Purpose of the Prestop®Vision kit The Prestop®Vision kit is an easy to use tool for revealing the presence of Gliocladium catenulatum fungal strain J1446 after aerial application of Prestop® onto plants.

Contents of the kit •

20 petri dishes (containing Sabouraud's growing medium in a gel form)

1 user guide

5 ampules of sterilising fluid

1 petri dish backing card for observation assistance

1 USB key with multimedia content

1 pen

Use of the kit 1

Apply Prestop® onto your plants.

Extract a piece of stem or leaf ten minutes after application. The sterilising fluid provided enables you to disinfect your cutting implement (knife, scissors, etc.) and your hands to prevent any non-target micro-organism contamination.

Open a petri dish. Put the sprayed side of the stem or leaf against the growing medium gel. Gently press the plant material so that it is in direct contact with the gel medium.

Close the petri dish and store for several days at room temperature.

Regularly return to look at the petri dish and monitor the Gliocladium catenulatum J1446 growth. Write down the results seen on an observations table.

At a macroscopic level, Gliocladium catenulatum strain J1446 normally creates a pinkish-white growth. This is the mycelium (formed by the massing of the fungal hyphae) of the growing micro-organisms.

Use of the PrestopÂŽVision kit Sabouraud's growing medium is a jelly which prevents the growth of bacteria and encourages the growth of fungi and yeasts. As this medium favours the growth of Gliocladium catenulatum fungal strain J1446 it will also favour the development of other kinds of mycetes that are naturally present on plant material. It is therefore normal to see the emergence on the growing medium of other micro-organisms such as Penicillium spp. or Trichoderma spp. which will have a green appearance. Contamination by non-target micro-organisms can be restricted by hygiene and disinfection measures being implemented, such as washing cutting tools and hands with the sterilising fluid provided, when preparing the samples for placement into the petri dish.
