Opportunities in US generic injectable market he Indian generic players have better opportunity in US generic injectable market with upcoming patent expiries, drug shortages, quality manufacturing and development resources, says a report by ICRA. Indian companies are increasingly focusing on complex generics and some of the leading Indian companies have carved a niche for themselves through the launch of differentiated and complex filings. The financial profile of generic injectable players in the US is characterized by healthy operating margins, RoCE, working capital intensity and moderate debtprotection metrics. The growth rate in CY2015 and CY2014 improved to 15.7 per cent and 13.3 per cent respectively, compared to over 7 per cent in CY2012 and CY2013 each. During the 2015-19 period injectables drugs worth US$ 16 billion are expected to go off-patent in the US alone. Key patent expiries for the US market would include Alimta (oncology in 2016 with CY2015 revenues of above $1 billion and Levemir (anti-diabetic in 2019 with CY 2015 revenues of above $2 billion. While significant price erosion is normal once the drugs go offpatent, the injectable segment still holds a sizeable opportunity to gain meaningful revenue growth for Indian generic companies on account of the upcoming patent expires. The US generic injectable market to grow at a CAGR of 10 over the next five years led by significant number of drugs going off-patent as well as supportive price environment in the USA. Over the past few years, there have been a sizeable number of acquisitions in the US generic injectable space. The acquisitions were driven by the need to acquire product portfolio, development

pipeline and manufacturing capabilities of existing injectable players to consolidate market share or accelerate entry into the promising US sterile injectable market. The continuing trend in M&A to acquire development and manufacturing capabilities for complex injectables/novel drug delivery systems. Notwithstanding the various factors, the drug shortages for injectables in the US have been fuelled by shifting of old molecules production to low cost countries such as India and China making high cost US facilities cost-inefficient, reduction in the number of players owing to M&A and subsequent closure and consolidation among such facilities in addition to phasing out of low value low margin products. Also stringent action by US FDA regarding compliance to GMP led to several warning letters and supply disruptions for injectable drugs is another major reason for drug shortages. Moreover, ongoing investment in maintaining quality standards not justified by margins offered by various Government-run programmes such as Medicare, also led to rationalization of product portfolio for several players. Drug shortages led to improved pricing power for the industry and reflected in the contrasting volume and value growth during the 2008-2013 period. During the period 2008-13, the injectable generic market value grew at a CAGR of 19.6 per cent, against volume growth during the same period of only 1.6 per cent implying a health pricing environment. The number of drugs in shortages in the US swelled to 251 in CY2011, of which 73 per cent were for injectable products. However, due to action taken by the

US FDA, since 2012, there have been drug shortages. For the ninemonth period ending September 2015, 22 new drugs (all forms) were reported to be in shortage, excluding 48 drugs, which continued to be in shortage, taking the total tally to 70. So, the injectable space is expected to continue enjoying a supportive pricing environment' albeit at a lower pace, due to the moderating trends in shortages. Issues with US FDA cGMP norms have led to demand for good quality facilities and contract manufacturers based out of India, China to support large volumes at low cost. Approximately 65 per cent of the 117 drug shortages in 2012 were on account of quality manufacturing/delays or capacity constraints â&#x20AC;&#x201C; thereby making it one of the critical success factors. Complex injectables include lyophilised products, high potent drugs, long-acting suspensions, liposomes, which require complex manufacturing capabilities or use of new drug delivery systems (NDDS). The complex development and manufacturing process of sterile injectables along with high capital and operational costs involved coupled with high compliance cost has led to relatively consolidated market. The market has seen further consolidation due to M&A activities within the sector over the last few years. Hence, the complexities involved in manufacturing of injectables have led to smaller number of players which compared to Oral Solids Generics Industry. A new player looking to gain entry into the US generic injectable market needs to have a relatively large basket of products and low cost advantage to gain traction with customers.

NEWS Biocon net surges by 17% to Rs.147 crore in Q1, scrip touches yearly highest level at Rs.786 Bangalore : Biocon, a Rs.3,500 crore major biopharmaceuticals company, has posted satisfactory financial performance during the first quarter ended June 2016 and its consolidated net profit as per IGAAP improved by 17 per cent to Rs.147 crore from Rs.126 crore in the corresponding period of last year. Its total sales increased by 9.5 per cent to Rs.903 crore from Rs.825 crore. EBIDTA increased by 14.8 per cent to Rs.271 crore from Rs.236 crore with EBIDTA margins of 28 per cent. The company's gross R&D expenditure remained at Rs.92 crore for the quarter. The revenues from small molecules increased by 9 per cent to Rs.357 crore, led by sales to its India based customers servicing the needs of the US market and to some key markets in the LATAM and Africa & Middle East regions. The biologics business improved by 53 per cent to Rs.107 crore which include rh-insulin, insulin analogs, monoclonal antibodies and recombinant proteins.The sales of branded formulations increased by 15 per cent to Rs.159 crore. Its research services business through Syngene improved by 18 per cent to Rs.263 crore. Biocon scrip has taken a jump and reached at its yearly highest level at Rs.803.85 in the afternoon session on BSE today on account of satisfactory financial performance and

announcement of acceptance by European Medicines Agency for proposed biosimilarPegfilgrastim, codeveloped by Mylan and Biocon. KiranMazumdar-Shaw, chairperson and managing director, said, “Our strong performance this quarter has been driven by an all-round growth of our business across small molecules, biologics, branded formulations and research services. Our biologics business delivered a growth of 53 per cent driven by the sale of biosimilars in emerging markets. The submission of Pegfilgrastim, our first biosimilar filing in EU, isa a critical milestone this quarter. Our insulins business made a mark with the launch of Insulin Glargine in Japan. In addition we received regulatory approvals from MoH, Malaysia, for rh-insulin and Glargine which will enable commercialization of these products. We are on track for filing some of our biosimilars and generic formulations in the developed markets later this year.”

Syngene International net up by 17.5% in Q1 Mumbai : Syngene International, a Rs.1,000 crore plus company engaged in contract research and manufacturing services, has posted net profit growth of 17.5 per cent during the first quarter ended June 2016 to Rs.51.1 crore from Rs.43.5 crore in the corresponding period of last year. However, excluding other comprehensive income of Rs 8.7 crore (Rs 3.1 crore in the last period),

net profit growth worked out to 28.3 per cent to Rs 59.8 crore during the June 2016 quarter as against Rs 46.6 crore in the similar quarter of last year. Its net sales also moved up by 17.9 per cent to Rs.269.2 crore from Rs.228.3 crore. EPS improved to Rs.3.08 from Rs.2.41 in the last period. EBDITA moved up 30 per cent to Rs.105 crore from Rs.80 crore. Jonathan Hunt, CEO, said, “We are pleased to report a good start for the year with all our three verticals performing as expected. First quarter revenue grew at 23 per cent compared to last year, while EBDITA and PAT grew by 30 and 28 per cent respectively. Phase 3 study of ABP 980 in patients with HER2-positive breast cancer mgen and Allergan have announced results from a phase 3 study evaluating efficacy and safety of ABP 980 compared with trastuzumab in patients with human epidermal growth factor receptor 2-positive (HER2-positive) early breast cancer. The results ruled out inferiority compared to trastuzumab but could not rule out superiority based on its primary efficacy endpoint of the difference of the percentage of patients with a pathologic complete response (pCR). The primary endpoint had a prespecified equivalence margin of +/- 13 percent and the observed upper end of the confidence interval was 13.4 percent. Overall, adverse events were comparable between ABP 980 and trastuzumab. In the neoadjuvant phase of the study, which included chemotherapy, there were more serious adverse events reported in the ABP 980 group, the majority of which were reported by the investigators as unlikely related to investigational product. In the adjuvant phase of the study, which did not include chemotherapy, serious adverse events were comparable between treatment groups. The overall results also showed comparable immunogenicity. "We believe this study confirms no clinically meaningful differences between ABP 980 and trastuzumab, and we look forward to continued discussions with regulatory authorities," said Sean E. Harper, M.D., executive vice president of Research and Development at Amgen. "Biosimilars are approved based on the analytical, nonclinical and clinical data, and we believe that the totality of the evidence we've generated supports ABP 980 as highly similar to the reference product.""These results provide significant clinical evidence that ABP 980 could be an important biosimilar treatment option for patients with HER2-positive early breast cancer," said David Nicholson, chief research and development officer, Allergan. "Allergan is committed to the continued development of ABP 980 and other biosimilars that provide safe, highquality and effective therapies in key disease areas." ABP 980 is being developed as a biosimilar to trastuzumab, a recombinant DNA-derived humanized monoclonal immunoglobulin G1 kappa antibody which targets HER2. Trastuzumab is approved in many regions for the treatment of HER2-positive early breast cancer,

metastatic breast cancer and metastatic gastric cancer. Amgen and Allergan are collaborating on the development and commercialization of four oncology biosimilars. Amgen has a total of nine biosimilars in development. Allergan is also independently developing biosimilars.This above referenced phase 3 study was a randomized, multicenter, double-blinded, active-controlled study (study number 20120283) that evaluated safety and efficacy of ABP 980 compared to trastuzumab in adult female patients with HER2-positive early breast cancer. There were 725 patients randomized, with 364 patients in the ABP 980 group and 361 patients in the trastuzumab group.In the neoadjuvant phase, enrolled patients received run-in chemotherapy consisting of epirubicin and cyclophosphamide (EC) every three weeks (Q3W) for four cycles. Once run-in chemotherapy was completed, patients with adequate cardiac function were randomized 1:1 to receive investigational product (ABP 980 or trastuzumab), plus paclitaxel, Q3W for four cycles. Surgery (breast and sentinel node or axillary lymph node resection) was complete three to seven weeks after the last dose of investigational product in the neoadjuvant phase, and pCR was analyzed. In the adjuvant phase, following surgery, patients received investigational product (ABP 980 or trastuzumab) Q3W for up to one year from the first day of investigational product administered in the neoadjuvant phase. Patients who received ABP 980 during the neoadjuvant phase continued to receive ABP 980 Q3W for the adjuvant phase. Patients who received trastuzumab during the neoadjuvant phase received either ABP 980 or trastuzumab Q3W for the adjuvant phase. The allocation to a treatment group during the neoadjuvant or adjuvant phase occurred at randomization. The primary analysis was conducted when the last patient completed the surgery following the neoadjuvant therapy. Clinical equivalence was assessed by comparing the confidence interval of the risk difference and risk ratio of the pCR in breast tissue and axillary lymph nodes with the prespecified equivalence margins. The final analysis of safety will be conducted when the last patient has completed their last study assessment in

the adjuvant phase. HER2-positive early breast cancer is a breast cancer that tests positive for a protein called human epidermal growth factor receptor 2 (HER2), which promotes the growth of cancer cells. Approximately 20 percent of all breast cancers are HER2-positive. Breast cancer is the second leading cause of cancer death among women and each year it is estimated that over 230,000 women in the United States will be diagnosed. HER2positive breast cancers tend to grow and spread more aggressively than HER2negative breast cancers. ABP 980 is being developed as a biosimilar to trastuzumab, a recombinant DNA-derived humanized monoclonal immunoglobulin G1 kappa antibody approved in many regions for the treatment of HER2-overexpressing early breast cancer, metastatic breast cancer and metastatic gastric cancer. The active ingredient of ABP 980 is a humanized monoclonal antibody which has the same amino acid sequence as trastuzumab. ABP 980 has the same pharmaceutical dosage form and strength as trastuzumab (US) and trastuzumab (EU). Phase 3b studies evaluating Odefsey to treat HIV-1 infection meets primary objectives Gilead Sciences, Inc. announced that two phase 3b switch studies evaluating Odefsey (emtricitabine 200mg/rilpivirine 25mg/tenofoviralafenamide 25mg) for the treatment of HIV-1 infection met their primary objectives. The ongoing studies were designed to explore the efficacy and safety of Odefsey among virologically suppressed adult patients switching from the tenofovirdisoproxilfumarate (TDF)based regimens Complera (emtricitabine 200mg/rilpivirine 25mg/tenofovirdisoproxilfumarate 300mg) (Study 1216) or Atripla (efavirenz 600mg/emtricitabine 200mg/tenofovirdisoproxilfumarate 300mg) (Study 1160). Odefsey combines Gilead's emtricitabine and tenofoviralafenamide with rilpivirine, marketed by Janssen Sciences Ireland UC, one of the Janssen Pharmaceutical Companies of Johnson & Johnson. Odefsey maintained similar rates of virologic suppression as the TDF-based regimens in both studies based on the proportion of patients with HIV-1 RNA levels (viral load) <50 copies/ml. ARTICLE 48, virologic suppression was maintained in 94 percent of patients taking Odefsey and in 94 percent of patients taking Complera in Study 1216 (difference: -0.3 percent; 95 percent CI: -4.2 percent to +3.7 percent), and in 90 percent of patients taking Odefsey versus 92 percent of patients taking Atripla in Study 1160 (difference: -2.0 percent; 95 percent CI: 5.9 percent to +1.8 percent). Compared to the TDF-based regimens, Odefsey demonstrated statistically significant improvements in bone mineral density (BMD) at the hip and spine (p<0.001) in both studies. Additionally, improvements in total and tubular proteinuria statistically favored Odefsey in both studies (p<0.001). Study regimens were generally well tolerated, and general safety and discontinuation rates due to adverse events were comparable in the two studies. The most commonly reported adverse events for Odefsey included upper respiratory tract infection, diarrhea, nasopharyngitis, cough and headache. Gilead plans to submit these data for presentation at scientific conferences in 2016. Odefsey was approved in the United States on March 1, 2016, and is indicated as a complete regimen for the treatment of HIV-1 infection in patients 12 years of age and older who have no antiretroviral treatment history and HIV-1 RNA levels =100,000 copies/mL.Odefsey is also indicated as replacement for a stable antiretroviral regimen in those who are virologically suppressed (HIV-1 RNA <50 copies/ml) for at least six months with no history of treatment failure and no known resistance to the individual components of Odefsey. No dosage adjustment of Odefsey is required in patients with estimated creatinine clearance =30 mL per minute.Odefsey has a boxed warning in its product label regarding the risks of lactic acidosis/severe hepatomegaly with steatosis, and post treatment acute exacerbation of hepatitis B. Study 1216 is a phase 3b, randomized, double-blind, multicenter study among 630 virologically suppressed adults (HIV-1 RNA levels

<50 copies/ml) on a stable regimen of Complera for = six consecutive months. Patients were randomized 1:1 to either maintain their Complera regimen or switch to Odefsey. Study 1160 is a phase 3b, randomized, double-blind, multicenter study among 875 virologically suppressed adults (HIV-1 RNA levels <50 copies/ml) on a stable regimen of Atripla for = six consecutive months. Patients were randomized 1:1 to either maintain their Atripla regimen or switch to Odefsey. The studies will follow patients for 96 weeks after randomization. The studies are ongoing. The primary objective of each study is to evaluate the efficacy of switching from Complera or Atripla to Odefsey in HIV-1 positive subjects who are virologically suppressed as determined by the proportion of subjects with HIV-1 RNA <50 copies/ml at week 48 as defined by the FDA snapshot algorithm. The secondary objectives are to evaluate the bone safety of the regimens by the percent change from baseline in hip and spine BMD at week 48 and week 96, to evaluate the safety and tolerability of the treatment arms through week 48 and to evaluate the efficacy, safety and tolerability of the treatment arms through week 96.

UV light oven has potential to decontaminate produce at home aiqiang Chen of the University of Delaware is developing a device using ultraviolet (UV) light to kill foodborne pathogens on fresh produce in consumers’ homes and elsewhere.His UV light oven, which is about the size of a microwave oven, will combine UV light with stirred-up water to reduce Salmonella and other bacteria and viruses that can contaminate fresh produce.“At home, when the fresh produce reaches you, it might not be completely free of foodborne pathogens,” Chen said in a news release. “Typically, consumers don’t wash fresh produce if it has been prewashed, and those who do generally just wash it a couple of times with tap water. The oven will have a simple control panel to allow users to adjust treatment time and will offer a fixed UV intensity. “The decontamination comes through two sources, UV and water. The UV will kill pathogenic bacteria and viruses but the bad thing about UV is that it doesn’t penetrate through solids, although it can penetrate through clear water,” Chen said. “The water will wash off the pathogens from a food surface

and whenever they get into the water, they will be killed almost immediately.” Chen compared the device’s efficacy to that of washing produce with plain tap water. He applied both under two simulated Salmonella pathogen contamination scenarios: spot-inoculation, in which he contaminated a piece of produce in a particular spot, and dipinoculation, involving contaminating the entire piece of produce. After using samples of lettuce, spinach, tomato, blueberry and strawberry in the two scenarios, Chen found that the UV light oven decontaminated the fresh produce much more effectively than washing it with plain tap water. The oven could kill 99.7 percent of the Salmonella on the dip-inoculated lettuce, while the tap water washing could only kill 59.3 percent, he noted.“For spot inoculation, the UV is a lot better. It showed a lot of reduction. It can kill 99.999 percent of Salmonella spot-inoculated on tomatoes – it’s basically gone,” Chen added.The professor said that the UV oven method will not heat fresh produce and will not have a negative effect on its sensory properties.“The produce will be cold.

Colorado town discovers local water supply may have gone to pot Hugo, CO, a town of about 730 people, located 110 miles from Denver on the Eastern Plains has reported its municipal water has tested posiive for THC, marijuana’s main psychoactive ingredient. Peptide Discovery Platform System technology eptiDreamInc, a public TokyoP based biopharmaceutical company, has entered into a Technology License Agreement with US-based Genentech, a member of the Roche Group, to nonexclusively license PeptiDream’s proprietary Peptide Discovery Platform System (PDPS) technology. Celt ACD, which previously received CE mark and is sold in Europe, is indicated for arterial puncture closure in both diagnostic and anticoagulated percutaneous interventional cardiology and radiology patients. The agreement gives Aelan world-wide rights to intellectual property describing a novel epigenetic biomarker with a granted United States patent. PHARMA DPCO ORDER S.O.2209(E) - Whereas in line with the directions given in the review order issued vide letter no. 31015/9/2013/PI.I dated 05th May 2016 passed by the Department of Pharmaceuticals in application of review from various pharmaceutical companies against S.O. No. 3127(E) dated 10th December, 2014, ceiling prices of Glucose 5% Injection, Sodium Chloride 0.9% Injection and Glucose 5% + Sodium Chloride 0.9% Injection were fixed by National Pharmaceutical Pricing Authority (i.e. NPPA) vide notification S.O 1993(E) dated 3rd Jun, 2016 and Corrigendum Order 2062(E) dated 09th Jun, 2016. Whereas the manufacturers have made representations about existing unit volume based pricing system because of which manufacturing of aforesaid formulations in all kind of packaging for some pack sizes has become non remunerative affecting the availability, which may become detrimental to the public interest. Whereas, the NPPA had invited manufacturers claiming entitlement for separate ceiling prices for demonstration as notified by S.O. 1993(E) dated 3rd Jun, 2016 and Corrigendum Order 2062(E) dated 09th Jun, 2016. Whereas based on the representation and feedback from various glass and non glass manufacturers and its own examination, NPPA modified the previous categorisation of different types of packaging as (a) glass (b) non glass and (c) non glass with special features. Whereas NPPA in its a Authority meeting held on 23rd Jun, 2016 decided to re-fix the ceiling price of aforesaid formulations under modified categories i.e. glass , non glass and separate ceiling prices for specific manufacturers of non glass with special features. In exercise of powers, conferred by sub paragraph (3) and (4) of paragraph 11, and paragraph 14 the Drugs (Prices Control) Order, 2013, read with S.O. 1394(E) dated the 30th May, 2013, S.O. 1192(E) dated 22nd March, 2016 issued by the Government of India in the Ministry of Chemicals and Fertilizers and in supersession of the order of the Government of India in the Ministry of Chemicals and Fertilizers (National Pharmaceutical Pricing Authority) No. S.O. 3127(E) dated 10th December, 2014, S.O. 619(E) dated 26th February, 2015 , S.O. 644(E) dated 2nd March, 2016 and S.O 1993(E) dated 3rd Jun, 2016 read with Corrigendum order S.O 2062 dated 9th Jun,,2016 , in so far as it relates to formulation packs of Glass and Non-Glass packaging mentioned in the table herein below, except in respect of things done or omitted to be done before such supersession, the National Pharmaceutical Pricing Authority, hereby fixes the price as specified in column (5) of the table herein below as ceiling price exclusive of local tax applicable, if any in respect of the scheduled formulations specified in the corresponding entry in column (2) of the said Table with the strength and unit/packaging specified respectively in the corresponding entries in columns (3) and (4), thereof:
