细胞凋亡概述 细胞凋亡(Apoptosis)或程序性细胞死亡(Programmed Cell Death)是指细胞为了清除自身不需要的和失去功能细 胞的一种高级调控过程。凋亡可以发生在细胞生长、 衰老、免疫反应或者当细胞受到疾病和毒害因素的破 坏，例如放射性药物和化学化疗等的过程中。

始类Caspase(2.8.9.10)、效应类Caspase(3.6.7)和激活类 Caspase(1.4.5)。凋亡的起始通过起始类Caspase的激活进 而起始Caspase扩大凋亡的级联放大信号。Caspase3是其 中关键效应执行Caspase分子之一，它可以切割其下游 底物，如PARP酶、细胞角蛋白、α-胞影蛋白等，从而造 成最终的凋亡效应，如细胞膜外表面暴露磷脂酰丝氨 酸(PS)，DNA断裂和结构蛋白降解。

CaspGLOWTM 活性Caspase染色试剂盒 CaspGlow™原位Caspase染色试剂盒，可以在活细胞内快速灵敏的原位检测到激活的Caspase。在该试剂盒中，FITC或 者罗丹明荧光团耦联到Caspase抑制剂上作为原位标记，这种无细胞毒性并且具细胞通透的Caspase抑制剂不可 逆的与凋亡细胞中激活的Caspase结合，由于存在FITC或罗丹明等荧光标签，这样可以通过荧光显微镜、荧光酶标 仪或者流式细胞仪直接检测凋亡体中活化的Caspase。此外试剂盒中还提供了无荧光标记的Caspase抑制剂作为 阴性对照。 B

凋亡细胞分离 BioVision凋亡细胞分离试剂盒提供了一种简易有效的方法，通过使用Annexin V/磁珠(MagBeads)，在细胞培养液或者组织 液中分离凋亡细胞或去除死细胞。Annexin V是一种钙离子依赖的磷脂结合蛋白，对磷酯酰丝氨酸（PS）具有高亲和 力。PS在凋亡细胞和死细胞中会重新从质膜内部向外部翻转。因此Annexin V/磁珠(MagBeads)中的Annexin V可以通过结合 PS而富集凋亡细胞，结合了磁珠的凋亡细胞由磁力富集，而非凋亡细胞则仍然处于自由状态。分离出来的凋亡细胞 和健康细胞可以用于各种凋亡机制和通路的研究。本试剂盒也可以用于在细胞培养时去除死亡细胞。

凋亡还可以不通过传统的Caspase介导切割级联反应来调控。有更多的证据证明改变蛋白水解酶，例如溶酶体组 织蛋白酶，可以起始和传递凋亡信号，但是目前还不能确定组织蛋白酶如何介导凋亡。溶酶体组织蛋白酶，常被 作为毒理学研究的敏感标记酶。Biovison最新提供的组织蛋白活性检测试剂盒都是基于荧光的实验，利用每种 组织蛋白酶偏爱的底物序列，标上AFC，这样包含有组织蛋白酶活性的细胞裂解物或者其他样本将该底物序列 切割，而被酶切后生成的自由AFC则释放出黄绿光荧光(λmax = 505 nm)，这种荧光可以用荧光计或者荧光分析仪 读取数据。对比处理的样品和正常对照组的样品发射的荧光强度，就可以得出钙活性的改变。该检测试剂盒步 骤简单，只需要大约2小时即可完成检测；试剂盒内包含了阴性和阳性对照，我们也可以单独提供活性的钙蛋白 酶、钙蛋白酶抑制剂以及抗体。
