T

he race to space has begun with various companies vying for first place. Each company wants the opportunity to be the first to launch a tourist space shuttle into space. With successful Amazon owner Jeff Bezos recently revealing the latest innovation from his company Blue Origins, namely the New Glenn space rocket, the competition for colonising the planets beyond the earth is slowly heating up.

In September, billionaire Bezos divulged the name and design of Blue Origin’s latest space craft. New Glenn, named after John Glenn who was one of the first American astronauts to orbit around the Earth, is a rocket designed with the ability to transport large quantities of people into orbit around the Earth in one trip.The New Glenn rocket is predicted to be one of the largest space vessels yet to be built.

According to the plans, there will be two different versions of the new rocket. The two- stage version of the New Glenn will have the ability to travel to low-Earth orbit (LEO), while the three-stage version of the rocket should be able to travel beyond LEO, which has not been done before. Each of CRS-6 Falcon 9 launch

the versions will be an estimated 7m in diameter with seven BE-4 engines and will lift off with 38.5 million pounds of thrust.

In an effort to make space flight more affordable and efficient, this new rocket also features reusable parts. In his announcement Bezos explained that the first stage of the vessel will have the ability to land post-launch. The ability to land a rocket after launching it into orbit around the Earth is a feature that has not been seen in any other rocket so far and it puts New Glenn leagues ahead in the race to space.

It will also make space travel more affordable in the long run.

However, while the details of the space rocket have been revealed, construction of this game-changing technology has yet to begin. According to Bezos, Blue Origins hopes to launch New Glenn from Launch 36 Complex 36 at Cape Canaveral in Florida (USA) before the end of the decade.

Other contenders Bezos’ Blue Horizon is not the only contender in the race to start offering tourism trips to space. In fact there are already several other companies that are fighting for this title.

Business magnate Elon Musk’s company SpaceX is in close contention for the number one spot in space. The latest offering from this company is the Falcon 9 space rocket. The Falcon 9 was designed as a two-stage rocket that could safely deliver satelites and the Dragon Spacecraft into orbit around the earth. In 2012, this spaceship safely transported the Dragon spacecraft into well-timed orbit so that it could rendevous with the International Space Station (ISS) and as a result of this successful mission, SpaceX went down in history as the first commercial company to visit the ISS. Since then the company has sent multiple flights to and from the space station to transport cargo back and forth.

SpaceX is currently in the process of building the Falcon Heavy, which is scheduled to launch later this year. This rocket will be extremely powerful with the ability to lift the equivalent of a 737 jetliner loaded with passengers, crew, luggage and fuel (over 54 metric tons) into orbit. However, it ranks second when compared to the payload of the New Glenn. Nevertheless, the launch of this rocket will see another rocket being created with the purpose of carrying people into space.

United Alliance The third competitor in the race to space is the company United Launch Alliance (ULA), which is a joint venture between Lockheed Martin and The Boeing Company.

ULA’s Vulcan is a rocket that has been under construction for the past two years and will only be ready for launch in 2019 but already has high expectations for future trips into space.

Although these vehicles are still in there early stages of testing, the success that the commercial space flight industry has seen in the past few years shows potential for the future of space travel. Who knows, in the not-so distant future, space travel might be as simple as logging onto the website and booking a seat onboard your preferred spaceline.

Recently, I was faced with a very difficult decision, either stay where I am and continue in my comfort zone or take a huge risk and see where it takes me. While I was wracking my brain trying to decide what to do, a famous quote by TS Eliot came to mind.

“Only those who will risk going too far can possibly find out how far it is possible to go,” and he couldn’t have been more right.

Take Amazon founder Jeff Bezos for example. He is an entrepreneur that has received great success because he was never afraid to take a risk. In 1994, despite being the youngest senior vice president at a successful investment company, Bezos decided to take the risk, quit his job and start a virtual bookstore. His blind leap of faith back then led to the creation of one of the most successful companies in the world, Amazon.

And Bezos is once again pushing the envelope with his plans for the New Glenn, which could catapult him directly into space.

So, why not take a page out of this successful entrepreneur’s book and take a chance because the real risk in life is to do nothing.

Part 2

Planning appropriate PT activity – dispelling the myths EP Tarnow, NMISA

I

n Part 1 of the article SANAS Technical Assessor and metrology consultant EP Tarnow explained some of the steps that are necessary in planning the appropriate Proficiency Testing (PT) programme for your specific laboratory.

Part 2 of this article continues to explore the considerations when planning suitable PT activity.

2.6 Identification of suitable participants The selection of suitable participants depends on several factors, not least of which is the PT activity Target Uncertainty of Measurement.

The following needs to be considered for PT schemes where the Reference Value is either obtained as a calibrated value or as the manufacturer’s accuracy specification as verified through calibration: All the participants must have a larger PT Activity Target Uncertainty of Measurement than that associated with the Reference Value.

The participants must be able to perform the measurements at or very close to the calibrated Reference Values (measurement points).

There should preferably be a minimum of two participants in addition to the Reference Calibration Laboratory.

The following needs to be considered for PT schemes where the Reference Value is obtained statistically from all the participants’ results: All the participants should have similar measurement uncertainties unless a weighted mean is used to determine the Reference Value.

All participants must have the technical capability to perform the measurements.

All participants should preferably use the same or similar methods. This is to reduce the impact of any bias.

2.7 The means of analysing the results and measuring performance There are several statistical

methods for the analysis of the results and measuring performance.

The most common one used by calibration laboratories in industry is: En =

(x – X)

√(U2LAB + U2REF)

Where: En is the Normalised Error (commonly known as the En value) x is the value reported by the participant X is the Reference Value ULAB is the measurement uncertainty reported by the participant UREF is the measurement uncertainty associated with the Reference Value In this case, satisfactory performance is demonstrated when a Normalised Error of between -1 and +1 is achieved by the participant.

2.8 Identification/ selection of a pilot laboratory if appropriate The pilot laboratory is not necessarily the source of the Reference Value. It is the laboratory tasked with the responsibility of planning, executing and reporting the results of PT activity. This responsibility must be assigned before the start of the PT activity and the pilot laboratory must have the necessary competencies to be able to take on this responsibility.

2.9 Drawing up a protocol The protocol sometimes referred to as the audit instructions is the document that contains all the information relating to the PT activity and is generated before the PT activity is implemented. This is the document that is usually circulated to all potential participants to give them an opportunity to evaluate whether they are technically capable of participating and achieving their objective of PT participation.

The protocol must contain at least the following minimum information: The identity of the pilot laboratory Identification of the PT activity participants The proposed measurement artefact or instrument The measurement parameters, including the range and measurement points Any special artefact storage or handling considerations Instructions on the conditioning of the artefact prior to performing the measurements if required Appropriate instructions on how to use the artefact How the Reference Value is defined Specific and detailed instructions on the manner of recording and reporting the measurement results and their associated measurement uncertainties How performance will be evaluated.

All PT activity must produce results that must be formally reported.

The report must contain at least the following minimum information and be circulated to all the participants upon conclusion of the PT activity: Identification of the participants The measurement protocol Identification of the measurement standard or artefact The detailed measurement results of all the participants with the confidentiality of the participants protected The Reference Values and Reference Uncertainties and how these were determined The evaluation of the measurement results An indication of the evaluated performance of each participant- Minimum acceptance criteria Conclusions and recommendations.

A conclusion needs to be made at the end of the PT activity as to whether or not the objectives set out at the beginning were in fact achieved.

These should include all aspects of the PT activity such as participation, resulting trends and performance of the artefact.

These conclusions should then be used to improve the planning of similar future PT activity.

2.12 Appropriate proposed recommendations Based on the conclusions made, certain proposed recommendations can and should be made to guide future improvement.

This could even relate to the performance of an individual participant as long as their identity is kept confidential.

In particular, result trends should be interpreted and recommendations made to explain the behaviour and how to correct it.

Participants in PT activity have an obligation to respond appropriately to the results they obtain.

Where results are deemed unsatisfactory, root cause analysis must be conducted and appropriate corrective actions implemented to rectify the problem.

It should be noted that part of the root cause analysis investigation must include the investigation into the potential impact erroneous results on certificates and reports may have had for customers.

Customers should be immediately informed of any potential impact and the calibrations or tests repeated where possible to provide technically valid results.

2.14 The PT Activity Plan All SANAS accredited laboratories are required to

have a 5-year PT activity plan.

This plan must address at least the following: Identification of planned PT activity Identification of participants or potential participants in the proposed PT activity The proposed measurement artefact/sample or instrument The measurement parameters, including range and measurement points How the Reference Value is to be established The method of evaluating performance and minimum acceptance criteria The identification of the pilot laboratory who has the responsibility of generating and distributing the report Where applicable, the link between the planned PT activity and the scope of accreditation – with specific focus on challenging measurements such as at the range extremities.

The PT Activity plan must be a controlled document with the historical as well future plans.

It must thus be possible for a SANAS Technical Assessor to compare what was planned to what has been executed over a specified time period, and to what degree the PT activity has satisfactorily covered the entire laboratory scope of accreditation within a 5-year period.

3. Avoiding common pitfalls/dispelling the myths Based on the author’s experiences, the most common problems relating to PT activity in the South African calibration laboratory community are:-

Unsatisfactory PT activity plans: Inadequate detail Plans do not adequately cover entire scope of accreditation Plans do not continuously cover a ‘rolling window’ of PT activity.

This makes it difficult for an assessor to adequately determine what was planned versus what has taken place and whether or not this adequately covers the laboratory’s scope of accreditation at least once every 5 years.

Total lack of documented protocols or protocols containing insufficient detail.

This often results in the different participants performing the measurements at different points and using different methods, than were originally planned, making direct comparison difficult. Furthermore, if the artefact setup is not carefully defined in detail, it is possible that the results will differ. For example the selection of the same filter settings, or the same sampling rates.

Poor selection of artefact/ sample or instrument, Often the artefact has not been adequately characterised before the PT activity to determine its stability and performance after having travelled between participants.

In addition, the artefact selected, is often not capable of performing in line with the desired Target Uncertainty of Measurement.

Unclear definition of Reference value: In many cases, no Reference Value is defined or determined. Usually the

participants just test to see whether an En value of between -1 and +1 has been obtained.

However, often this result indicates nothing more than the participants are equally wrong.

The Reference Value must be defined to ensure there is a link to the True Value.

Unclear definition of PT activity Target Uncertainty of Measurement: There is very seldom a Target Uncertainty of Measurement defined at the start of the PT activity. This often results in two participants having vastly different reported measurement uncertainties in which case there is no benefit to the participant with the smaller uncertainty.

Furthermore, the common objective of demonstrating proficiency in being able to perform measurements to within an accredited CMC is often not realised.

Poor determination of PT activity Reference Value Uncertainty: It would appear that there is a general lack of understanding regarding the measurement uncertainty associated with PT activity. This is especially important when participants are aiming to demonstrate proficiency in being competent to within their accredited CMCs. A common mistake is to assume that if two participants participate in PT activity, both having reported measurement uncertainties at their CMCs, obtaining a Normalised Error value of ≤ |1| is an indication that they have both demonstrated proficiency to within their CMCs. This is not the case since the Reference Value Uncertainty is

actually the Root Sum of their CMCs squared which is larger than either CMC Bilateral comparison PT activity with incompatible Measurement Uncertainties: It is common to find results of PT activity where the participants have vastly different measurement uncertainties. While this may benefit the participant with the larger uncertainty, it does not demonstrate the proficiency of the participant with the smaller uncertainty.

Refer to Fig 4 below. In this case, participants 7gv2 and 5ht9 have not demonstrated ‘proficiency’ to within their reported measurement uncertainties since the Reference Uncertainty as indicated by ‘NLA 1’ and ‘NLA 2’ is significantly larger.

Lack of formal report.

In many cases the Technical Assessor is presented with an informal report recording PT activity, which is inadequate in that insufficient information is contained therein and the minimum SANAS requirements have not been met.

4. Summary In summary, PT activity has to be a planned activity with a specific objective. While it is obligatory to meet specific requirements if a laboratory is accredited by SANAS, PT activity should be enthusiastically embraced to manage the risk of a laboratory reporting technically invalid results. The cost of participation therefore, so often used as the excuse for inadequate participation, should perhaps be balanced against the potential legal and other costs, which would be incurred if the impact of a laboratory issuing erroneous results was severe.

Debunking the 5-second rule

O

ne of the most well-known and widely accepted notions is the 5-second rule, which states that food that has been dropped on the floor can still be consumed if it is picked up within 5 minutes of it falling on the floor. However, a group of scientists from Rutgers University in the US recently disproved this notion. Their research was published in Applied and Environmental Microbiology.

The 5-second rule is a belief that is accepted around the world by individuals of different ages, however new research on the topic renders this idea invalid. Previously scientists thought that bacteria required longer than 5 minutes to transfer from one surface to another, however research from the Rutgers team have revealed that this is not always the case. The type of surface, contact time as well as moisture all plays a role in

how bacteria spread from one area to another.

In their study, the researchers looked at how long it took for cross contamination of Enterobacter aerogenes, a nonpathogenic relative of Salmonella on four different surfaces, including wood, stainless steel, ceramic tile and carpet. They also used four different types of food (watermelon, bread, bread and butter, and gummy candy) and tested four different contact times namely less than 1s, 5s, 30s and 300s. The researchers set up different scenarios with the various foods and surfaces to test how long it would take bacteria to be transferred from the different surfaces to the food.

Their study revealed that watermelon appeared to have the most contamination with gummy candy having the least. The results also showed that carpet had a

much lower bacteria transfer rate as compared to tiles and stainless steel. Their research also revealed that while longer contact times result in greater transfer of the bacteria from one surface to another, contamination can also occur immediately.

Therefore, while the 5-second rule is not necessarily incorrect, it can’t always be applied to every

situation. These researchers have shown that there are a number of factors that play a role in crosscontamination of bacteria between surfaces and food. Therefore, the next time you drop food on the floor, it might be better not to pick it up and pop it back into your mouth, because you never know what might have had the chance to contaminate it.

First DNA sequenced in space

N

ASA astronaut Kate Rubins has become the first ever person to successfully sequence DNA in space. Kate was aboard the International Space Station (ISS) when she carried out this procedure using the Biomolecule Sequencer. The success of her DNA sequencing in microgravity could have a large impact on future missions to space.

Virologist turned astronaut Rubin has made history by being the first ever person to sequence DNA in the microgravity environment of space. In her experiments Rubin successfully sequenced three different types of DNA, namely mouse, virus and bacteria DNA onboard the ISS. While the DNA samples were all prepared in a laboratory on the ground and sent up with Rubin, the actual sequencing process

of the DNA took place onboard the spacecraft.

In order to ensure that the environment was the only variable factor in the study, they also had a group of researchers perform the same sequencing technique on the same types of DNA in a laboratory on the ground.

How DNA sequencing works?

Each DNA strand within the body is made up of a specific order of nucleotides (adenine, guanine, cytosine and thymine).

DNA sequencing is a method that involves the determining of the specific order of these nucleotdes, which can in turn help to provide important genetic information. For the DNA sequencing of the three

samples, Rubin used a device called MinION. This works by sending a positive current through nanopores embedded in membranes inside the device.

At the same time, fluid containing a DNA sample passes through the device. Individual DNA molecules then partially block the nanopores and change the current in a way that is unique to that particular DNA sequence.

By looking at these changes, researchers can then identify the specific DNA sequence of the sample.

While the methods of DNA sequencing have become quite advanced over the years, one of the major challenges that this process poses in space is overcoming the effects of microgravity. For example on Earth air bubbles always float

to the top of a substance and can be removed. However, in an environment with microgravity, air bubbles are less predictable and could end up blocking the nanopores of the device.

However, Rubin successfully overcame these microgravityrelated problems to be the first person to sequence DNA while orbiting the Earth.

Having the ability to sequence DNA in space could have a myriad of benefits for future astronauts including helping them to have better understanding of the different environments they find themselves in. It could also help them identify illnesses or other organisms, which can help them to stay healthy during extended trips to space. The possibilities are infinite.

Speeding up Salmonella testing

S

cientists from the University of Florida Institute of Food and Agricultural Sciences have made a scientific breakthrough by creating a faster method for

detecting Salmonella in food.

Speeding up this food testing process could play a huge role in the prevention of illness.

Salmonella is a bacterium found in food that is responsible for causing food poisoning, which can have some serious consequences. Therefore, it is essential to test food prior to it being made available for consumption. This new test could reduce the testing time for

Salmonella from 2-3 days to just 15 hours.

In their study, the group of researchers artificially contaminated a number of samples of beef and chicken with Salmonella. The scientists then tested each of these samples using a combination of a Salmonella-specific antibody and a unique signal amplification method. Their unique blend produced results in just 15

hours. Their research has since been published in the Journal of Food Safety.

According to the World Health Organisation, globallly 1 in 10 people fall ill every year from eating contaminated food.

Therefore, while this test is still in its early stages, it has the potential to make monitoring pathogens more efficient, which could greatly improve health in the long run.

t can be a fine line between success and failure in the laboratory. Some experiments can be more sensitive than others and often when things go wrong, you don’t realise until perhaps it’s too late. Sometimes it can be one failed attempt too many that sends you over the edge, leaving you facing many hours of troubleshooting and repeated experiments long after everyone has left for that conference in Pape’ete, the one where you were hoping to present your data to your peers. Don’t worry, it happens to the best of us but it helps if you can see it coming and take steps to ensure it doesn’t happen in the first place (or ever again with any luck).

Water is an important solvent for almost all procedures in a laboratory. In fact, I can’t think of one where water isn’t involved at some point or other, although granted some most definitely rely on it more than others.

Water purity is fundamental if you are going to succeed. Some of the more merciful techniques in the laboratory can tolerate a bit of a hit in the water quality department and a dash of human error but the more sensitive ones require the finer things in life.

Yes, champagne, caviar and diamond-studded pipettes would be a nice touch to your daily laboratory routine but we are talking about ultra-pure Type I+ water of course.

A new test for asthma

T

he World Health Organisation estimates that there are 235 million around the world currently living with asthma. However, despite being so prevalent globally asthma is still one of the most under-diagnosed medical conditions. A new diagnostic test created by a group of researchers

at Loughborough University (UK) could help to change this by creating an easier and more reliable method to detect asthma.

The research was recently published in the journal Analytical Methods.

Despite effective treatment being available for asthma, this condition kills thousands of people every year.

Many people succumb to this illness as a result of not being diagnosed with the illness and thus, not receiving necessary treatment for the condition.

However, with this new asthma test, diagnosis of the condition could be as simple as collecting a sample of the patients saliva.

In their test, the Loughborough University researchers in collaboration with the Nottingham City Hospital, collected different saliva samples from a variety of people, some of whom had asthma and others that didn’t.

They then peformed liquid chromatography-mass

spectometry on the samples to detect the metabolic biomarkers in the saliva. By measuring the amount of metabolic biomarkers in the sample, the scientists could determine whether a patient had asthma as well as how severe the condition was.

While diagnostic tests for asthma exist, including the lung function test that measures an individuals airflow into the lungs, many of these tests are stressful and inaccurate. Therefore, this new diagnostic method for asthma that requires just a small amount of saliva could revolutionise how we test for the condition on a gobal scale, which in turn could affect how many people are treated for asthma.

While this certainly is one step closer to creating an easier and more accurate method of testing for asthma, further research still needs to be done before this diagnostic method can be introduced at medical facilities.

However, we can breathe easier knowing that researchers are trying to develop better and more efficient ways to test for life-threatening medical conditions like asthma.
