Cool Crones et al!
Dear Reader
Welcome to another year! Over this festive season, I have been contemplating what 2025 may hold for many of us. Having thought I would have two weeks of sunshine in Johannesburg - time to swim, time to start considering my new year’s resolutions (which, by the way, I never make!) - I now realise I was wrong. Rain, rain and more rain. But on reflection, how could I possibly be even mildly irritated by this. Gratitude is the only word that should be rolling off my tongue. How fortunate are we to have had so much rain, and to see our surroundings so lush.
So, that being said, I have been able to mull over this past year, which I don’t think has been an easy one globally, and to focus on what I hope will be some positive trends in 2025.
From AI-driven innovation to eco-ambition and the rise of older women as powerful leaders, here are the currents driving change and transformation.
AI and us
AI is clearly going to play a major role in our future. According to Forbes, the global AI market is projected to grow annually by 36.6% until 2030, and by next year many businesses (64%) expect it to routinely work alongside us to boost productivity and creativity, assisting with research, writing and problem-solving. In areas such as customer service and translation, AI will really come into its own, while quantum AI, although still in its early stages, has the potential to transform fields such as medicine by vastly accelerating the amount of data that can be processed.
Downsides? There are commonplace predictions that jobs destroyed by technology will be replaced by new openings, but there's a frustrating vagueness about where these new roles will appear, other than in AI-related tasks such as prompt engineering. It seems unlikely, say, that a coder whose once-prized skills are now surplus to requirement will happily retrain as a low-paid carer. According to estimates, almost half of current employees will need further training to adapt to new technologies. Will companies with one eye on a lower headcount foot the upskilling bill? In short, AI is a positive tool – but only if we take the necessary steps to ensure it doesn’t just leave people behind.
Trump effect
Whatever your view of the US election result, most of the consequences of chapter two of the Donald Trump presidency remain a matter of conjecture for now. What we do know is that his decisions are likely to have a huge impact in South Africa and elsewhere, certainly when it comes to trade.
Trump is a fan of tariffs (which he has described as “the most beautiful word”). During his first term, to give just one example, he slapped a 25% import tax on single malt scotch whisky, which ran from October 2019 to March 2021. It was the result of a long-running dispute in the aerospace sector, with Boeing complaining that the EU was subsidising its rival, Airbus. A five-year truce was called in 2021 – and that expires in June 2026. The scotch whisky industry estimates those duties cost it in the region of £600m, more than £1m a day, and understandably it’s keen to avoid a repeat.
“We know that a lot of people are thinking about this and there’s some crisis management and planning going on in companies that are likely to be targeted,” said Duncan Edwards, Chief Executive of British-
American Business trade association, shortly before the election. You can be sure that planning will have gone up a few gears since then.
Eco-ambition
Today’s youth are set to inherit the worst consequences of global warming and the political decisions of recent leadership – but it’s not all doom and gloom.
Directly challenging this narrative of “eco-anxiety” (a 2021 report found three-quarters of people aged 16 to 25 in 10 countries think the future is scary due to climate change), the Villars Institute’s annual Global Issues Survey has also revealed some more optimistic trends. “Eco-ambition” is the key phrase among young people prioritising biodiversity, climate and energy reforms in their careers – according to a 2022 survey by the Jack Petchey Foundation, one in three young people are already interested in pursuing a career in the environmental and sustainability sectors. Meanwhile, a survey from UWE Bristol has found that 72% of young people are willing to change their lifestyles for climate action, compared with 62% of adults, while 82% support shifting away from fossil fuels.
Cool Crones
Where once they were expected to fade into the background and await their status as grandmother, older women are now finding themselves liberated and moving into their power, emerging as a symbol of knowledge, experience and leadership. Yes, the age of the ‘Cool Crone’ is upon us, and we’re just starting to understand the impact of this all-butinvisible social group.
The most recent Forbes 100 Most Powerful Women list was heavily skewed towards the Cool Crone: 80% were over the age of 50; 60-somethings Ursula von der Leyen, Christine Lagarde and Kamala Harris topped the list; Taylor Swift was the sole under-40 in the top 40. Meanwhile, as a recent piece on the ‘Power-Ager’ in the DailyMail pointed out, “76% of 50+ women are confident in their own skin, spending more than ever on fitness, beauty, health supplements and travel.”

FRITSCH SAMPLE PREP WORKSHOP AT LABOTEC
Milling & particle analysing efficiency
Labotec hosted a highly interactive in-person workshop on 14 November, ‘Latest Sample Prep technology for Milling and Particle Size Analysis’, at their Midrand HQ in conjunction with special guests from Fritsch Germany, Maik Paluga and David Oduor from Fritsch Africa. After the opening speech, ‘New dimensions for sample preparation and particle sizing’, which spoke of the theoretical advancements and device-related implementation, the delegates were invited to bring their own samples for processing and analysis using the Fritsch mills, grinders and Analysette Particle Sizer.
A variety of samples from mining, food and industrial industries that included, among others, samples of corn, car tyre, cheese, aluminium powder, bricks, glass sand, butternut, cement, etc, were physically prepared and analysed on the day. According to Maik Paluga, “An eye-opener was analysing the dispersion of an oil-coated metal, which refused to dispense. After using a surfactant (tensile detergent), the measurement worked on the particle sizer.”
He added: “Another highlight was seeing the micrometer measurement difference of three alloy mixes, each with a different compositional twist. We reliably identified the particles at 10, 10.5 and 11 microns respectively."
David Oduor explained: “It was interesting to see the difference in sample grinding time and effects of the milling process, type of mill used, and use of different ball sizes, which all added up to producing the required homogenous fineness needed for the specific application. For instance, we had glass sand from different geographical locations
in SA, and they all yielded different grinding results.”
The interaction, practical demonstrations and discussion of each of the results was of great value to the participants. Some of the more challenging samples for particle sizing were taken by Paluga back to Germany for further inspection by the Fritsch application laboratories.
The objective was to show analysts how to get rapid, reproducible results every time samples are prepared, using the correct mill/ grinder, and to introduce the completely revised ANALYSETTE 22 NeXT Micro and Nano particle sizers with different measuring ranges, 0.5 - 1500μm and 0.01 - 3800μm respectively, that will automatically and simply analyse particle size. The Fritsch range covers size reduction and sample processing, particle sizing and analysis, and sample dividing and feeding and particle size analysis.
A big thank you to all speakers and attendees who made this day a success.

A TRUE MULTIPURPOSE MILL –RETSCH MIXER MILL MM400
This compact benchtop unit is designed for dry, wet and cryogenic grinding of small volumes of up to 2 x 20ml.
It is suitable for classic homogenisation, as well as biological cell disruption of DNA/RNA and protein extraction, and mixes and homogenises powders and suspensions with an unbeatable frequency of 30 Hz within seconds. There is no equivalent to the technology of this mill available on the market. With regard to performance and flexibility, this mill is in a class of its own.
Functional principle
The grinding jars of the mixer mill MM 400 perform radial oscillations in a horizontal position. The inertia of the grinding balls causes them to impact with high energy on the sample
material at the rounded ends of the jars and pulverise it. Also, the movement of the jars combined with the movement of the balls results in the intensive mixing of the sample.
The degree of mixing can be increased even further by using several smaller balls. If several small balls are used (e.g. glass beads) then, for example, biological cells can be disrupted. The large frictional impact effects between the beads ensure effective cell disruption.
RETSCH mixer mills are true allrounders. They homogenise, for example, alloys, animal feed, bones, ceramics, cereals, chemical products, coal, coke, drugs, electronic scrap, glass, grains, hair, minerals, oil seeds, ores, paper, plant materials, plastics, sewage sludge, soils, straw, tablets, textiles, tissue, tobacco, waste samples, wood, wool and more.
The MM400 is the first laboratory mill that can be calibrated, making it suitable for testing labs, accredited labs applying ISO/IEC 17025 or ISO 9000ff, and pharmaceutical products.

MULTI-PARAMETER WATER ANALYSIS IN LESS THAN HALF THE TIME
Save up to 64% of time when performing four titrations in parallel
As the capacity of turntable sample changers is inherently limited, pushing the limit used to mean investing in another autosampler and a titrator, possibly more, depending on the sample volume. Not so with the OMNIS platform. The OMNIS platform includes an x-y-z sample robot that can accommodate up to 175 samples on
a smaller footprint. Furthermore, the OMNIS Sample Robot can be scaled up module by module from small to medium to large. Fully automated parallel titrations at four workstations means getting the same number of samples analysed a lot faster than by sequential analysis. The titrations performed at the four workstations can be

of the same kind or different types. In terms of space-saving, the square design of a modular x-y-z system makes a substantial difference when compared to the traditional turntable design. No matter whether you are interested in determining several parameters or just a few, the automated OMNIS system can be tailored to your needs and requirements. If these change after some time and more detailed ion analysis becomes necessary, then other techniques, e.g. ion chromatography, can be easily added to the existing system setup.

METROHM POTENTIOSTATS
USED TO DEVELOP NEW CATALYSTS FOR GREEN HYDROGEN PRODUCTION
clean and green?
Due to its high gravimetric energy density and zero pollution emission, ‘green hydrogen’ is a clean and sustainable energy carrier that is expected to become one of the fuels of the future. Green hydrogen is produced with renewable energy sources, and it can help to mitigate global warming by using cleaner fuels.
Producing green hydrogen with electrolysis
The most favourable way to produce green hydrogen is via water-splitting electrolysis, where water (H2O) is broken down into its counterparts by using a direct electric current. Electrolysis is a sophisticated technique that has been used for many decades in industry. When using this technique to produce hydrogen, drawbacks are the sluggish reaction kinetics when using inexpensive catalysts, or the high costs for more optimal catalysts (e.g. platinum). To produce hydrogen in an efficient and economical manner, the goal for researchers around the world is to develop catalysts for this purpose that are highly active, inexpensive, and stable over long periods.
Metrohm has a wide range of potentiostats that can be used to characterise recently developed catalysts for electrochemical hydrogen production.

GET A MORE COMPLETE VIEW OF BIOLOGY WITH HiFi READS LIFE SCIENCES/ COMPANY
Africa’s Genomics company, inqaba biotec, has been at the forefront of providing genomics services for the past 22 years to researchers across Africa. In 2017, inqaba biotec partnered with Pacific Biosciences based in California, a leader in long read sequencing, to bring the Single Molecule Real Time sequencing (SMRT) technology to the African market. SMRT sequencing involves a single strand DNA molecule bound to DNA polymerase, forming a DNApolymerase complex that is sequenced in SMRT cell containing millions of nano wells, known as Zero Mode Waveguides.
What are HiFi reads?
SMRT sequencing technology has evolved to a different type of long reads, known as highly accurate long reads or HiFi reads. HiFi reads are produced using a circular consensus sequencing mode (CCS) on PacBio systems (Sequel IIe, Revio and Vega) and provide base-level resolution with 99.9% single-molecule accuracy, allowing researchers to accurately detect all types of variants, from single nucleotide to structural variants, with high precision and recall and phase haplotypes, even in hard-to-sequence regions of the genome.
Benefits of HiFi reads:
• Long and highly accurate reads: HiFi reads provide the information needed to confidently call and detect all variants with superior accuracy (>99.999%) with long reads spanning thousands of bases without compromising read integrity.
• Uniform coverage across the molecule: Unlike other methods, HiFi sequencing does not show bias based on GC content and other genome complex regions, making it possible to sequence challenging genomic regions.
Single-molecule resolution: The technology captures sequence data directly from natural DNA molecules, ensuring the generation of highly accurate long reads with stunning single-molecule precision.
Application of HiFi reads:
• Comprehensive variant calling with phasing + 5mC and 6mA: Access high accuracy for all variant types - SNVs, indels, structural variants, tandem repeat expansions, and methylation -
including in challenging regions.
• A truly complete assembly of complex genomes: Accurately construct the full sequence of chromosomes, including telomeres and centromeres.
• Targeted sequencing to study genes of interest: Focus the power of HiFi variant calling by enriching for regions of interest using hybrid capture or PCR amplification.
RNA sequencing with Kinnex:
The Kinnex kits are based on the MAS-Seq method, which concatenates smaller amplicons into larger fragment libraries for throughput increase.
Kinnex kits increase throughput for fulllength RNA, single-cell RNA, and 16s rRNA sequencing on PacBio long-read sequencing systems to enable large-scale studies at a resolution difficult to attain with short-read RNA sequencing.
