Making Sense of Antisense and Nonsense

ne of four major initiatives

announced last fall as a result of the unprecedented influx of donations resulting from the 2014 ALS Ice Bucket Challenge is Neuro Collaborative, which employs a three-pronged approach drawing on the unparalleled expertise of three labs. One of these labs, located at CedarsSinai Medical Center in Los Angeles under the direction of Clive Svendsen, Ph.D., is developing a Stem Cell and Motor Neuron Core Facility with the purpose of creating clinical-grade induced pluripotent stem cell (iPSC) lines. iPSCs have emerged as a key research tool and potential source of therapeutic cells in ALS. As the cell lines become available, they will be openly shared with the ALS research community. “The idea is,” said Dr. Svendsen, “to essentially create ALS in a petri dish.” Cedar-Sinai is at the forefront of this kind of research, and the lab is working with the C9orf72 gene, which is the most common genetic cause of ALS, responsible for up to 40 percent of inherited cases and about six percent of sporadic cases. “Antisense oligonucleotide (ASO) therapy” and “nonsense-mediated decay” (NMD) don’t sound all that sensical, but each has recently emerged as a potentially important idea in developing new treatments for ALS. ASO therapy is believed to offer the best hope for the two major genetically based forms of ALS, due to mutations in either the SOD1 gene or the C9orf72 gene. NMD is a newly discovered cell pathway that might be exploited to help preserve motor neurons. In this article, we will try to make some sense of these concepts and understand what they might mean for therapy.

Nucleotides and Antisense Both ASO therapy and NMD are linked to DNA and RNA. You may remember from high school biology that DNA is the hereditary material, composed of building blocks called nucleotides. There are four types, abbreviated A, T, C and G. DNA is composed of two strands, each a long string of nucleotides. The chemical structures of the nucleotides allow them to pair up, so that A on one strand is always matched with T on the other strand; similarly, C always pairs up with G. For instance, if the nucleotides on one side read ATTCAG, the other side will read TAAGTC. The two Continued on page 4
