Presidents Report

As another month has flown by, so many projects have been wrapped up. Harvest completed, hives treated and fed. Now we are busy wrapping and putting things away for the season. Mites always seem to be the biggest concern before winter. I’ve been hearing some alarming stories of very high mite loads and some hives showing damage before winter. It seems we have to work harder to control mites then ever before. Many producers have been doing mid summer treatments as well as multiple rounds of oxalic and formic in the fall. Hopefully we get some better options soon that will help as it seems Apivar resistance seems to be quite widespread as well. Hopefully the persistence of the beekeepers applying the acids will pay off and we will have good wintering success.

Members of the ABC board had an opportunity to meet with Minister Sigurdson and some of his team. We had an opportunity to cover a wide range of topics and issues including honey bee health and the challenges of bringing new treatment tools to market, stock replacement and a north American beekeeping industry strategy, working with Alberta Sugar Beet Growers on a more reliable/consistent supply of beet syrup, challenges in trade with Japan and long-standing issues such as commercial insurance for farm vehicles and Alberta health care cards for seasonal workers. Overall, Minister Sigurdson was receptive, interested and expressed interest in learning more about beekeeping, the needs of the industry, and how government can support a viable industry into the future. He did indicate there is much he still has to learn about beekeeping in Alberta. We are looking forward to Minister Sigurdson presenting opening remarks and welcome at the 2023 convention.

ADM Rosser was the chair for the Bee Sustainability Working Group. He was very interested in learning more about beekeeping in Alberta and discussing some of the issues and recommendations raised during the Bees SustainabilityWorkingGroupmeetings.

When he’s not touring bee farms ADM Rosser’s responsibilitiesincludetradenegotiations, agriculture and agri-food sectoral competitiveness, anddepartmentalregionaloffices.

September 1 is the start of a new fiscal year for the Commission. Office staff are busy working with executive on wrapping up the 2022/23 fiscal yearandplanningforthe2023/24year.

Challenges and trade-offs to Queen Immunity

Queen immunity begins with social immunity via worker behaviour. Workers perform behavioural tasks to defend against pathogens and parasites (e.g., grooming, removing diseased or parasitized larvae, using propolis in nest building, and removing dead adults from inside the colony)1,2. Social immunity plays a critical role in preventing disease transmission within the colony, including preventing transmission to the queen. For example, queens forced to interact with only viral-infected workers have similar viral loads and show similar symptoms to their infected worker counterparts. However, when queens have access to both healthy and infected workers, the queens have lower viral loads3. It is not clear if infected workers avoid the queen when healthy workers are present, or if the queen avoids interacting with infected workers

when healthy workers are present, but behavioural strategies that limit queen interactions with diseased individuals play an important role in social immunity for the queen and may explain why queens are considered less susceptible to pathogens compared to workers4

Like workers, queens also possess a system of individual innate immunity, and this is where things get complicated. (see figure 1) Innate immunity begins with the exoskeleton and alimentary tract designed to limit invader passage through a combination of physical and chemical barriers5. For example, much like the hairs in our noses and on our eyelids and eyebrows, hairs on a bee help keep pathogens from penetrating their exoskeleton. In fact, shaving the hairs on a queen’s thorax is a common method used

in experimental research when inoculating queens with chronic bee paralysis virus3 Interestingly, even queens that have their thorax shaved but have no virus topically applied have higher viral loads than queens that do not have their thorax shaved3. Similarly, like intestinal mucus in mammals, the epithelium in the midgut of honey bees (and other insects) produces a gelatinous thick covering called the peritrophic membrane that protects the epithelium from food abrasions and direct contact with pathogens.

Should invaders breach the exoskeleton line of defence, the queen’s cellular and humoral (i.e., body fluid) immune pathways are ready to defend her. For small invaders like bacteria and unicellular fungi, the queen’s immune system can use a mechanism called phagocytosis to remove the invader - a cell ingests the invader and packages it for removal from the queen’s body. For larger invaders, the queen’s immune system uses haemocytes (insect “blood” cells) to bind to invaders. These haemocytes encapsulate the foreign bodies, thereby restricting any further growth and movement, often killing the invader6. These two mechanisms (phagocytosis and haemocytes) can be used to defend the bee against both familiar and novel pathogens. A third line of cellular and humoral defence includes induced responses, used when the bee immune system recognizes its invader. This mechanism involves the production of short chains of specific amino acids (the building blocks of proteins) called antimicrobial peptides (AMPs)7. AMPs are small proteins that are synthesized

in response to immune priming in the fat body and hemocytes of honey bees and secreted into the hemolymph8. AMPs protect against microorganisms but take time and energy to synthesize8

While generally more protected than workers, queens are susceptible to numerous viruses including black queen cell virus, deformed wing virus, chronic bee paralysis virus, and sacbrood virus. Highly infected queen larvae will often be removed from the colony by workers9, and therefore the prevalence of symptomatic queens and the impacts of disease and viruses on queen health and performance are not well studied. However, wrinkled wings from deformed wing virus (DWV) have been seen in queens10. Further, DWV has been associated with ovary degeneration in queens11, although high DWV titres in queens are not always associated with ovary degeneration11. Despite appearing asymptomatic, queens can be important vectors of bee viruses through vertical transmission to the eggs they lay12,13. Thus, global queen trade and pollination services is potentially an underestimated source of disease transmission in apiculture.

A queen’s ability to invest physiological resources into the cellular, humoral, or AMPs pathways of innate immunity depends on her environment, resources, and exposure to stress. Queens are already challenged by common hive pathogens but can be further challenged in the presence of additional stressors. For example, pesticide residues found in the nectar and pollen of contaminated plants are taken back to the hive by foragers and subsequently fed to larvae, workers, drones, and the queen, and these pesticide residues may work in concert to impact queen health. Researchers at the USDA in Arizona showed queens grafted from colonies exposed to field realistic levels of insecticides (e.g., chlorpyrifos) were significantly less likely to emerge

than queens grafted from colonies not exposed to insecticides but found even lower queen emergence success when queens were grafted from colonies exposed to insecticides and fungicides14. Further, newly emerged queens grafted from colonies exposed to insecticides and fungicides had higher titers of DWV than newly emerged queens grafted from control colonies14, suggesting exposure to pesticides may challenge queens’ immunity to common hive pathogens. Indeed, queens exposed to sublethal doses of neonicotinoids via direct feeding or trophallaxis show signs of suppressed innate immunity15

Interestingly, researchers at the University of British Columbia16 recently demonstrated a trade-off between the viability of stored sperm and immune function in queen honey bees, where resources are invested in the long-term storage of sperm at a cost to immune function. Queens mate early in life, taking multiple mating flights and storing sperm from multiple drones in a sperm storage organ called the spermatheca. Because queens do not re-mate later in life, a queen’s reproductive success is limited by the number and viability of her stored sperm. Thus, it makes sense that a queen

might divert energy and resources from her innate immunity to the storage of her sperm. The number of stored sperm has been linked to immunity in other invertebrates as well. For example, in the leaf cutting ant, Atta colombica, queens downregulate their immune systems when establishing a new colony and trade-off short-term survival against long-term reproductive success17. Lower stored sperm viability has also been found in field crickets18 and in queen ants16 in response to an immune challenge.

With immune function linked to reproduction in queen honey bees16, investment in sperm storage may make queens more susceptible to diseases, particularly when exposed to multiple stressors, potentially impacting queen quality and failure. (see figure 2). The added protection from social immunity behaviours may allow queens to invest fewer resources into their innate immunity and more resources into sperm storage. Therefore, we need to encourage these social immunity behaviours in queen breeding programs. For example, selecting for hygienic workers has been demonstrated to increase colony survival in the face of disease and pest challenges20

Saving Bees with Synthetic Biology: ImmuniBee’s Solution to Deformed Wing Virus

The iGEM club at the University of British Columbia, Okanagan, is a gathering for inquisitive minds, serving as a platform for students to explore, experiment, and expand their scientific knowledge. With its engaging activities, the club aims to foster a love for synthetic biology and nurture future scientists.

Every year, the iGEM club forms a competitive team that competes at an international jamboree in Paris. Hundreds of teams from around the world gather to present their projects and contributions to the field of synthetic biology. Key industry partners, startup sponsors, and government experts attend to support these future advancements. The field of synthetic biology creates or modifies new genes in order to modify living systems for useful purposes. Synthetic biology is not just for movies, it can be applied to so many global problems!

The club is entering its third year, and the second competitive team is wrapping up their project. This year, it’s all about saving the bees! Members have learned about the vital role these pollinators play in our ecosystem and economy, and the horrific losses experienced by honeybee populations. At Okanagan iGEM, team members are working to create the first biosynthetic vaccine that can give bees and their offspring immunity to Deformed Wing Virus. This project is called ImmuniBee!

Back in February, some team members visited Planet Bee, a honey farm in Vernon that aims to educate and inspire the public on honeybees. There, they learned that a significant contributor to honeybee decline is the Deformed Wing Virus (DWV) and its transmission through the Varroa destructor mite. When the time came to brainstorm this year’s project, team members were inspired to make a difference in the beekeeping community by tackling the most prevalent virus threatening BC honeybees. A love for bees was born out of the project, and the team has been working tirelessly to make this idea a reality.

Bee immunity is complex, and common methods of initiating an immune response aren’t possible when tackling a virus. So, what’s their approach? Enter trans-generational immune priming (TGIP)! First, surface proteins from the Deformed Wing Virus capsid are expressed on a bacterial surface. The deactivated

bacteria are introduced to worker bees, who feed it to the queen, and her immune system responds via TGIP. The viral proteins are also carried to her ovaries, effectively immunizing not only herself but also her offspring against DWV.

A large part of iGEM focuses on looking at how your project impacts the community, and how the community impacts your project. The Human Practices team with ImmuniBee have been collaborating with beekeepers and experts from around the Okanagan and Alberta to explore the feasibility of the project and gather opinions from key stakeholders. The team was made aware of the communication gap between researchers and beekeepers, leading to projects that work in theory, but not practice. To avoid this, they’ve actively engaged with numerous beekeepers and sought input on the preferred method for administering ImmuniBee to bees and discussed various concerns related to testing and outcomes. They also met with Samantha Muirhead, the Alberta provincial apiculturist, to learn about government varroa testing and bee-related legislation. These interactions have added to their understanding of bee losses and increasing awareness on the issue.

The team has worked on multiple projects to engage with the community and inspire others to learn about bees. Next month, the group will be speaking about their project at the BC Honey Producers Association and aims to use this opportunity to learn from industry professionals and engage with the community. They are also in the process of applying to become a Bee Campus under Bee City Canada. As part of this effort, team members are working with UBC staff to develop a pollinator trail on campus that will feature native flowers and emphasize the importance of native bees in our ecosystems.

So, what’s next? The team is working hard to bring their project to fruition. If they succeed, this could have world-wide implications, and the technology could be applied to other viruses affecting bees. The team will be competing at the Grand Jamboree from November 2nd-5th along with over 7000 other competitors

Mimi's Honey Vanilla Banana Bread

Ingredients

• 3-4 large ripe bananas, mashed

• olive oil or melted butter cooled

• brown sugar

• sugar

• honey

• eggs

• pure vanilla extract or Madagascar Vanilla

• all-purpose flour

• baking powder

• baking soda

• Pinch of salt

Instructions

1. Preheat the oven to 350°F. Spray with non-stick spray a 9x5 loaf pan.

2. Mix together the melted butter or olive oil, bananas, honey, sugars, eggs and vanilla together.

3. In another bowl, add the baking powder, baking soda, salt and flour together and whisk and then add to the wet mixture and mix until just incorporated.

4. Pour mixture into a loaf pan and bake for 1 hour.

5. Let cool in pans for 10 minutes, then remove from pans and cool on a wire rack until completely cool.
