Microscopy

Selected Opti-Lab® Microscopes feature the revolutionary X-LED2 illumination system. A new high-efficiency single chip LED improved by a special optical system doubles the light intensity generated by an ordinary LED. The resulting light is comparable to a 30W halogen lamp but with a colour temperature of 6300K... which means a white light instead of the yellow one produced by filament lamps. At the same time, the system’s high efficiency uses only 10% of a 30W halogen lamp’s consumption and has a lifetime of 50,000 hours instead of the more typical 1,500 hours of an ordinary halogen lamp.

Glossary of Terms Microscope types: Compound

Diaphragm

The most common type of microscope, found in any school or microbiology lab. It is used to greatly magnify tiny specimens like amoebas or thin sections of specimens such as leaves or muscle tissue mounted on microscope slides. Compound microscopes are ideal for observing cells and cell structure. Typically they provide high-power magnification (x40 to x1600), a narrow field of view and short working distance.

There are two types of diaphragm used in microscopes:

Stereo & Dissecting These microscopes have lower magnifications (typically x10 to x45) and are designed for viewing whole, threedimensional objects such as rocks, insects, flowers, and dissection specimens. They are important for advanced nature studies and ideal for children as an object can be viewed without preparing a slide.

Microscope operating parts: Base Our bases generally house the illumination system.

1. Disc diaphragm used with the stage condenser; a revolving plate with a series of different size holes fitted beneath the stage. 2. Iris diaphragm used with the Abbe condenser; a set of leaves that can be adjusted to form a larger or smaller hole. Usually, the lower the magnification of the objective the bigger the hole in the diaphragm. The higher the magnification the smaller the hole.

Eyepiece A primary image is formed by the objective lenses; the eyepiece forms a secondary image, which is the image you see. There are two types of eyepieces in the main: 1. Widefield - allows a wide view of the specimen. 2. Huygenian - a more narrow view of the specimen. Generally graticules can be fitted to both of these eyepieces, however they may fit in different places.

Eyepiece or ocular tube

Condenser The condenser comprises a lens or set of lenses that focus the light onto the object. The two types of condenser are as follows: 1. Stage Condenser of Numeric Aperture (N.A.) 0.65 which is fixed into a hole in the stage. 2. Abbe Condenser of N.A. 1.25 which is mounted below the stage of the Microscope, concentrating and controlling the light that passes through the specimen prior to entering the objective. It has two controls, one which moves the Abbe Condenser closer to or further from the stage and another, the iris diaphragm which controls the diameter of the light beam. Abbe Condensers are particularly important for magnifications above x400.

The eyepieces fit into the eyepiece or ocular tube.

Field of view The area which is visible through the eyepiece when the microscope is in focus.

Filter holder and filters Some microscopes come with a swing-out filter holder at the bottom of the condenser. Blue and yellow filters can be used to enhance the image in some situations.

Focusing

Objective lens types

There are two types of focusing mechanisms:

Objectives come in many different designs. The most common are Achromatic, Semi-Plan and Plan:

1. Rack and pinion - separate coarse and fine focusing controls are used. 2. Coaxial focusing - combines both coarse and fine focusing controls around a common axis.

Head The area that incorporates the eyepiece tube assembly, this is either monocular, binocular or trinocular. The head on most angled eyepiece tube microscopes can rotate through 360°. In a binocular microscope, there is one optical path originating from the microscope objective. This optical path is then split into two paths that are brought to each eye. If your microscope has two eyepieces, but one objective, then chances are it is a binocular microscope. A trinocular microscope works in the same way, but the optical path is split into three paths - two for your eyes and a third port usually for a camera connection. In a stereo microscope, there are two paths originating from the microscope objective that travel essentially parallel up to the eyepieces. The advantage of using a stereo microscope over a binocular microscope is in the depth perception. Having two separate optical paths makes depth perception and three-dimensional viewing of an object possible. Using a binocular microscope, you see only a flat field and would be unable to discern any height differences on the object that you are viewing.

A semi-plan, or semi-planar objective, is an intermediate between the achromatic and the plan objective. Semiplan (sometimes called micro-plan) objectives have an 80% flat field. A plan (or planar) objective better corrects for colour and spherical aberration than either the semi-plan or the achromatic objective. Plan objectives have a flat field about the centre 95% of the image. While plan objectives give you flatter fields than achromatic objectives, they also have a higher price.

Oil immersion objective When high magnification and high resolution are required it is necessary to exclude air, which can cause surface reflections between the microscope slide and the lower surface of the objective. Exclusion of air is achieved by using a drop of immersion oil which forms an airtight connection between the two surfaces. 100x oil immersion is necessary for viewing small specimens such as bacteria and cell organelles.

Parcentered

Illumination Illumination is provided by several methods. It can either be by incident (from above) and/or transmitted (from below) in the case of Stereo Microscopes and substage for Compound Microscopes. The types of illumination include:

Magnification The factor by which the object is enlarged. Calculated by multiplying the magnification of the objective by the eyepiece.

When the condenser, objective and the eyepieces are aligned.

Parfocal The ability of the microscope to stay in focus when the objectives are changed. Applies when high magnification to low magnification is selected.

Polariser An addition to a microscope which reduces light to a single plane allowing vivid polar spectrum colours to be viewed. Ideal for use with rock section or other crystalline specimens.

Retractable Objective Spring-loaded objective that prevents microscope slide damage should the objective be lowered too far.

Slip Clutch Safety mechanism to prevent gear stripping if the coarse focus is racked down beyond the normal stop position.

Stage There are three types:

Numerical Aperture (N.A.) The Numerical Aperture of the lens determines its ability to resolve fine detail. The greater the N.A. value of the optical system (condenser and objective lens) the greater the power of resolution. (Numerical Aperture is marked on both the condenser and objective). Numerical Aperture of various optical elements: x10 Achromats 0.25; x100 Achromats 1.25; Abbe condenser 1.25

Nosepiece The part of the microscope where all the objective lenses are fitted. The nosepiece rotates allowing different magnifications of objective lens being used to view a specimen. The nosepiece of microscopes, depending on the model, can hold three or more objectives.

1. Fixed - static platform 2. Floating - adjustable in any direction for easy viewing 3. Mechanical - gives precise control of slides, includes sure-grip slide holder. Mechanical ‘2-layer’ with coaxial adjustment is available on advanced models, providing ultimate precision.

Stage Clips These hold the slide in place on the stage.

Microscopy

1.Plano/Concave mirror - usually two-sided with one side flat (plano) and the other curved (concave). Always use the plano surface with a lamp. Use the concave surface for very low N.A. objectives and with natural light. 2. LED - offers longlife illumination and produces no heat. Ideal for live specimens and offers near-natural light. 3. Fluorescent - gives an even and cool white light. 4. Halogen bulb - a low voltage lamp which is more efficient, has longer life and delivers a brighter, more intense light than tungsten. 5. Tungsten - the most common and least expensive form of illumination but brighter than fluorescent.

In an achromatic objective, the most common type, there is one achromatic lens. Achromatic objectives correct for colour and have a flat field in about the centre 65% of the image. This is not to say that the outer 35% of the image will be blurry - just that if there are aberrations, they occur in the outer 35% of the image.

Working distance (Stereo microscopes only) The distance between the top of the specimen and the bottom of the objective lens when the sample is in focus.

A Edu-Cam Multimedia Video Camera The Edu-Cam® video cameras are especially designed to meet the various requirements in the educational field. When connected to a monitor or even to a TV, Edu-Cam® is able to carry out many different functions. It can be used as an episcope, for the reproduction of texts, documents, photographs; an enlarger, to enlarge small objects, insects, minerals; for video-microscopy; an overhead projector, for the projection of drawings; a camera for conferences, assemblies, meetings; a camera for filming, with the help of a video recorder. Its ultra-high sensitivity enables clear recording even in low light. The special lens enables you to focus from 0.76mm up to infinity. An extremely sensitive microphone (only on Multimedia models), captures the teacher's voice during the lesson (or surrounding sounds) that can be heard via a TV or separate amplification system. The microphone can be switched off if required. The special flexible arm (12mm dia.) and the heavy weight of the base (approx. 2.7 kg) make Edu-Cam® versatile, sturdy and stable. The CCD camera is placed on the end of the flexible arm that can be safely twisted in any position (even projecting from the base) without affecting the system's stability. A special joint allows the head to rotate without damaging the wires inside. All models are equipped with an external power supply and dual adapter for videomicroscopy (for biological and stereo microscopes). For connecting to a PC, a USB Video Grabber (code: 601-255) is available. Electronic shutter, Automatic gain control, Automatic white balance, Microphone, Microscope adapter included.
