1 | General Summary The TN9000 Semi-Automatic Cryostat Microtome is a medical device used for rapid freezing of pathological sections of human and animal tissue. It can be widely used for pathological diagnosis, analysis and research in hospitals, medical colleges, and by legal medical experts and institutes.
Instrument Consists of 4 Major Parts: • The touch screen display is located on the upper part of the instrument. This area displays the general working conditions and the temperature of the instrument. • The chamber area is used to rapidly freeze the tissue and to conduct cutting. The microtome assembly is located inside the chamber area. • The compressor, cooling condenser and evaporator are located in the lower part of the instrument. • Behind the chamber is the mechanical transmission and motor drive.

2 | Safety Precautions This user manual includes important instructions and data related to the operating safety and maintenance of the instrument. The user manual is an important part of the product. It should be read carefully before using the instrument for the first time and must always be kept with the instrument. This cryostat may only be operated by qualified personnel. In case of malfunction, contact a qualified service technician. Never try and solve the problem yourself.
Power Supply • A professional electrical engineer should inspect the electrical circuit before installation to ensure smooth operation of the instrument. • The instrument uses a single phase AC power supply: the voltage is 110V±10%. If the power does not meet this requirement, then a 2000-3000VA AC voltage regulator shall be connected for usage. • A dedicated 20AMP circuit is required. • Confirm the power supply is well grounded (triple socket) before connecting the instrument with the main power socket. Turn on the power switch in the back of the instrument to start the machine.
Temperature • The cryostat’s working (environment) temperature is 5°C ~ 23°C (40°F ~ 73°F).
High temperature or humidity will impact the chamber temperature and shorten compressor life.
Operating • Exercise extreme caution when handling blades. Always remove when cleaning the inside of the chamber. Never place a blade face up as the sharp surface can lead to serious injury. • NEVER TRY TO CATCH A FALLING BLADE! • Before changing specimens, always lock the hand wheel and cover the blade edge with the blade guard. Make sure the hand wheel is in the locked position when the instrument is not in use.

4 | Installation Transportation/Unpacking • Unit must be shipped in the upright position. If the unit was shipped or stored lying down, it must stand upright for 36 hours before use so all lubricating oils enter the compressor. It should only be used after 4 hours of standing upright to ensure proper lubricant distribution. • Inspect the package for damage. If damaged, do not open and immediately contact Tanner Scientific ® and the transportation carrier.
Site Requirements • Do not operate in rooms with or near flammable chemicals. • Place instrument at least 10” from other instruments. Areas on both sides of the instrument should be clear to allow proper air flow to the cooling unit. • Do not place in rooms above 75°F or in a humid environment directly in sunshine, OR NEAR AC VENTS OR CEILING FANS. The environment’s relative humidity should be no more than 80%. Failure to follow this procedure could create frost and icing in the chamber.
Placement • The cryostat is a movable floor model with 4 caster wheels and 2 stabilizing legs in the front that are adjustable to level the instrument.
Hand Wheel Assembly • Insert the pin of the hand wheel shaft into the hole of the hand wheel. • Mount the spring washer on the screw (M10x15). • Tighten the screw with a #6 Allen key.
Freezing Chamber Assembly • Place the waste tray in the freezing chamber. • Install blade holder on the microtome base and lock. • Place all tools needed for specimen preparation in the chamber. • Close glass window. • Connect power.
Check your power socket to ensure it is well grounded and that the power supply is stable, constant and accurate.

| Blade Holder Adjustment
Turn the blade holder lock handle clockwise to release it. Manually move back and forth and lock in the desired position. When the holder moves to the terminal point, the entire blade holder can be taken out and put back by reversing the procedure. Turn (counter clockwise) the blade holder lock handle. Manually move left and right and lock in the desired position. Notice: Remember to lock the blade holder in the correct position after moving it forwards or backwards.

| Changing the Blade
Release the clamping lever on the upper right to open the blade holder. Insert blade from left to right and then lock it in place.
Note: Blade is extremely sharp. Use caution when handling or replacing blades.

| Adjusting the Angle of the Blade
Loosen the lowest adjustment handle on the right side of the blade holder. Refer to the reference scale to turn the rotator in order to choose the best cutting angle of the blade. The angle should be no more than 10°. Retighten the adjustable handle.

| Adjust Anti-Roll Plate Angle with Blade
1) Make the edge of the anti-roll plate parallel to the edge of the blade. • Insert blade and lock the clamping lever. • Lock two knobs. • Use hexagonal wrench to make blade and anti-roll plate parallel. • Fasten hexagonal screw. • Turn knob to adjust height of the anti-roll plate. 2) Make the edge of anti-roll plate and the edge of the blade flush. • Turn height turning knob to adjust height of the anti-roll plate. • Adjust accordingly until you get quality sections. Adjust with Hex Wrench. Make the Edge of Anti-Roll Plate and Edge of Blade Flush.
1) The upper part of the anti-roll plate should not touch the specimen. 2) When the quality of the tissue section is lacking, check the edge of the blade for foreign matter. 3) Avoid touching the front of the plate.

| Specimen Disc Installation
• Adjust the specimen clamp accordingly. • After the specimen has been frozen on the specimen disc, insert specimen disc shaft into the hole in the center of the clamp. Join the front surface of the clamp with the rear surface of the specimen disc. Rotate the screw to lock the front of the clamp. • Loosen adjustable clamp screw, turn the lever to adjust the direction of the specimen disc surface and lock.

5.3
| Freezing Specimen
The specimen freezing station is located at the left side of the chamber. There are 18 holes to place the specimen disc, 2 of which are semiconductor refrigeration holes. The temperature of the freezing station is always colder than the freezing chamber’s temperature. • At room temperature, cut the specimen to an appropriate size. Place on disc and fix with tissue freezing medium. • Insert specimen disc with specimen into the freezing hole. • Hold the freezing clamp on the specimen for about 30 seconds or until specimen is completely frozen. • After specimen is frozen, insert specimen disc into the specimen clamp, lock, and begin sectioning process.

| Defrost
In some situations, it makes sense to allow the machine to defrost and eliminate excess ice buildup. When the defrost cycle runs, ice will melt and drain into the waste container. • The defrost cycle runs for almost 15 minutes. • Upon completion of the defrost cycle, the machine will automatically return to its original cold state.
Automatic Defrost Timer Set the automatic defrosting time accordingly. When the chamber’s temperature reaches -5°C, defrost automatically terminates.
Instant Manual Defrost Set the manual defrost at any time. It can be turned off if needed.

Delay Startup Mode When the compressor’s downtime is less than three minutes, and is started again, the compressor will enter delay startup mode. The compressor will become fully operational after 3 minutes.

Note: UV disinfection will only activate when the glass door is closed, which means the UV lamp can not work when the glass door is open. When the lamp is on, opening the door causes the light to automatically turn off. Close the glass door and press the UV button, the lamp will light up again.

Set the Time of Day the Cryostat Enters Defrost Mode and Sleep Mode.

1) Manual Defrost 2) Automatic Defrost Settings from Sunday to Saturday. Press square below each day to activate. Selected Unselected 3) Timer. Press the black square to display timer. Choose hour and minutes. Press to increase or decrease time. Click SAVE or CANCEL to save or cancel the time setting. Each defrost time takes 10 minutes.

| Sleeping Mode Setting
1) Manual Sleeping Mode. Chamber should keep running at -5°C while the display light dims. To deactivate sleep mode, press the button at any time. 2) Automatic Sleeping Mode. Without any activity after two hours, the machine automatically enters sleep mode. 3) Automatic Sleeping Mode Start Time and End Time. Note: After setting up the time, you need to press the SAVE button. Otherwise, the setting will not enter sleep mode.

| System Time Setting
1) Select the box under the workday you want to set a time. refers to the corresponding workday you selected. 2) Select the black square to display time. Choose hour and minutes. Select to increase or decrease time.
3: Press SAVE to save the setting. At the same time, the gray bar in the lower right corner of the screen will display your set time.

| Setting Retraction Value of Section
Select (+/-) to increase or decrease the retraction value. The range of retraction value is 0-100μm with 5μm increments. REMEMBER TO SAVE.

| Course Feed Control Panel
TIP: Before adjusting any specimen, make sure the hand wheel is locked.
Located to the left of the main chamber, the course feed control panel allows you to advance or retract the specimen. Additionally, you can control the thickness by using the +/- buttons. You can increase the speed at which the specimen advances or retracts by holding down the “F” button at the same time pressing the or buttons.

| Trimming
Trimming can be controlled on the display screen and the touch pad located to the left of the chamber. Use the +/- buttons to increase or decrease the trimming thickness with each turn of the hand wheel. Trimming allows you to cut away unwanted portions of the specimen. • Place the blade into the holder and lock. Adjust the anti-roll plate. • Adjust the blade holder’s angle 1°~2°, or to the angle that works best. • Adjust holder and sample, but keep enough space for depth of cutting. • Open the anti-roll plate. • Unlock the hand wheel. • Move the sample close to blade. Turn the handwheel to section. Thickness can be set to 20μm. Reduce gradually to the desired thickness. • Close the anti-roll plate. • Turn hand wheel to trim excess specimen away.

5.9.2
| Sectioning Mode
• Press the “Switch” button to enter sectioning mode. • Turn the hand wheel one rotation. The instrument will cut one piece of section at a time. • It’s best to turn the hand wheel at uniform speed. • Adjust speed until you receive quality sections.
5.9.3
| Sectioning Considerations
1) When using the cryostat, controlling the blade’s speed and correctly adjusting the anti-roll plate are important factors in cutting high quality sections. The proper speed of the blade can only be acquired by practice and experience, along with skillful control of the hand wheel. The proper adjustment of the anti-roll plate requires even more practice. Sometimes they interfere with each other and require careful adjustment. 2) When freezing tissue, the water within the tissue will condense into ice and the tissue will harden. The hardness changes with the temperature: the lower temperature, the harder the tissue becomes. Each tissue reacts differently under various temperatures. Achieving high quality sections can only be obtained through practice. Cutting temperatures of the specimen tissue without fat and formalin should be between -13°C ~ -25°C. 3) For high quality sectioning, pay attention to the following: • Proper working temperature in the chamber • Correct hand wheel speed • Fine adjustment of the anti-roll plate • Sharp blade edge and correct blade angle 4) Frozen sections should not be cut in wide sections. Cutting thinner sections, so that the line of contact is shorter, is ideal.

| Maintenance
5.10.1
| Daily Finishing Work
• Lock the hand wheel. • Remove the blade from the holder accordingly. • Remove waste from the chamber. • Clean blade holder and brush holder. Tip: General cleaning can be used with 95% ~ 98% alcohol.
• Close the glass window. • Turn off the lights. • Do not turn off the main switch. Do not stop the cooling system when the compressor is running.

| Power Up/Down
If you go several weeks or longer without using a machine, you can turn off the main power switch. • Turn off the machine’s power switch when compressor is not running. • Open the glass window, ventilate the freezing chamber and let dry. • Remove all specimens from the freezing shelf. • Lock the hand wheel. • Remove the blade from the blade holder. • Remove waste. • Remove the dust pan and brush to clean. Before restarting the machine, make sure all parts in the freezing chamber are thoroughly dry or icing could occur.

6 | Cleaning & Maintenance
Since these machines are precision instruments, it’s important to pay attention to cleanliness, maintenance and repair work. Do NOT use organic solvents or any aggressive substances to clean and sterilize the chamber! Only use the cleaning agents specified in this manual. If necessary, the external surface may be cleaned with light-duty commercial household cleaner. Use a wet cloth and rub until dry. Do NOT apply xylene or glass cleaner.

| Daily Cleaning
• Dispose of any waste daily. • Clean the waste pan. • Clean the brush holder. • Waste water treatment must comply with local environmental regulations. • Observe the water level in the bucket and discard accordingly.

| Maintenance
• Clean the machine daily. • Add droplets of low temperature lubricating oil on the plastic coupling interface every week. • Lubricate the Specimen Feed Mechanism Weekly: Click the fast forward button to make the clamp move to the front position, drip a drop of the low temperature lubricating oil on the copper bushing, then click the fast rewind button to make the clamp move to the back position. • Lubricate the Eyelet Cover As Needed: Turning the hand wheel, drip some drops of the low temperature lubricating oil on the decorative sheeting when the specimen clamp on the highest and lowest position, then turn the hand wheel again to make the oil distribute evenly. • Clean the Condenser As Needed: Open the vent panel at right of the machine, clean the dust on the cooling fin of the condenser with brush, or jet it with high-pressure airflow.

| Replacing Fuses
• Turn off the machine and unplug the power cord. • Replace the Fuse: There are 5 fuse boxes in the back of the instrument. Open the cap and remove the broken fuse tube. Insert a new fuse into the lid. Close the lid on the fuse box and tighten. Note: You can only use the same type of fuse! 1

| Waste Bottle
The waste bottle is used to collect the liquid from defrosting tissue, washing fluid and waste. Pour 200mL of 10% formalin solution or other disinfectant into the water bucket in order to prevent waste damage on personnel and environment. Waste discharge should be in accordance with local regulations. Use correct methods to treat the waste water and waste.
