A Sensitive Amplified Amperometric DNA Sensor Using Hybrid Nano-materials Probing for Human Papillomavirus (HPV) Detection *

Results

Human papillomavirus (HPV) is although virtually induced in each type of cervical cancers. Epithellial cells which are infected from the high risk human papillomaviruses are also associated with cervical cancer . The molecular epidemiologic studies provide risk estimates and functional evidence of oncogenic capacity of different types of HPV. Electrochemical detection of hybridization is mainly based on the differences in the electrochemical behaviour of the labels with or without double-stranded DNA (dsDNA) or single-stranded DNA (ssDNA). The hybridization between PDNA (probe-DNA) and TDNA (Target-DNA) was detected by reduction in current, generated by interaction of anionic mediator, i.e., methylene blue (MB) with free guanine (3′G) of ssDNA.

Introduction

The proper diagnosis of HPV infection is essential for the prevention of cervical cancer. The implementation of effective electrochemical DNA sensor is introduced for the detection and identification of HPV types. Electrochemical detection of hybridization is mainly based on the differences in the electrochemical behavior of the labels towards the hybridization reaction on the electrode surface or in the solution. Use of functional nanomaterials offers new possibilities of improving the performance of electrochemical sensors due to their unique array of structural and chemical properties. GONRs are highly soluble in water and polar solvents, with an attractive surface and edges containing oxygen functional groups. Bimetallic gold-silver core-shell nanoparticles were prepared by chemical reduction in aqueous solution, following a method that was friendly to the environment, allowing us to use this for medicinal purposes. The objective of this study is to develop and evaluate biosensor prototype for detection of high-risk HPV types.

The DNA sensor for rapid detection of Human Papillomavirus using GONR@Ag coated AuNP was developed. The DNA-based electrochemical sensor has a wide linear range, low detection limit, good reproducibility and stability. Therefore the development of biosensor based technology for high risk HPV genotype detection provided a potential tool in diagnosis.
