Inschools,twotypesofmicroscopesarecommonlyused:thecompoundmicroscopeandthestereomicroscope.
Compoundmicroscope
Alsoknownasabiologicalmicroscope,thistype usesaseriesoflenses,includingob ective lenses(typically4x,10x,40x,and100x), magnifiedfurtherbytheeyepiece(usually10x).

The numerical aperture is related to the angles of light which are collected in a lens which in turn affects the image resolution - the abilitytodistinguishdetailswithinthespecimenyouareviewing.ThehighertheNAvaluethebettertheresolution.
Please note that the numerical aperture also relates to the condenser. The numerical aperture of the condenser must be equal to or exceedthenumericalapertureoftheob ective.
For example, the ob ective pictured above has a numerical aperture of 1.25 and would be best suited to use on a microscope with an Abbecondenserwhichwillofferanumericalapertureof1.25too.Itwouldnotbeadvisabletousethisob ectiveonamicroscopewitha 0.65NAcondenserasthebestresultswouldnotbeachieved.
What is the objective tube length?
Thisisthedistancefromtheopeningofthenosepiece,wheretheob ectiveisscrewedin,to thetopoftheobservationtube,whereyouinserttheeyepiece.
Ifthisdistancewaslengthened,forexamplebyaddinganaccessorylikeapolarizer,then theimagewouldbedistorted.

Onceyouhaveobservedtheheartrateyoucancarefullyremovethewater(using theedgeofatissue,andwithouttouchingthedaphnia)andreplaceitwithyour testsolution(usuallyasolutionofethanolorcaffeine). 4.
Youcanagainrecordtheheartrateforafewminutes.
Replacethefirsttestsolutionwithwaterandallowthedaphniatorestbefore replacingthewaterwiththesecondsolution.
Onceyouhaveallyourrecordingsreplacethesecondsolutionwithwaterand returnthedaphniatothebeakerofstockwater.
Ifyourdaphniaisswimmingaboutontheslidetoomuch, youcancorralitintoalittle‘pen’madeof2cotton strands(nomorethan1cmeach):placethemaroundthe daphniaontheslidetoformatrianglewiththedaphnia trappedinside.
