NormaJeaneMortensonwasgiventhe nameMarilynMonroeonthedayof herbirth,June1,1926,attheLos AngelesGeneralHospitalinLos Angeles,California.GladysPearl Baker(néeMonroe;1902–1984),her mother,wasborninPiedrasNegras, Coahuila,Mexicotoalow-income familyfromtheMidwestthathad movedtoCaliforniaattheturnofthe century.GladysPearlBakerdiedin 1984.GladysBakerwasjust15years oldwhenshetiedtheknotwithJohn NewtonBaker,amanwhowasnine yearshersenior.Robert,wholived from1918to1933,andBerniecewere theirtwochildren(1919–2014).In 1923,shehadherpetitionfordivorce andrequestforexclusivecustodyof thechildrengranted.However,shortly afterwards,Bakerabductedthe childrenandtransportedthemtohis homestateofKentucky

ThefactthatMonroehadasisterwas notrevealedtoheruntilshewas12 yearsold,andthetwodidnotinitially meetuntil1944,whenMonroewas either17or18yearsold.Followingthe endofhermarriage,Gladysfound employmentatConsolidatedFilm Industriesasafilmnegativecutter.She wedMartinEdwardMortensenin 1924,butthecouplesplitonlyafew shortmonthsaftertheirmarriageand werelegallyseparateduntil1928.The resultsofDNAtestingconductedin 2022suggestedthatMonroe's biologicalfatherwasCharlesStanley Gifford(1898-1965),acoworkerof Gladys'swithwhomshehadanaffair intheyear1925.Thisinformationwas revealedin2022.Doris(1920–1933), theolderofMonroe'stwootherhalfsiblings,andCharles,theyoungerof Monroe'stwootherhalf-brothers,were bothbornduringGifford'smarriageto hisfirstwife.

DespitethefactthatGladyswas emotionallyandfinanciallyunprepared forakid,Monroehadasecureand happylifeinhisearlyyears.Inthe

Shehadprominentpartsinthefilm noirNiagara,whichexplicitly dependedonhersexappeal,andthe comediesGentlemenPreferBlondes andHowtoMarryaMillionaire,which cementedherstarimageasa"dumb blonde."By1953,Monroewasoneof themostmarketableactressesin Hollywood.Thephotographsofher nakedbodywereonthecoverand

centerfoldoftheveryfirstissueof Playboy,whichwaspublishedthe sameyear Throughouthercareer,she hadalargepartintheformationand controlofherpublicimage,butshe wasdisheartenedwhenshewas typecastandunderpaidbythestudio. Thepublicwasveryinterestedinthe turbulentpersonallifeofMonroe.She battledaddictionaswellasmental issuesthroughoutherlife.Her marriagestoJoeDiMaggio,aformer baseballstar,andArthurMiller,a writer,bothreceivedagreatdealof mediaattentionbutultimatelyresulted indivorce.HerdeathonAugust4, 1962,attheageof36,wastheresultof anoverdoseofbarbituratesthat occurredatherresidenceinLos Angeles.Itwasdeterminedthatsuicide wasthemostlikelycauseofherdeath.

Eventhoughthereisalotof innovationintheindustry,biopharma companiesarefacingacomplexand difficultenvironmentduetomore competitionandlongerR&Dcycle times,lesstimeonthemarket,patents thatareabouttoexpire,fallingpeak sales,pressurearoundreimbursement, andmorescrutinyfromregulators.As we’veshowninourseriesofreportson “Measuringthereturnfrom pharmaceuticalinnovation,”these factorsarecausingtheprojectedreturn oninvestmentthatlargebiopharma companiescouldgetfromtheirlatestagepipelinestodropbyanalarming amount,whichthreatenstheirlongtermfuturesAItoolsarefinally changing.

HerearesomewaysthatAItoolsare finallychangingtheworldof biopharma:

Nomatterwhattheirjobtitleis,those inchargeofdigitaltransformationin biopharmacompaniesneedtousedata andnewdigitaltechnologiestothe fullesttochangetheirbusinessand operatingmodels.Themostanticipated andimportantofthesetechnologiesare thosethatfallundertheterm“AI.”

Atthemoment,machinelearning(ML) isthemostimportantpartofmanyAI technologies.Recentimprovementsin MLalgorithmsandthemore complicatedpartofMLcalleddeep learning(DL)arewhataregetting peopleexcited.Untilrecently,many companiesdidn’thavetheskillsto makethemostofAItoolsarefinally changing.Thisisbecausefindingthe rightusecases,makingcustom solutions,andusingthesesolutionsat thescalerequiredaccesstolargedata sets,specializedinfrastructureand processingpower,andateamofdata andanalyticsexperts.7Usingitona largescalealsorequiredalotofmoney tobespent.

Today,thisisquicklychangingastech giantsandstart-upsthatwereearlyto useAIofferAI-baseddevelopment toolsandappsasproductsand services,sothatothercompaniescan alsouseAI.Biopharmacompanies haveaccesstolargedatasets,andthe resourcestogetthecomputingpower andtoptechnicaltalenttheyneedand areinagoodpositiontoprofitfromAI iftheyinvestearlyAItoolsarefinally changing.

1.ThemarketforAIinbiopharma

MarketsandMarketspredictsthatthe marketforAItoolsarefinallychanging inthebiopharmaindustryandwill growfrom$208millionin2018to $3.88billionin2025,whichisa compoundannualgrowthrate(CAGR) of52.9%.

Between2018and2025,itisexpected thatdrugdiscovery,precision medicine,medicalimagingand diagnostics,andresearchwilldrive mostoftheAItoolsthatarefinally changinginbiopharma.Duringthe periodoftheforecast,drugdiscovery willbethebiggestmarket.Itwillgrow from$159.8millionin2015to $2,999.7millionin2025,whichisa CAGRof52.0%.Overthenextfive years,themarketforprecision medicineisexpectedtogrowby60.2% peryear,from$16.3millionin2018to $440,9million.CAGRsofover50% arealsoexpectedformedicalimaging anddiagnosticsandresearch(see figure4).

2.ThingsthataremakingAIgrow acrossthevaluechain.

ThegrowthofglobalAItoolsare finallychanginginbiopharmaandthe potentialfordigitaltransformationare causedbyseveralthings.Froma technologyanddatapointofview, transformationisbeingdrivenbythree thingsinparticular:moredata,more powerfulcomputers,andcheaper computers.

3.Biopharma’sfuturewillbedriven byAI

Biopharmacompanieswilluse technologiestospeedupinnovation, streamlineprocesses,andgetridof

barriersasaresultofdigital transformation.AItechnologieswillbe attheforefrontofthischange.They willhelpimproveefficiency,power newproductsorservices,enablenew businessmodels,andcreatea biopharmaindustrythatisvery differentfromwhatitisnowAItools arefinallychanging.

4.AIwillaffecteverypartofthe biopharmaceuticalvaluechain.

AIwillaffectthewholebiopharma valuechain.Atfirst,itwilldothisby collectingandcombininginformation fromdata(seefigure15).Hereare someexamplesofhowAIcouldbe usedinthedifferentpartsofthevalue chainAItoolsarefinallychanging.

5.Researchandfindout

UsingAIandcomputationalbiologyto findnewdrugsandmakethembetter couldmakeahugedifferenceinhow productivepeopleare.Inturn,thiswill makeresearchanddiscoveryless expensive,faster,andbetter.Itwillcut downonthenumberofdrugsthat don’tworkbyfindingpatternsinalot ofdata.Itwillalsoimprovetarget identification,safetyandeffectiveness assessments,andmodelingthat predictswhatwillhappenand“testing inthecomputer.”

OneimportantthingthatAItoolsare finallychangingispredictinghow moleculeswillinteractwitheachother Thishelpsusfigureouthowdiseases work.Inturn,thesemechanismscould helpfindnewbiomarkers,find,design, validate,andoptimizenewdrug candidates,andfindexistingdrugsthat couldbeusedforotherpurposes.

CombiningAIwithothertechnologies likeorgan-on-a-chip,3Dcellcultures, andothercellmodelsthatmimic humanbiologyin-vitrocouldreduce theneedtotestonanimalsandcould eventuallyleadtotheendofPhase1

clinicaltrialsinhealthyvolunteers.

Atthemoment,over148start-up companiesandover33large biopharmacompaniesareusingAIto researchandfindnewdrugs.Theydo thisthroughpartnershipsorontheir own.

40–41.Companiesarealsolooking intothepossibilityofusingAIto designandrunpreclinicalexperiments, whichcouldhelpcutdownthetimeit takestofind,develop,andreleasea newdrugAItoolsarefinallychanging.

6.Clinicaldevelopment

Clinicaltrialsarereadyforchange becausetheyaretakinglonger,cost more,andhavealotofmanualsteps. Also,alotoftrialsareneverfinished. ClinicalTrials.govwasusedtoregister morethan30,000clinicaltrialsin 2018.Sincetheyear2000,nearly 300,000trialshavebeenregistered. Butlessthan5,000trialsreportedtheir resultsin2018,andsince2000,less than35,000registeredstudieshave reportedtheirresultsAItoolsare finallychanging.

Biopharmacompaniesarecurrently lookingintohowtheycanuseAI technologytoimprovetheirclinical trials.Thisincludesimprovingtrial designandpatientrecruitment, analyzingbothtrialresultsand evidencefromtherealworld,and automatingthepublicationofresultsto makeitfasterandmoreefficient.Ifthe implementationgoeswell, developmentcostscouldgodown, peoplewouldbemorelikelytofollow therules,andtheresultswouldbe better&AItoolsarefinallychanging.

Also,havingaccesstolargeamounts ofdataandAI-poweredanalyticscould leadtothecreationofhighlyeffective personalizedtherapieswithouttheneed forlarge-scaleclinicaltrials.

7.Makingthingsandgetting supplies

Biopharmamanufacturingandsupply chainscreatealotofreal-timedatathat hasn’tbeenusedinausefulwayfor manyyears.ButAItoolsarefinally changingtechnologiesthatcould changetheseprocessesbyprocessing andmakingdecisionsaboutdatainreal time.Thiscouldmakesupplychains trulydata-driven.Thepotentialfor end-to-endvisibilityinmanufacturing andsupplychainswillhelpbiopharma companiesimprovetheirsupplychain planning,inventorymanagement, demandforecasting,logistics optimization,workforceplanning,and purchasing.

Biopharmacompanieswillbeableto managetheirsupply,quality,safety, security,andcostsbetteriftheycan changeprocessesandtrack medicationsinreal-time.Thiswilllead tosmartermanufacturingandsupply chaindecisions.

Thebusinessworldislikelytomove awayfromlinearsupplychainsand towarddigitalsupplynetworksthatare open,flexible,andlinked(DSNs). DSNsusedigitaltechnologieslike sensors,roboticprocessautomation (RPA),blockchain,andadvanced analyticstoimprovethevisibility, traceability,andcontrolofproducts andinventory.MLappliedto productiondatatobuildcompliance andqualityriskmodelsthathelp predictfailuresandcompliance problemsisalreadyhelpingseveral companies.Atthesametime,RPAand NLPcouldautomatethecreationand filingofcompliancereportsAItools arefinallychanging.

Cellandgenetherapies,whichare examplesofnext-generationproducts, willprobablyneedadifferentsetof skillstomakeandshipthantraditional molecules.Thesecapabilitiesshould

includeadvancedanalyticsand workflowautomationtomakeiteasy tocoordinatepatientcellcollection, manufacturingcapacity,coldchain logistics,anddeliveryofviable productstothetransfusioncenter

8.Launchandbusiness

Biopharmacompaniescouldbenefit greatlyfrombeingabletoseehow theirproductsaresoldfromstartto finish.UsingAIcanhelpcompanies planproductlaunchesbetter,get patientsmoreinvolved,helpdoctors makedecisionsandrunmarketing operations,andpredictdecisionsabout marketaccessandpricing.AItoolsare finallychangingandarealreadybeing usedtotrytoimproveengagementby customizingbehavioralnudgestoeach person’spersonality,motivations,care journey,andengagementchallenges.

Customizedengagementwith healthcareprofessionals(HCPs)could helpmakesurethattherightHCPsget therightinformationattherighttime. AIcanbeusedtodivideHCPsinto different“personas”basedontheir habits,preferences,andhowopenthey aretomarketingmessages.Tosupport reimbursementmodelsfornew curativetherapiesandservices,itwill likelybecomeimportanttogather evidencetoshowproofofvalueAI toolsarefinallychanging.

Itwillalsobeimportanttolearnmore aboutpatientmonitoringthanjust safety,andmanybiopharmacompanies andstart-upsarefocusingonthisarea. AI-poweredpatientsupportprograms couldchangethewaybiopharma interactswithitscustomers,improve enrollment,adherence,andretention, andimprovetheoveralloutcomefor patients.
