Introducción Introduction

Hemos llegado a un punto sin retorno con la extracción y el uso de nuestros recursos naturales. Como diseñadores desempeñamos un papel importante en la elección y el uso de materiales, nuestras elecciones deben estar dirigidas hacia aquellos que causen el menor impacto ambiental. [1] Este proyecto busca el desarrollo de objetos utilizando micelio. El micelio es el conjunto de células filamentosas altamente interconectadas, las cuales forman estructuras similares a redes. Esta constituye la fase de crecimiento vegetativo de los hongos, la cual permanece durante todo su ciclo de vida. El micelio puede descomponer la materia vegetal degradando extendiendo su red hifal. El crecimiento del micelio se produce mediante la colonización de sustratos, principalmente vegetales, y se asocia directamente a su alimentación por degradación del mismo. A diferencia de los materiales convencionales, este material crece y, por lo tanto, tiene un proceso de desarrollo diferente.

Cuando se combina, el micelio coloniza y degrada parcialmente el sustrato para crecer y a través de este proceso une todas las partículas a su alrededor creando un material compuesto.

Debido a sus propiedades mecánicas, este material puede obtener cualidades análogas a la espuma de poliestireno y otros plásticos que se utilizan en diversas industrias basadas en agua y petróleo.

El micelio en particular, puede generarse a partir de especies locales y materias primas, representando una alternativa atractiva a los productos sintéticos tradicionales, ya que es 100% natural, totalmente compostable y resulta de corrientes de desechos.

Este publicación busca difundir el proceso que involucra cultivar un microorganismo hasta crear un biomaterial. Cada una de las etapas del proceso es descrita, desde su cultivo, inoculación y mezcla hasta las piezas de diseño obtenidas de dicho proceso. La gama de objetos y artefactos desarrollados son parte de una investigación que opera en la intersección del diseño y la ciencia, ya que trabaja en consulta directa con científicos, adoptando y desarrollando metodologías y técnicas novedosas, como parte de su práctica.

El contenido que se encuentra en esta publicación fue desarrollado gracias al apoyo del Fondo Metropolitano de Cultura, las Artes y las Ciencias de la Ciudad de Buenos Aires y el Fondo Nacional de las Artes de Argentina.

We have reached a point of no return with the extraction and use of our natural resources.

As designers we play an important part in choosing and using materials, our choices should be targeted at materials causing the lowest environmental impact. [1] This project seeks the development of objects using mycelium. The mycelium is a rhizomatic network that constitutes the vegetative part of fungi. It can break down plant matter by degrading it by spreading its network of hyphae. Unlike conventional materials, this material grows and, therefore, has a different development process.

When combined, the mycelium obtains nutrients from the substrate to grow and through this process binds all loose particles around it creating a composite material. Due to its natural ability to fuse very small pieces of organic material into a larger whole, this material can obtain qualities similar to polystyrene foam and other plastics that are used in various oil and water-based industries.

This publication seeks to disseminate the process that takes to cultivate a microorganism until the creation of a biomaterial. Each stage of the process is described, from its cultivation, inoculation and mixing to the design pieces obtained from the process. The range of objects and artifacts developed are part of a research that operates at the intersection of design and science, since it works in direct consultation with scientists, adopting and developing new methodologies and techniques, as part of its practice.

This publication was developed thanks to the support of the Metropolitan Fund of Culture, Arts and Sciences of the City of Buenos Aires and the National Fund of Arts of Argentina.

Mycelium in particular can be generated from local species and raw materials, representing an attractive alternative to traditional synthetic products, since it is 100% natural, fully compostable, and resulting from waste streams.

¿Qué es el micelio?

What is Mycelium?

Si bien podemos admirar un hongo que crece del suelo o en un tronco, es el micelio que está fuera de la vista (y a menudo olvidado), siendo éste la parte esencial del organismo.

El micelio se forma durante la etapa de crecimiento vegetativo del ciclo de vida de los hongos y consiste en una red de filamentos interconectados llamadas hifas. El micelio se lo puede encontrar en el suelo y en diferentes sustratos vegetales como troncos, hojas, corteza, etc. A pesar de aparentar ser un objeto estático, este forma parte del ciclo de vida de los hongos por lo que crece, se reproduce y muere. Los hongos interactúan con su entorno modificando su crecimiento respecto a diferentes condiciones ambientales, recursos y otros organismos, produciendo diferentes formas o estructuras de crecimiento.

Cada hifa crece en longitud y es capaz de ramificarse, generando una gran red intercomunicada en donde se produce el transporte de nutrientes y otros compuestos, que favorecen su crecimiento. El micelio a medida que crece se expande colonizando su sustrato. [2] Debido a su metabolismo absorbotrófico los hongos requieren que sus nutrientes sean degradados en el ambiente.

Para esto secretan a través del ápice de las hifas, enzimas degradadoras una gran cantidad de compuestos como polisacáridos, proteínas, lípidos, fenoles, etc.

Una vez degradados estos son absorbidos y asimilados por el metabolismo. En la naturaleza los hongos cumplen principalmente el rol de degradadores de materia orgánica y dependiendo de los sustratos de los que son capaces de colonizar y degradar se hace referencia su metabolismo. En nuestro caso vamos a trabajar con hongos descomponedores de madera y dentro de este grupo, con hongos de pudrición blanca (White Rot Fungi). Este grupo se caracteriza por ser el único capaz de degradar completamente la madera ya que puede actuar sobre la lignina, uno de los constituyentes de la pared celular vegetal y de los polímeros más recalcitrantes de la naturaleza. [3] While we can admire a fungus that grows from the ground or on a log, it is the mycelium that is out of sight (and often forgotten), this being the essential part of the organism. The mycelium is formed during the vegetative growth stage of the fungal life cycle and consists of a network of interconnected filaments called hyphae. The mycelium can be found in the soil and different plant substrates such as logs, leaves, bark, etc. Despite appearing to be a static object, this is part of the fungal life cycle, so it grows, reproduces and dies. Fungi interact with their environment by modifying their growth for different environmental conditions, resources and other organisms, producing different forms or structures of growth.

Each hypha grows in length and is capable of branching, generating a large intercommunicated network where the transport of nutrients and other compounds occurs, which favor their growth.

The mycelium as it grows expands colonizing its substrate. [2] Due to their absorotrophic metabolism, fungi require that their nutrients be degraded in the environment.

For this they secrete through the apex of the hyphae, degrading enzymes a large number of compounds such as polysaccharides, proteins, lipids, phenols, etc. Once degraded these are absorbed and assimilated by the metabolism.

In nature, fungi mainly play the role of degraders of organic matter and depending on the substrates of which they can colonize and degrade their metabolism is referenced. In our case we will work with wood decomposing fungi and within this group White Rot Fungi. This group is characterized by being the only one capable of completely degrading wood since it can act on lignin, one of the constituents of the plant cell wall and the most recalcitrant polymers of nature. [3]

El micelio como biomaterial tiene una gran variedad de propiedades. Estudios indican que, estas dependen de la especie fúngica utilizada, los sustratos, las condiciones de crecimiento y el procesamiento del material, así como sus aditivos.[4]

Ganoderma Lucidum (Reishi) Para realizar el Biomaterial se utilizó el hongo de pudrición blanca Ganoderma lucidum (Reishi).

Hacen parte del grupo de los hongos filamentosos, los cuales tienen la capacidad de descomponer la madera ya que producen enzimas que le ayudan a degradar la lignina y/o celulosa.

The white rot fungus Ganoderma lucidum (Reishi) was used to perform the Biomaterial.. They are found within the filamentous fungal group. This fungus can break down wood and degrade it by producing enzymes that help degrade lignin and/or cellulose.

La semilla de Reishi se comercializa para la producción de hongos comestibles y se pueden encontrar fácilmente en el mercado.

Reishi seed is marketed for the production of edible mushrooms and can be easily found in the market.

Aserrín Sawdust Para la producción del aglomerado se recomienda utilizar aserrín de madera blanda no resinosa como el álamo. También se puede utilizar pino o eucalipto los cuales contienen resinas o fenoles que pueden impactar en la velocidad de crecimiento del hongo, pero no en la formación del aglomerado. En lo posible evitar aserrín que provenga de MDF y aglomerados ya que puede tener resina y pegamento.

For the production of the chipboard it is recommended to use non-resinous softwood sawdust such as poplar. You can also use pine or eucalyptus which contain resins or phenols that can impact the growth rate of the fungus, but not the formation of the agglomerate. If possible, avoid sawdust that comes from MDF and agglomerates as it may have resin and glue.

Semilla de avena Oat seed Utilice para el cultivo del hongo la semilla de avena sin ningún tratamiento, es decir que no sea la avena arrollada.

Use oat seed for the cultivation of the fungus without any treatment, i.e. it is not the rolled oats.

Autoclave Se utiliza para esterilizar los sustratos y las herramientas que vamos a manejar durante el proceso. Puede ser reemplazado por una olla a presión.

It is used to sterilize the substrates and the tools that we are going to handle during the process. It can be replaced by a pressure cooker.

Es un espacio diseñado para material manipular en esterilidad y evitar la contaminación de material biológico. Este tipo de instrumental es propio de laboratorios de cultivo, por lo que no siempre está a disposición. También se puede utilizar en reemplazo un mechero ya que, al igual que la campaña de flujo laminar, establece flujo de aire estéril y crea una zona aséptica de trabajo.

It is a space designed to handle material in sterility and avoid contamination of biological material.

This type of instrument is typical of culture laboratories, so it is not always available. An alcohol burner can also be used as a replacement because, like the laminar flow campaign, it establishes sterile air flow and creates an aseptic work zone.

Cámara de cultivo Temperature chamber El micelio debe mantenerse en un rango de temperatura ideal para mejorar su crecimiento.

Se puede usar como alternativa un espacio desinfectado en donde se pueda controlar la temperatura.

Growing should be kept in an ideal temperature range to improve its growth. The alternative is a disinfected space where the temperature can be controlled.

Horno Oven La etapa final de la producción del aglomerado constituye su secado, por lo que se necesita un horno o estufa. Las piezas se secan a 80° durante 8 horas aprox. Otra posibilidad es dejarlo secar al sol por varios días.

The final stage of agglomerate production constitutes its drying, so an oven or stove is needed. The pieces are dried at 80 ° for 8 hours approx. Another possibility is to let it dry in the sun for several days.

Protocolos de limpieza Cleaning protocols Usar guantes siempre que se está en contacto con el hongo, salvo en el caso en que esté expuesto a fuego como en las etapas en las que se utilizan mecheros o esterilizadores.

Wear gloves whenever you are in contact with the fungus, except in the case where it is exposed to fire as in the stages in which lighters or sterilizers are used.

Limpiar todos los elementos a utilizar (moldes, instrumentos, vasos, guantes, etc) con mezcla de alcohol y agua ( 70% de alcohol (etanol) y 30% agua destilada).

Clean all the elements to be used (molds, instruments, glasses, gloves, etc.) with a mixture of alcohol and water (70% alcohol (ethanol) and 30% distilled water).

Los elementos que se usen para manipular material estéril, aparte de desinfectarlos con alcohol 70%, se deben esterilizan por incineración, esto se puede hacer con el mechero (de alcohol o gas) o con un incinerador. Para esto se somenten los elementos metalicos a la llama sin soltarlos (con sumo cuidado ya que levanta mucha temperatura) se espera hasta observar un cambio en la coloración del metal, se saca y enfría la zona que va a entrar en contacto con el material estéril sumergiendolo en un frasco con alcohol (etanol 100%) y después se lo usa inmediatamente.

The elements used to handle sterile material, apart from disinfecting them with 70% alcohol, should be sterilized by incineration, this can be done with the lighter (alcohol or gas) or with an incinerator.

For this the metallic elements are subjected to the flame without releasing them (with great care since it raises a lot of temperature) until a change in the color of the metal is observed, the area that will come into contact with the sterile material is removed and cooled immersing it in a bottle with alcohol (100% ethanol) and then using it immediately.

Es muy importante que el espacio de trabajo permanezca limpio durante todo el proceso. La contaminación es el desafío más común que encontrará al cultivar micelio, es decir evitar tener moho no deseado en su experimento.

It is very important that the workspace remains clean throughout the process. Pollution is the most common challenge you will face when growing mycelium, that means, avoid having unwanted mold in your experiment.

La avena se debe hervir en agua durante 1 hora aprox y luego escurrirla con un colador. Una vez que ya no caiga agua se mete en un frasco de vidrio llenando hasta la mitad del volumen.

Los frascos con avena, se ponen con la tapa floja en el autoclave durante 30 min a 121°. Después se deja enfriar y se cierran los frascos dentro del autoclave para evitar contaminaciones.

The oatmeal should be boiled in water for about 1 hour and then drained it with a strainer. Once water no longer falls, it gets into a glass jar filling up to half the volume.

The jars with oatmeal, are placed with the lid loose in the autoclave for 30 min at 121°C (250°F). Then it is allowed to cool and the jars are closed inside the autoclave to avoid contamination.

En la cámara de flujo laminar, utilizando guantes, desinfectar la superficie y todos los elementos de trabajo rociando con alcohol 70% y esparciendolo con papel absorbente. La pinza y otros elementos metálicos deben ser estirilizados con calor usando el mechero.

Pasar un pedazo no muy grande material colonizado (inóculo) por el reishi al frasco con avena. Para trabajar con semilla se debe ser muy cuidadoso con todo el material ya que se puede infectar muy fácil.

In the laminar flow chamber, using gloves, disinfect the surface and all work items by spraying with alcohol 70%. The clamp and other elements must be sterilized with heat.

Pass a not very large piece of colonized material (inoculum) by the reishi to the jar with oatmeal. To work with seeds you must be very careful with all the material since it can be infected very easily.

El frasco de vidrio con avena inoculado, se mete en la camara de cultivo a 28°C y en oscuridad dejando la tapa un poco abierta. Cada 2-3 días ir agitando el frasco de vidrio para que la colonización sea homogénea (cerrar el frasco para agitar y después dejarlo un poco abierto de nuevo).

Una vez la semilla este completamente colonizada por el micelio es momento de mezclarla con el aserrín.

Tiempo de crecimiento en semilla: 5/6 días.

Once the seed is completely colonized by the mycelium, it is time to mix it with the sawdust.

Seed growth time: 5/6 days.

The glass jar with inoculated oatmeal is placed in the culture chamber at 28°C (82°F)in the dark leaving the lid a little loose. Every 2-3 days shake the glass jar so that colonization is homogeneous (close the jar before shaking it and then leave it a little loose again).

Cultivos en medio agarizado Culture media Una de las formas de conservar el inĂłculo es realizar cultivos en medio agarizado. Para esto se arman medios de cultivo ricos como extracto de malta-glucosa y se les agrega agar. El agar es un gel similar a la gelatina compatible con el crecimiento de microorganismos como los hongos. Este se coloca en placas de petri y se agrega inĂłculo el cual crece y luego permite multiplicar el micelio.

One of the ways to conserve the inoculum is to perform cultures in an agarized medium. For this, rich culture media such as malt-glucose extract is assembled and agar is added. Agar is a gel similar to gelatin compatible with the growth of microorganisms such as fungi. It is placed in petri dishes and inoculum is added which grows and then allows the mycelium to multiply.

Este procedimiento se realiza en la cabina de flujo laminar o en un ambiente esterilizado.

Se llenan las placas de petri hasta la mitad y se espera hasta que se solidifique el agar.

Se toma un pequeĂąo pedazo del hongo con una pinza esterilizada y se coloca en el centro de la placa de petri encima del medium de agar. Es importante inocular al menos tres placas para aumentar la probabilidad de crecimiento.

This procedure is performed in the laminar flow cabinet or in a sterilized environment. The petri dishes are filled halfway and wait until the agar solidifies.

A small piece of fungus is taken with a sterilized clamp and placed into the petri dish on top of the medium agar. It is important to inoculate at least three plates to increase the probability of growth.

Para preparar el aserrín se lo coloca en contenedores capaces de resistir temperaturas cercanas a 121°C llenandolas hasta la mitad.

Una vez estén listas se tienen que humedecer hasta llegar al 80% de humedad (lo que pesa el sustrato + X de agua)

La ventilación es es un factor clave tanto para el crecimiento de los cultivos, como tambien para que el vapor del autoclave llegue a la madera y logre esterilizarla. Para esto se puede crear un tapón usando un caño de pvc con algodón y gasa. Este paso se puede reemplazar por “bolsas de cultivo” que contengan filtros.

Ventilation is a key factor for both, the growth of crops, as well as for the steam in the autoclave to reach the wood and achieve sterilization. For this, a plug can be created using a pvc pipe with cotton and gauze. This step can be replaced by plastic bags containing filters.

Una vez listas las bolsas se autoclavan durante 45/60 min a 121°C. Es importante que el aserrín esté bien humedecido.

La mezcla de los materiales se realiza en un ambiente estéril como la cámara de flujo laminar, para esto se combinan 4 partes de aserrín con 1 parte de mezcla de semilla micelada (25%) en la bolsa de aserrín. Cierre la bolsa inmediatamente con el anillo y el tapón de algodón, mezclar hasta conseguir una distribución homogénea.

Once the bags are ready, they are autoclaved for 45/60 min / at 121°C (249,8°F). It is important that everything is well moistened.

The mixing of the materials is carried out in a sterile environment such as the laminar flow chamber, for this, 4 parts of sawdust are combined with 1 part of micelled seed mixture (25%) in the sawdust bag. Close the bag immediately with the ring and the cotton cap, mix until you get a homogeneous distribution.

1. Colocar en los moldes Place in the molds

Una vez preparado el material, es el momento de ponerlo en los moldes. El molde debe ser fácil de esterilizar, limpiar y fácil de desmoldar para poder sacar las piezas una vez estén listas.

Es importante que los moldes tengan agujeros para que haya intercambio de aire durante el crecimiento.

Desinfectar los moldes con alcohol 70% y esparcir con el papel hasta asegurarse de que el alcohol se haya evaporado. Volcar la mezcla de aserrín inoculado a los moldes. Rellenar ejerciendo una leve presión para que la mezcla alcance todo el volumen y tapar con film.

Once the material is prepared, it is time to put it in the molds. The mold should be easy to sterilize, clean, and unmold in order to remove the pieces once they are ready. It is important that the molds have holes for air exchange during growth.

Disinfect the molds with alcohol 70% and spread with the paper until making sure that the alcohol has evaporated. Dump the sawdust mixture inoculated to the molds. Fill with light pressure so that the mixture reaches full volume and cover with film.

Llevar los moldes a la cámara de cultivo a 28°Cy en oscuridad por 7 días o el tiempo que le tome al micelio colonizar todo el sustrato. El micelio crecerá adoptando la forma del molde. Una vez este completamente blanco se debe desmoldar.

Una vez desmoldada, la pieza fuera del molde puede estar 7 días más dentro de una bolsa o un contenedor más grande en la cámara de cultivo cuidando que no se seque. Después se debe secar el biomaterial a 80°C hasta alcanzar su sequedad. La forma de saber esto es que pesar ell objeto durante el proceso de secado y continuarlo hasta que permanezca constante.

Put the molds into the culture chamber at 28°C (°F82,4 )and in darkness for 7 days or the time it takes for the mycelium to colonize the entire substrate. The mycelium will grow by adopting the shape of the mold. Once this completely white must be unmold.

Once unmolded, the piece outside the mold can be 7 days more in a bag or a larger container in the culture chamber taking care not to dry. Then the biomaterial should be dried at 80°C (249,8°F). until it reaches dryness. The way to know this is to weigh the object during the drying process and continue it until it remains constant.
