Microscopes may seem complex, but they are essentially just a series of lenses that magnify tiny images, allowing us to see down to the cellular level

Inschools,twotypesofmicroscopesarecommonlyused:thecompoundmicroscopeandthestereomicroscope.

Compoundmicroscope

Alsoknownasabiologicalmicroscope,thistype usesaseriesoflenses,includingobective lenses(typically4x,10x,40x,and100x), magnifiedfurtherbytheeyepiece(usually10x).

Totalmagnificationiscalculatedbymultiplying theeyepiecemagnificationbytheobective magnification,withschoolrequirementsranging from40xto1000x.Itprovidesatwo-dimensional imagewithabrightlightsourcebeneaththe specimen.

Stereomicroscope

Usedtostudysolidsurfaces,likerocks,leaves, andflowers,orduringdissection,the stereomicroscopedirectslightfromabove.

Top tips for observing Brownian motion

Itistheuseofcontrastandfocusonthemicroscopethatallowsagoodviewofthefatparticlesofthemilkmovinginthis demonstrationanditcanbetrickytogetright.Alteringthelightlevelsonthestageaswellasthefocusingwillimprovethecontrast ontheimage.

Makeasolutionofmilkandwater:approx.10cm³waterwith2-3dropsofmilk.

Place1-2dropsofwater/milksolutionontoaslide(orcavityslideifavailable)andaddacoverslip.

Usethediaphragmonthemicroscopetoaltertheamountoflighthittingtheslidesothatthereisminimallightcomingontothe image.

The eyepiece:

Oftencomeswithafixedpointerinside,allowingyoutoshare areasofinterestontheslide:linetheimageupwiththepointer. Whereagraticulewillsitwhich,inconunctionwithastage micrometer,allowsaccuratemeasurementofthespecimenon theslide.

The numerical aperture is related to the angles of light which are collected in a lens which in turn affects the image resolution - the abilitytodistinguishdetailswithinthespecimenyouareviewing.ThehighertheNAvaluethebettertheresolution.

Please note that the numerical aperture also relates to the condenser. The numerical aperture of the condenser must be equal to or exceedthenumericalapertureoftheobective.

For example, the obective pictured above has a numerical aperture of 1.25 and would be best suited to use on a microscope with an Abbecondenserwhichwillofferanumericalapertureof1.25too.Itwouldnotbeadvisabletousethisobectiveonamicroscopewitha 0.65NAcondenserasthebestresultswouldnotbeachieved.

What is the objective tube length?

Thisisthedistancefromtheopeningofthenosepiece,wheretheobectiveisscrewedin,to thetopoftheobservationtube,whereyouinserttheeyepiece.

Ifthisdistancewaslengthened,forexamplebyaddinganaccessorylikeapolarizer,then theimagewouldbedistorted.

Choosingthecorrectthicknessofcoverslip

Foroptimumresultsusethethicknessofcoverslipidentifiedon yourmicroscopeobective

Acoverslipisusuallymadefromthinborosilicateglassandisusedtocoverspecimenstobeviewedonamicroscopeslide.

Thestandardthicknessforacoverslipis0.17mm,thisisanumber1½coverslip.Highernumbersindicatethickerglass.Foroptimum resultsusethethicknessofcoverslipidentifiedonyourmicroscopeobective.

Asmallscrewatthebackofthestagepreventsstudentsfrommovingthemicroscopeslidetooclosetothe obective,causingcontaminationordamage.Thisispre-setforeachmicroscopebutcanbeeasilyadusted.

Microscopefocusingmechanisms:

Focusinginvolvesmovingtheslidecloserorfurtherawayfromtheobective.

Entrylevelmicroscopescomewithonlyacoarsefocusasthemagnificationrangetendstobelower.

Mid-rangemicroscopescomewithseparatecontrolforcoarseandfinefocus.Turnthecoarsefocus slowlytogetthespecimenintofocus.Thenmakesmallfinaladustmentswiththefinefocustogeta sharpimage.

Coarsefocusshouldbeusedforlowermagnification,switchingtofinefocusathighermagnification. Somemicroscopeshaveatensionadusteronthecoarsefocuswhichhelpsifthesamplebeingviewed driftsoutoffocus.YourBMSmicroscopewillcomewithasmallwrenchthatcanbeusedtotightenthis. Higherspecificationmicroscopesareequippedwithcoaxialfocusingwhereboththecoarseandfine focusingareonthesameaxle.Thisismoreconvenientastheuserdoesnothavetotrytofindthecoarse andfineknobswhilststillviewingtheirsample.

Theentryrangeofmicroscopeshaveafixedcondenserandmoveablediaphragm.Thiscanbeopenedorclosed,allowingmoreorless lighttohittheslidefrombelow. Thewiderthediaphragmthelowerthedepthoffieldbutbetterresolution,themoreclosedthe diaphragmis,thelowerresolutionwithahigherdepthoffield.

What is birefringence?

Birefringenceoccurswhenarayoflightisrefractedthroughanobectin2differentdirections,givingadoubleimagewhenyoulook throughit.Apolarizingfilterwilleffectively‘remove’oneofthoseimages.

Polarizationcanbeusedwithbothstereoandcompound microscopes.

Stereomicroscopes:

Polarizationcanreducetheglareofreflectedpartsofthe specimen.
