIs are manufactured in India, with 57% of these APIs included in the World Health Organization's prequalified list. The Indian API market is projected to grow at a Compound Annual Growth Rate (CAGR) of 13.7% over the initial four years, which is approximately 8% higher than the generic API industry (Invest India). The Indian API sector has become attractive to various investors and venture capitalists due to India's strong domestic market, advanced chemical industry, skilled workforce, strict quality and manufacturing standards, and lower costs (about 40% less than in the West) for establishing and operating modern facilities.
AI, in conjunction with big data and ML, represents the future of the pharmaceutical industry. It covers all aspects of
pharmaceutical operations, from drug research and production to marketing and supply chain management. AI applications are proving to be essential in reducing costs, supporting a profitfocused innovative ecosystem, and boosting productivity, resulting in improved business outcomes, as more Indian firms embrace digital technologies.
Indian companies are harnessing AI's power by analyzing extensive data sets to enhance business processes with insightful findings. This approach enhances the success rate of new drugs through quality control and addresses supply chain issues to minimize waste in production. AI tools also decrease human error, ensuring greater efficiency, and cutting operational costs through predictive maintenance, facilitating the development of more costeffective drugs. Clearly, AI driven analytics can significantly enhance the value proposition by fostering innovative business models.Pharmaceutical firms are increasingly using AI to streamline drug discovery by simplifying the identification of complex patterns from datasets. This aids in evaluating drug candidate safety and efficacy, solving problems involving intricate biological networks. AI's predictive and analytics capabilities help researchers assess new drug implications, benefits, and success rates by analyzing dataset patterns.
Additionally, machine learning plays a critical role in realworld evidence studies and clinical trials using techniques like few shot learning, inferring results based on limited data to test experimental drugs and identify candidates with high potential. These interventions reduce reliance on large datasets and expedite the drug discovery and development process.AI extends beyond drug discovery to improve marketing campaigns by offering measurable results and predicting the success of marketing activities accurately. Leveraging artificial intelligence during production can enhance product stability by identifying and rectifying errors promptly. This approach aids in safeguarding the integrity of the final product's active pharmaceutical ingredients against manufacturing process variations.
The Development Path of Indian Pharmaceutical Research and Development: Obstacles and Opportunities.
The Indian pharmaceutical sector is widely recognized for its production of affordable generic drugs and vaccines, positioning it third globally in terms of output volume. It leads in the global generics market, accounting for more than 20% of global generic supply by volume and meeting around 60% of global vaccine demand.Despite this highvolume ranking, the industry ranks 14th globally in terms of the value of its pharmaceutical production. Since the 1990s, Indian pharmaceutical companies have been investing in their own research and development (R&D) efforts, identifying over 200 compounds in preclinical and clinical stages of development. However, only a few of these have successfully made it to market. For instance, Biocon was a pioneer in introducing novel biologics developed in India, and recently, the Central Drugs Standard Control Organization (CDSCO) approved ImmunoACT's CART cell therapy as India's first domestically developed treatment of its kind.Despite having a strong network of pharmaceutical firms, extensive manufacturing capabilities, and a large pool of scientists and researchers, Indian companies have yet to make a significant impact in discovering new blockbuster drugs.
Indian pharmaceutical companies face challenges in advancing drug discovery primarily due to resource limitations when competing against major
companies in the United States and Europe. Another hurdle is the struggle to progress beyond Phase 2 clinical research in certain initiatives.Although most new drugs globally originate from the United States and Europe, China has recently made significant strides in this area. Notably, Hong Kong based biotech startup Insilico Medicine achieved a milestone by developing the first fully AIgenerated drug to enter Phase II clinical trials with patients in June 2023. Insilico leveraged AI extensively in the preclinical drug discovery process, including molecule target identification, generation of novel drug candidates, assessment of binding efficacy, and prediction of clinical trial outcomes. This approach drastically reduced costs and time compared to traditional methods, completing milestones in two and a half years that would typically take six years and cost over $400 million.Insilico utilized a Gen AI platform with multiple AI models, each trained on vast datasets to perform various tasks. Notably, the PandaOmics tool efficiently identified diseaserelated targets, such as the spike protein linked to COVID 19. The Chemistry42 engine, another critical AI component, designed potential drug compounds tailored to these identified proteins using deep learning techniques to create molecular structures from scratch.

Government initiatives
The integration of AI into the healthcare industry will significantly impact various factors
such as cost, quality, efficiency, and accessibility of healthcare services to underserved areas. The primary goal is to extend healthcare services to regions lacking infrastructure or where primary healthcare quality is in question. However, the complete replacement of physicians and doctors by automation remains a contentious topic that will be addressed socially in the future.Moreover, AI's broader impact in India is directed towards addressing issues related to economic inequality. Government initiatives include collaborations between the National Institution for Transforming India Aayog (NITI Aayog) and Google to promote AI research, thereby contributing to a technologically empowered New India (NITI Aayog, 2018). Additionally, NITI Aayog is spearheading a national effort to implement an analytical portal with AI to transform the National Health Scheme (NHS) into a centralized health record system, facilitating effective health information management through big data analytics and machine learning (Dhupar, 2017).
The Indian government is actively pursuing initiatives to amplify AI's impact on healthcare to improve health quality in rural India. The Ayushman Bharat National Health Protection Scheme aims to deliver quality and affordable healthcare to all citizens, making it the world's largest healthcare scheme and addressing challenges in database management and quality control through AI, deep learning, and machine learning technologies.In August 2023, a comprehensive five year policy aimed at enhancing research and development (R&D) in the pharmaceutical and medtech sector was introduced by the Health Ministry, as per top government sources. This initiative involved an allocation of Rs 5,000 crore, spanning from FY2428. The primary goal of the scheme was to propel the value of the pharmaceutical industry from USD 50 billion to USD 130 billion by 2030. PRIP Scheme for innovation
The Indian government aims for significant expansion in the pharmaceutical industry, targeting a USD130 billion milestone by 2030 by boosting research and development (R&D) and fostering innovation. Recent efforts such as the National Policy on Research and Development and Innovation in the PharmaMedTech Sector, alongside the Promotion of Research and Innovation in Pharmaceuticals and Medical Device (PRIP) scheme, are designed to promote R&D in critical sectors. The PRIP scheme, backed by a financial allocation of INR 5,000 crore over five years, seeks to cultivate a supportive R&D environment within India.

IMPORTANCE OF AI IN INDIAN PHARMA INDUSTRY

Introduction
There could be no development without invention. For the pharmaceutical sector to continuously advance healthcare, innovation is crucial. Manufacturers are able to develop medication on a commercial scale to better meet the public's health requirements because of pharmaceutical innovation. Research and development of novel active chemicals has long been the only focus of innovation in the pharmaceutical sector; in the meantime, batchwise methods continue to dominate production structure.
Batch procedures are not as advantageous as continuous manufacturing (CM), as several other industrial industries have previously shown. With a far greater degree of quality assurance, manufacturing may be designed that is faster, less expensive, and more adaptable. The major regulatory bodies have been encouraging pharmaceutical businesses to develop and modify continuous technologies since they realised that medication manufacture needed to alter in recent years. Because of this, a great deal of study has been done in the disciplines of pharmaceutical technologies up to this point, ranging from the manufacture of drug substances to drug products. Numerous articles discuss continuous crystallisations and synthetic stages conducted in flow reactors. Continuous filtering, drying, granulation, and blending have all been researched in some capacity on the formulation side. Furthermore, in addition to converting existing conventional processes to continuous operation, new, inherently continuous technologies are also being researched. The mostly independently created processes must be combined to construct endtoend systems that include raw materials and final dosage forms in order to fully use the benefits of continuous manufacturing. Even yet, it's a difficult effort to integrate two technical advances. Deep process
knowledge and an allencompassing approach to process development and optimisation are necessary for the creation of endtoend systems.
Past perspectives
Lyophilization
While AM advances mostly concentrate on solid oral dosage formulations of small molecule APIs, injection or infusion are the primary methods of administering the vast majority of largemolecule based treatments. According to Alexeenko and Topp (2020), lyophilization has been necessary for over half of the recently authorised injectable or infusible pharmaceuticals in order to assure product stability. Lyophilization, as it is now done, is a very ineffective batch procedure that involves sublimating a solvent—typically water—at low pressure and temperature in order to extract the solvent from a liquid formulation. There is strong motivation to develop and implement new technologies due to the restricted capacity for lyophilization on a worldwide scale and the growing quantity of items that necessitate it. The LyoHUB consortium released a technological roadmap towards the end of 2017 that fills in the gaps in process technology, product design methodologies, and process equipment.
A technique that prints an APIcontaining solution onto an excipient tablet and then dries the solution to produce the dosage form is an example of the liquid solidification approach (Clarke et al. 2017). Although the manufacturing process has been
widely used, no product that makes use of it has received approval as of yet. There have been reports of research successes utilising alternate fluid formulations, such as polymer melts and API powder suspensions (Içten et al. 2017; Radcliffe et al. 2019). Drying to extract the solvent or chilling to solidify the fluid formulation and create a stable dosage form always results in the production of a solid. Nevertheless, no authorised products have been developed using the liquid solidification approach.
AM techniques that rely on extrusion, particularly those that employ drug polymer filaments (Awad et al., 2018), are among of the most widely documented techniques. They can benefit from a large variety of commercially accessible 3 D printers that are capable of using thermoplastic polymeric filaments. A filament made of an acceptable polymer—a drug combination with the required API composition and mechanical or rheologic properties—must be manufactured in order to generate a dosage form. That method necessitates a preliminary hotmelt extrusion process.

With the Indian pharmaceutical industry expected to reach a valuation of $130 billion by 2030, its unwavering dedication to the advancement of medical knowledge highlights its critical role in determining the future direction of healthcare. Industry 4.0's vertical integration will transform the pharmaceutical manufacturing facility into a "reconfigurable factory," with a highly adaptable, nimble, and intelligent production line that can handle mass customisation of customised medications to meet various needs. Effective crossorganizational communication combined with big data analytics may enhance process monitoring capabilities and detect and minimise material waste, overproduction, and energy usage.
Augmented and Virtual Reality
AR and VR form an integral part of future industry, helping manufacturing companies to face challenges related to volatile demand and changing requirements from customers and suppliers. The literature identifies the primary advantages of AR and VR applications as follows:
• Production plant control and error diagnostics
• Production system safety and security
• Product design and reconfiguration
• Provision of required information at operational and enterprise level
• Complex task training to increase safety and productivity
The concepts of AR and VR have existed for more than 20 years, but these technologies have only recently entered the industrial environment. VR applications in pharma allow companies to decrease design and production costs, maintain product quality, and reduce the time needed to go from product concept to production.
AR, defined as a system “in which 3D virtual objects are
integrated into a 3D real environment in real time,” can “increase the operator’s natural feedback with virtual clues.”In other words, AR enables users to visualize and interact with 3D objects, in the real environment, more easily than they can through a simulation or a computer screen.
AR is recognized as a support for industrial applications including product assembly, improving assembly quality and efficiency, and reducing assembly costs and maintenance. However, AR visualization tools must be improved to allow for seamless and complete integration into manufacturing, as current technology is not suitable for continuous use and harsh working environments, such as those found in the pharma industry.
3D Printing of pharmaceutical industry
3D printing has had a big impact on the pharmaceutical industry since it makes it possible to create customised medications that are suited to each patient's particular requirements. 3D printing provides an alternative to traditional pharmaceutical production, which usually produces standardised dosages that might not be appropriate for particular populations, such as youngsters or the elderly. Pharmacists and other medical professionals possess the capacity to formulate and dispense drugs with precision. This breakthrough enables the production of tablets with controlled drug release, guaranteeing optimal therapeutic results. This technology is especially helpful for people who have trouble swallowing medicines or who need dosages that are not typical.
Digital Twin and Simulation
A digital twin is the virtualized version of a real system or product. It may be thought of as a virtual reflection of reality. It can
offer information on motions, machine functions, and process parameters in real time, in simulation, or in the past. Using a digital twin in realtime simulation reduces setup times for machines and improves quality by enabling testing and process optimisation before a job is carried out. Benefits of the digital twin include shorter cycle times for process development and cycle times for cycle time and system confidence, which in turn results in shorter regulatory deadlines. This is because the digital twin may be an exact copy of shop floor systems.
In summary, the introduction of artificial intelligence (AI) into the pharmaceutical sector signifies a radical paradigm change with broad ramifications. This analysis makes clear how artificial intelligence (AI) technologies have transformed several facets of medicine research and delivery. The drug discovery process has been greatly accelerated by artificial intelligence (AI) driven algorithms and machine learning models, which improve target identification, lead optimisation, and compound screening. Furthermore, through patient categorization, protocol optimisation, and real time monitoring, AI powered systems facilitate more effective clinical trials, which in turn lower costs and shorten the time it takes for novel treatments to reach the market. All things considered, the combination of artificial intelligence and medicines has great potential to transform healthcare delivery, stimulate innovation, and ultimately enhance patient outcomes. To fully realise the promise of this disruptive technology, a cautious yet aggressive strategy is necessary.

COLLABORATES WITH IVDP

Karkinos Healthcare a purpose driven technology led oncology platform in an impactful collaboration with the Integrated Village Development Project (IVDP), has concluded a groundbreaking series of cancer screening camps, marking a pivotal moment in the fight against cancer in underserved regions, held at the IVDP SHG Industrial Training Centre nestled within the SIDCO Industrial Estate, Gandhi Nagar, Krishnagiri, Tamil Nadu. These camps witnessed an inspiring turnout of women from rural areas engaged in various small businesses such as tea shops, tailoring shops, and catering ventures.
The demographic footprint of participants spanned across 16,000 selfhelp groups under the IVDP, originating from four districts and clustered into 82 distinct units. Over a span of 14 meticulously organized camps, conducted from
November 12, 2022, to February 1314, 2024, a staggering 10,738 registrations were tallied, with 10,101 individuals undergoing crucial HPV screening. Notably, the latest camp held on February 1314, 2024, recorded 456 registrations, and collected 456 cervical samples for HPV testing, marking yet another milestone in this transformative journey.
Dr. R. Sankaranarayanan, Director of Preventive Oncology at Karkinos Healthcare emphasized the profound impact of community focused healthcare interventions in the battle against cancer. "Initiatives such as the IVDP camps underscore the critical importance of communityfocused healthcare interventions in combating cancer.
