IMPRINT 2 Impacting Research Innovation and Technology – Call For Proposal MHRD and DST official notification for 2019 IMPacting Research INnovation and Technology (IMPRINT-2).

INTERESTED AND ELIGIBLE CAN APPLY ONLINE FOR THE CALL VIA SERB. IMPRINT-2 OFFICIAL CALL FOR APPLICATIONS FROM FACULTY MEMBERS AND RESEARCHERS IN ALL GOVERNMENT OF INDIA FUNDED HIGHER EDUCATION INSTITUTIONS (HEIS), CHECK DETAILS ON THE SAME BELOW: By Diluxi Arya

Call for proposal under IMPRINT will open on 15th Dec 2018 What is IMPRINT? India, a mighty global economy which will soon be crowned the youngest nation in the world faces multiple daunting challenges in terms of energy/physical/cyber security, potable water scarcity, environment and climate change, poverty, affordable health care for billions, etc. A vast majority of these tasks demand engineering intervention and technological innovation. A clarion call was made by the Honourable President and Prime Minister to address all engineering and technological challenges faced by the nation through a nation-wide unique initiative called IMPacting Research INnovation and Technology (IMPRINT) launched on November 5, 2015 from the Rashtrapati Bhavan. IMPRINT India was a multi-stakeholder mission programme aimed at bridging the gap between scientific knowledgebase created through fundamental and applied research and fruitful translation of the same through engineering invention and technogical innovation for the betterment of society. What is IMPRINT-2?

IMPRINT-India initiative was both novel and unique. Despite unprecedented interest, wide spread enthusiasm and unique scope and utility of the initiative, a need was felt to sharpen the focus, streamline the process of review and allotment, and showcase the fruits of innovation pursued and achieved through IMPRINT in public domain by creating a ‘knowledge portal’. Thus, IMPRINT has now graduated into the next edition, IMPRINT-2, with wider scope, simpler guideline for participation and better synergy among investigators, partners and collaborators. It is also realised and appreciated that IMPRINT-2 should be undertaken in a more inclusive manner by expanding the catchment of implementing institutions, by adopting a more demand-driven strategy of solution development and by incorporating the specific externalities of the states of India so as to make end-user translation and technology adoption easier. SERB in the DST is proposed to be made the nodal agency for implementing the IMPRINT-2.

ELIGIBILITY Who can apply Principal Investigator (PI): The applicant or the Principal Investigator (PI) should be an Indian citizen. The PI must hold a regular academic/ research position (Faculty member or Scientist or Scientific Officer) of Centrally Funded Technical Institution (CFTI), MHRD funded Central University (CU) or MHRD funded Higher Education Institution (HEI) offering/pursuing research and development (R&D) in engineering and technology (E&T), having at least 4 years of regular service left before superannuation as on date of submission of the proposal. For a given project, there will be only one PI. Scientists who has project under IMPRINT-2A and IMPRINT- 2B are not eligible for IMPRINT-2C. The PIs should be confident of developing and demonstrating new and novel technology solutions borrowing from already established and doc-

umented knowledge base. The following points will be given priority during the project evaluation: • Projects which address any of the ‘themes’ identified by one of the 10 Technology Domains and ‘topics’ proposed by stake holder Ministry or Departments. • Projects which qualify as translational research initiatives leading to viable technology, prototype or working model development. • Projects with industry participation or startup company involvement that may lead to developing commercially viable products and thereby creating jobs. CRISPR Code Cracked For Precise Human Genome Editing CRISPR Cas9 has been a trending technique effective in editing genes in various organisms. Tons of experiments have been conducted using CRISPR, latest being the CRISPR baby born in China claimed by a Chinese scientist. HOWEVER, THERE HAS BEEN NO LEAD IN PREDICTING THE EDITING OUTCOME IN SPECIFIC SITES. By Preety Suman

In a major breakthrough, scientists at the Francis Crick Institute have unveiled a simple set of rules determining the precision of CRISPR/Cas9 genome editing in human cells. This new discovery will pave ways for genome editing both in labs as well as clinics with an increased efficiency and safety. The above findings were published in Molecular Cell. CRISPR has been a topic of discussion Worldwide owing to its ethical usage. Pertaining to the unpredictable outcome of genome editing which may result in random insertions & deletions of DNA fragments. The effects of CRISPR editing was studied in 450 genes at 1491 target sites in human cells. The researchers concluded that the outcome of the CRISPR based genome editing depends on a simple rule, a single genetic ‘letter’ occupying a particular position in the region recognized by the ‘guide RNA’ to direct Cas9 for precision editing. Synthetic molecules were designed called Guide RNAs, that comprises of 20 A,T,G,C’s designed specifically to bind to a specific fragment of the target gene DNA. The genetic letters in Guide RNA bind complementarily and stick firmly with A binding to T and C binding to G. Cas-9 enzyme guided by RNA molecule further scans the genome until the region of interest is located. Upon locating the correct DNA sequence the RNA molecule sticks to it, while cas9 cuts the DNA fragment into three letters from the end of the target sequence. Pieces of genetic code are either inserted or deleted randomly in an attempt by the cells to repair the break. As per the above experiment, researchers concluded that the result of CRISPR genome editing depends on the fourth letter from the end of

the RNA guide that is adjacent to the cutting site. They further noted that if this fourth letter is either A or T, precise genetic insertion can be expected. Whereas if it’s a C, it will lead to a relatively precise deletion and a G will lead to many more imprecise deletions. Thus by avoiding editing sites containing G as the fourth letter after cuts made will lead to predictable genome editing results. The researchers also discovered the outcome of gene editing is also affected by how the target DNA open or close after the cut is made by cas9. Upon addition of compounds that force DNA to open and by allowing Cas9 to scan the genome, more efficient editing takes place, which could help when modifications are needed to be introduced in particularly closed genes. Crick group leader, Paola Scaffidi said, “The effects of CRISPR were thought to be unpredictable and seemingly random, but by analyzing hundreds of edits we were shocked to find that there are actually simple, predictable patterns behind it all. This will fundamentally change the way we use CRISPR, allowing us to study gene function with greater precision and significantly accelerating our science.” Read Further: Chakrabarti et al., Target-Specific Precision of CRISPR-Mediated Genome Editing, Molecular Cell (2018), https://doi. org/10.1016/j.molcel.2018.11.031

First Xenotransplantation Trial Of Genetically Engineered Skin Cells XenoTherapeutics gets a green signal to FDA to conduct the world’s first human clinical trial that involves live skin cells derived from genetically engineered pigs for treatment of intense burns. AS SAID PAUL HOLZER, CEO AND CO-FOUNDER OF XENOTHERAPEUTICS, XENO-SKIN IS GOING TO BE THE FIRST NON-HUMAN ORGAN TRANSPLANT CLEARED BY THE FDA FOR INVESTIGATIONAL USE. By Preety Suman

Xeno’s biologically active skin product comprises of animal-derived dermal and epidermal tissue layers and is sterilized to be free of the viral pathogens. As per XenoTherapeutics, In the past, this used to be an obstacle for usage in porcine xenotransplants. For patients with intense burns, this transplantation can serve as a temporary yet immediate treatment option. The process of enrolling patients at Massachusetts General Hospital is to be initiated by the end of the year after FDA cleared it for human trials. Holzer in a statement said the com-

pany’s main motto with this product was to advance the science of xenotransplantation for practical therapies, which would benefit patients all around the globe. No immediate treatment option was available until now to meet the needs for severe burns, this revolutionary

product would pave ways for further organ transplantation research, he added. Xeno-Skin will be mass-produced, cryopreserved & shipped across the globe. Patients with intense burns risk infections & fluid loss that can

lead to organ failure. With XenoSkin, deceased donor allograft which is available in very limited quantities can be either replaced or complemented.

Scientists Crack Complex Enzyme Structure With Google’s Help In a latest research study published in PNAS, a chemistry professor named Victor Batista & his fellow researchers at Yale have successfully identified key amino acids that regulate bacterial enzyme essential for almost all microorganisms with the use of Google’s PageRank algorithm. VIA THIS NOVEL APPROACH, THE RESEARCHERS HAVE CRACKED DOWN THE COMPLEX STRUCTURE OF ENZYMES & ITS REGULATION. By Preety Suman

Enzymes have the unique property to accelerate chemical reactions. The substrates bind to the active site of the enzymes, where the reaction takes place. However, the acceleration or inhibition of a reaction takes place when a molecule binds to another site of the enzyme called the allosteric site. After decades of research, still, no proper information exists on how exactly the active site & the allosteric site of an enzyme communicate with each other. High flexible structurality of the enzyme & presence of an infinite number of atoms has made the identification process difficult. Yale researchers noted that a similar question was addressed years before in the world of computer science.

Researchers at Google had studied the flow of information online, using PageRank, an algorithm to signify the importance of each web page concerning the quantity and quality of links to other Web websites.

glycerol phosphate synthase (IGPS), a bacterial enzyme found in most microorganisms.

This revolutionary research will set forth further experimentation & validation in this field keeping the current research outcome as a base. Advanced Using the same PageRank, the team experimentation on IGPS activity identified amino acids important for may lead to the development of new the allosteric process in imidazole antibiotics, pesticides, and herbicides.

This research was supported partially by National Institutes of Health and the National Science Foundation. The study’s other co-first author is Christian Negre of Los Alamos National Laboratory. Co-authors are Batista and J. Patrick Loria of Yale; former Yale researchers Heidi Hendrickson, Rhitankar Pal, and George Lisi; Ivan Rivalta of the University of Lyon; and Junming Ho of the University of New South Wales.
