Universal Cancer Biomarker Discovered That Detects Cancer In 10 Mins A single test to diagnose if one is suffering from Cancer. This has been made possible by researchers from the University of Queensland’s Australian Institute for Bioengineering and Nanotechnology (AIBN). THE RESEARCHERS HAVE IDENTIFIED A UNIQUE DNA EPIGENETIC PATTERN THAT APPEARS TO BE COMMON IN ALL TYPES OF CANCER. By Preety Suman

The test works on circulating free DNA fragments that can be easily obtained in affected persons body fluids. Via this simple diagnostic test results can be obtained in just 10 minutes and is visible to naked eyes in the form of a color change or an electrochemical signal. This groundbreaking discovery was led by Matt Trau, Ph.D., professor of chemistry at the University of Queensland and deputy director and co-founder of AIBN. A comparison was made between the epigenetic patterns on the genomes of cancer cells and healthy cells, specifically looking for the presence of methyl groups in the DNA. These methyl groups play an important role in the cell function as they act as signals that control which of the several genes should be turned on & off at a specific time, based on the requirement.

These unique DNA signatures are termed “methylscape” by the researchers and it appeared in every type of breast cancer, prostate cancer, colorectal cancer, and lymphoma that were examined. Every cancerous cell examined by the scientists has this highly predictable methylscape marker pattern.

This revolutionary one-step, highly sensitive, selective electrochemical or colorimetric cancer diagnosis test was designed by exploiting the difference in the methylscape present in the healthy and tumor cells.

As per reports till now this test has been performed on 200 samples across different types of tumor & healthy cells from humans, out of which the accuracy of cancer detection is as high as 90%. The researchers are currently in talks with University of Queensland’s commercialization company, UniQuest, to develop and license the technology. This revolutionary discovery can be a game-changer in the field of cancer diagnostics. The team acknowledged that the test still needs improvisations and its accuracy will be 100% incoming time.

Cure For HIV Moves A Step Closer In A Breakthrough Discovery In a breakthrough discovery, Researchers from the University of Illinois, Chicago have successfully figured out a way to manipulate TAT Gene circuit, a genetic switch that helps the hidden HIV virus under the cells to replicate. BY MANIPULATING THIS GENETIC SWITCH LATENT HIV VIRUS CAN BE COMPLETELY ERADICATED FROM THE BODY. By Preety Suman

While a human body gets infected with an HIV virus, the DNA of the virus penetrates deep into the nucleus of the host genome. As per reports published in National Academy of Sciences Journal, the HIV DNA has the Tat gene circuit as a key component, which controls the transcription and activation of the virus in the host body. Upon activation, the complete cellular mechanism of the host is taken over by the HIV virus to produce its replicas, simultaneously infecting the neighboring cell. As per a research, only when the Tat gene circuit is on, and the cells are being used to produce more of the virus, the HIV-specific immune effector cells can kill the virus-infected cells. Whereas the Tat gene circuit is off in cells that are latently infected, thus the cell goes about its normal business all the while harboring quiescent HIV.

Professor Jie Liang at the University of Illinois said that if we target the Tat gene circuit with a drug molecule to activate it, the latently-infected cells then can be modified to produce more of virus which can be further destroyed by the host’s immune system. But as of now, there are no drug molecule studies to attack the circuit. The HIV virus has the tendency to hide inside the host cells in an inactivated state, thus making it extremely difficult to flush out the latently-infected cells out of their

latency. Currently, Lifetime antiretroviral therapy is the only option for HIV infected patients. As per Liang, the reactivation of the Tat gene circuit in HIV infected cells is a very rare phenomenon. It has a random probability of being active or inactive. Advanced computational algorithms are being developed by researchers to study the functionality of the Tat gene circuit under different conditions. Researchers identified ways to manipulate the Tat gene circuit to make the “shock and kill” technique

more effective. They also studied the “block and lock” strategy, where latent viral particles are locked into latency by permanently blocking activation of the Tat gene circuit. The scientists concluded that by controlling HIV latency via Tat gene circuit manipulation, an effective therapeutic strategy can be chalked out which could cure HIV completely.

FOR THE FIRST TIME, RESEARCHERS FROM LUND UNIVERSITY IN SWEDEN HAVE DEVELOPED A MECHANISM TO CONVERT THE HUMAN SKIN CELLS INTO IMMUNE SYSTEM SENTINELS CALLED DENDRITIC CELLS, WHICH CAN COMMAND THE IMMUNE SYSTEM – T CELLS TO FIGHT TUMOR CELLS. By Preety Suman

This will lead to the development of better and much more effective cancer immunotherapies. This latest study was published in a journal called Science Immunology. Via this new mechanism called as direct reprogramming, the researchers could convert the cells of the skin – fibroblasts into immune – dendritic cells. Dendritic cells play a very important role in cancer immunotherapy by commanding the T-Cells target the cancerous cells. Since the dendritic cells are hard to produce in a large quality and are

rare, this research can prove to be a useful alternative source. In words of Filipe Pereira, research team leader, the entire process of reprogramming millions of cells derived from human tissue section into dendritic cells took only 9 days. He further added that these modified fibroblasts can be used to develop cancer gene therapy. Different transcription factor genes

were screened using transgenic mice as a medium to that could trigger the transformation process. As a result, they could find a combination of three transcription factors sufficient to complete the reprogramming of fibroblasts into dendritic cells. The transformed fibroblasts cells could engulf antigens and also interacted with T cells, similar to that of the dendritic cells.

A company called Asgard Therapeutics has been formed by Filipe Pereira in order to explore the clinical potential of this new technology. This research will open up newer avenues in the area of cellular immunotherapy for cancer treatments in the future.
