Director Comments

I

want to welcome you all to the Land of Lincoln, Illinois. Come join other dairy enthusiasts, as it will be a joint convention with Ayrshire and Brown Swiss. I commend the states that had spring sales this year. They did outstanding jobs with fabulous results. The quality has been exceptional, and the prices reflect such. We will have the best North America has to offer at the Grand National Convention Sale, so come and find something to improve your genetics. We have two tours scheduled for convention - one to show you the sights of Peoria and another to see the latest in innovation and technology in the dairy industry at River Valley Farm, one of the top Jersey herds around. Come for the comradery; and most of all, come to your national meeting for this is where your Association’s future is molded. Truly, as Ayrshire breeders, it is your future being developed. It only makes sense that attending the annual meeting should be high on all of our priority lists. Thanks to all who have made this convention possible. See you in Illinois! ~Steve McDonald

GENOMICS By: David J Kendall, Director of Genetic Development, Taurus Service, Inc, Mehoopany, PA

T

he April 2013 U.S. Genetic Evaluations introduced the power of genomic evaluation to the tool chest of Ayrshire breed improvement. Last summer, Adam Oswalt gave an excellent introduction to genomics in the Digest. This article is part of the continuing education process to help you better understand and utilize this technology. As Adam wrote, genomics allows us to see pieces of the genetic inheritance that the parents, grandparents and further removed ancestors have transmitted to an animal. These pieces of genetic material are denoted by Single Nucleotide Polymorphisms or SNPs. An SNP is a change in the sequence of the amino acids that are the building blocks of DNA. It is important to know that these SNPs, in the vast majority of cases, do not directly represent a gene. Rather, through extensive statistical analysis, SNPs allow us to correlate these changes in the amino acids with changes in animal performance. Most of these changes are very small; they have to be added up across not only on an individual chromosome, but across all 30 chromosomes as well, to get a true understanding of the total potential that an animal has to both perform in its own right as well as to pass on to any offspring. Dr. John Cole at the USDA has developed a fantastic tool to help visualize how these process works. If you go the Council on Dairy Cattle Breeding website, go to the left hand column under General Inquiries and click on “Get Chromosomal PTA by ID” you will get this:

If you type in the registration number of an animal that has been genomically tested, such as AY50803818 for Hidden Valley Garth’s Brandon, you will get a graph as shown below. What this graph shows us is the total contribution from each parent on each chromosome for each trait that is currently measured. This graph shows Brandon’s Chromosomal PTA for Net Merit. For instance on Chromosome 1 Brandon has inherited, as measured by the SNPs, +6.352 NM$ of value from his sire (always coded in Lavender) and -11.96 NM$ from his dam (always in peach) for a net contribution of -5.608 NM$ on this chromosome. This is repeated for each chromosome, added together and then regressed to give the Direct Genomic Value (DGV).

DGVs are the important first step of genomic evaluations. DGVs are based on comparison with the Reference Population, all the information from all the Ayrshire animals that have been tested correlated with each trait that is evaluated, such as NM$. This step does not

include any pedigree information. The second step is the inclusion of the polygenic effect. The polygenic effect is the first place where information from an animal’s traditional pedigree is incorporated. The polygenic effect is included in recognition that not all information about inheritance of genetic material is directly measured by the SNPs. SNPs measure the highlights, and they do a very good job of capturing the majority of the information, but is it not all the information. The third piece is contribution of non-genotyped ancestors, in Brandon’s case, his dam. This piece is complicated and important. Genomics does see the SNP information that a nongenotyped parent contributes, and it does measure that contribution against the reference population. The complication is that without the direct contribution of the SNPs from that animal combined with that animal’s performance it is not as accurate. This step recognizes this fact. This is where the increase in reliability will come from as more animals are tested. The result for Brandon is that of his +122NM$, +136NM$ comes from his sire as compared to the reference population, -45NM$ comes from his dam compared to the reference population for a DGV contribution of +90NM$. The 46 NM$ difference between Brandon’s DGV and official NM$ comes from inclusion of the polygenic effect and the performance of his dam and other relatives that have not been genomically tested. I hope this article helps you to better understand the process and the value of genomics. There is much more that we can learn from and apply to improving Ayrshires from this great tool. In future articles, we will look at some of those opportunities.

Breeders RoundTable on Genomics As genomic testing has become more readily available and breeders have embraced this new scientific availability, we asked four breeders to respond to a series of questions regarding their thoughts and use of genomics. Thanks to Dandy’s Cherry Knoll, Dan Carpenter, of Dansville, Michigan; BarVue Farms, John Irrthum of Wanamingo, Minnesota; Grand-View Ayrshires, Tim Busch of Cecil, Wisconsin, and Palmyra Farm, the Shank & Creek Families of Hagerstown, Maryland, who all provided input to this roundtable. How are you using genomics in your herd? Bar-Vue Farms: We are using genomics to

6 Ayrshire Digest

compare full brothers that will be going to AI. Dandy’s Cherry Knoll: To identify the best of my herd bulls. Grand-View: We just started testing a couple of females and males from our best cow families. Palmyra: We have Ayrshires and Holsteins and currently use the genomics very differently for each breed. For both, we consider genomics a tool to be used to help make breeding and marketing decisions. The Ayrshire genomics are still in a very early stage, therefore, are not highly reliable. We are actually at a point where Holsteins were six to seven years ago. Primarily we are sampling animals to add more data to the research pool. We are sampling full siblings to compare their information amongst themselves and to their parental averages, trying to see if a particular line or animal is transmitting more consistently than another. But at this time, the genomic results are limited, and we rarely make decisions based on their values. For the Holsteins we are always reviewing and updating our mating list. We list the bulls we are considering using based on the goals of our herd production and type and then we may narrow that list down by using their genomic values. How long have you been genomic testing? Bar-Vue Farms: One year. Dandy’s Cherry Knoll: Just started this year. Grand-View: We have been doing genomics on males for about nine months now through bull studs and just started in February on our own, with males and females. Palmyra: We began testing Holsteins six years ago, soon after it became commercially available. We have been working with AI companies for over two years submitting samples to help increase the data basis for the Ayrshires. We began personally sampling Ayrshires in October 2013. Do you use genomics differently today than you did when you first started? Bar-Vue Farms: No. Dandy’s Cherry Knoll: No Grand-View: Genomics are still being used the same. Still waiting for more results to guide us into what direction we should go next. Palmyra: We have not changed our use of Ayrshire genomic data. At this time, because it is still so unreliable, we just study the results and wait to see if there are any trends within family lines. As the data increases, and it becomes more reliable, we will consider it more when making May/June 2014

breeding and marketing decisions, but it will never be our only consideration. For the Holsteins, we now put more emphasis on the genomics than we did six years ago since the reliability has greatly increased. But when we started out with the Holsteins ,we used them in much the same way we are using the Ayrshire results. However, it is still only part of our decision making process. What advantages does genomic testing provide for you? Bar-View Farms: It lets us determine the differences in the full brothers. Dandy’s Cherry Knoll: I am intrigued by the polled trait, and am working toward a polled herd with good type and production averages. Since polled breeding stock is very difficult to find—with any choice of polled sires being virtually impossible— genomic testing allows me to identify the better of my own polled bulls for future use. It should also allow me to quantify improvement from generation to generation. Grand-View: Genomics gives us just another tool to use to find what offspring are the best at a young age. Palmyra: Genomic testing gives us more insight into the genetic potential of individuals at a younger age. It can also increase the marketing value of our animals and herd. In the future, as it becomes more reliable, it can greatly add to the economic value of our animals.

breeders to identify and use more unrelated quality bulls, reducing inbreeding in the breed overall. Grand-View: Hopefully with more people using genomics we can find some different cow families with some outcross genetics. Palmyra: At this time Ayrshire breeders will not see a greatly added value to their animals as we see in some of the other breeds, but this can change as the data becomes more reliable. It will have the potential to identify which animal might transmit superior genetic traits more consistently or at a higher level and recognize this at an earlier age. An example is we recently received results on six full siblings. The evaluation showed that two of them should be far superior to the other siblings, and they were also much higher than you would expect from their parent averages. Based on this (if it were more reliable) we should use these two for AI or flushing before the other four are considered. Are there any disadvantages in your opinion about genomics and what are they? Dandy’s Cherry Knoll: 1) At least with bulls, the cost discourages its use. The individual test price may not be that high, but you’d better not let anybody else use him, or the costs jump significantly. 2)

Cont. on pg. 13

What advantages does genomic testing provide for Ayrshire breeders? Dandy’s Cherry Knoll: I recently did an unscientific survey of the newer Ayrshire bulls offered by commercial AI companies, and found a disturbingly large percentage to be the sons or grandsons of one or two particularly well-known bulls. While I have nothing against any of these sires, that emphasis on a very small number of sires cannot, in the long run, be good for the breed. Genomic testing could (if utilized) allow more 7

Samantha Ann Rathjen is the 18-year-old daughter of Bridget and Josh Geigle of Walcott, Iowa. She is currently a senior in high school with plans to attend a local college after graduation and pursue a farm management and agriculture business program. She is currently active in 4-H, FFA, has participated in school softball and local leagues, volunteered with the Muscatine Special Olympics teams, is active in the community and church activites and works at the local vet clinic. She has been showing registered Ayrshires at the local and state levels. She is currently the Iowa Ayrshire Princess and plans to run for the National Ayrshire Princess. She owns twenty-five Ayrshires and has been a junior member of the ABA for 11 years. “My ultimate goal after graduating college is to apply my degree and experience into the development of a small farm operation that is geared toward people with special needs. This type of farm will help these individuals develop life skills while participating in a farm

setting. They will be able to interact with animals on a day to day basis. There have been many studies conducted that show the positive influence of animals and people with special needs. The most notable to me is Dr. Temple Grandin at Colorado Sate University. She was diagnosed with autism at a young age, but she worked on a ranch. She went to college, and now does extensive work on the design of livestock handling facilities. I feel if I develop such a farm, using my educationa and farm life experience, I may be able to help the next Dr. Temple Grandin.” Genomics cont. from pg. 7

Animal breeding has, and will probably always be, an art as well as a science. I’m afraid that too much emphasis on the genomic numbers may not be as successful as we may be led to believe. ,

MISSOURI

Grand-View: The reliabilities for Ayrshire genomics are very low right now....... there needs to be some type of an incentive for breeders to use it. Palmyra: Besides the low reliability level for Ayrshire genomics, our biggest concern is when individuals base mating, marketing and purchase decisions almost exclusively on genomic values. Even in the Holsteins, at this time, genomics are still a tool to help make decisions and should not be relied upon singularly. We are also concerned that progress toward more reliable information for the Ayrshire breed can be slowed because of the diverse bloodlines found throughout the population. Any other thoughts on the subject of genomics? Bar-Vue Farms: Yes, I think that it will narrow the genetic base even more. All the

AYRSHIRE ASSOCIATION

studs will pass over an older cow who has proven herself and possibly an out cross as a bull mother. Dandy’s Cherry Knoll: Although the science of genomics has come a long way in the last 10 years or so, I would be surprised if scientists had discovered every gene that should stay and every gene that should be eliminated. Sometimes I worry that one day we’ll turn around and realize that in our rush to genomically quantify dairy cattle we’ve inadvertently lost desirable alleles, or build up undesirable traits. Palmyra: Genomics will continue to evolve and become a very useful tool, but at this time it needs to be used as a supporting aspect of your decision making process as you develop your breeding program.

I’m running for director of Region 3. I would really appreciate your vote!! Please support the Ayrshire youth calf raffle fundraiser! Send all tickets and money to me...please...not the ABA.

OCTOBER 18, 2014 NE Ayrshire Club Sale

What’s the purpose of Ayrshire clubs? Our industry has changed and will continue to change. Shouldn’t our state and regional clubs continue to evolve? What’s the mission, the need, the effectiveness? The creation of the Ayrshire Foundation and the upcoming U.S. hosted World Ayrshire Federation Conference make state clubs vital. Let’s have some meaningful dialouge about this.

The Ayrshire Future QUEEN’S Corner

Happy Spring!!! Finally, all the snow is melting, syrup is boiling, and the t-shirts are emerging! As this spring comes around, I find myself scrambling for finals and finishing off scholarship applications. Some things I am sure my fellow college students can easily relate to. As my reign as your National Queen is quickly coming to a close, I have been squeezing in some time to reflect on this past year. As an individual, I have had the chance to grow a lot (or at least I feel I have). It’s crazy to think that, at this time last year I was filling out my application and preparing myself for what competing would be like. To some people, being Queen just may be wearing a crown and looking pretty in the pictures. For me, the Ayrshire breed is partially responsible for where I am. Without the ever amazing breed I would never have gotten involved with dairy cattle, and I never would have considered studying in agriculture. Being National Queen has opened many doors for

me. I have been able to meet so many people, and gotten to know people I have met even better. It has given me a chance to network within the breed, which is a very valuable resource! I really see the breed as more of a family now, like a giant connected web. One of the benefits of being able to wear the crown as it does draw attention to you. By doing that, you are able to meet even more people than if you were just walking around. And who wouldn’t love to see the smile and awe on little girls faces when they see the sparkle of the crown and ask you questions with such admiration. It really makes you feel special, and I hope that I can convince those little girls that someday they can wear the same tiara, because once upon a time, I was just like them. I still remember the night at my first Ayrshire convention when I stayed awake on my mom’s lap just long enough to see who was crowned the princess. It has been amazing. I never would have thought that my reign would have been like this. I always thought the personal

Meet Your National Ayrshire Youth Board Xavier Altmeter, Director-at-Large Xavier Altmeter, 19, hails from Arcade, New York, and is our National Youth Board Director-atLarge. Xavier is currently a freshman at the State University of New York at Cobleskill, where he is a major in agricultural engineering with a minor in dairy science. The Altmeter’s milk their 85 head of various breeds in a tie-stall barn in Wyoming County in Western New York. “Even though both my parents grew up on farms, we are a first generation dairy farm which in today’s economics, has had some challenging times,” said Altmeter. Xavier’s family attempts to grow all of their own forages, however, competing land issues with large dairies makes this difficult. The herd receives a TMR consisting of corn silage; haylage; dry hay and a purchased grain supplement. During the warmer months the herd is grazed. Xavier owns 28 head of Ayrshires of which 18 are cows and 10 young stock. He owns one cow who is classified Excellent. Xavier is very involved on his family farm as the work force is made up of his family who includes his parents, Cory and MaryAnn, and brother Zac. “Zac shows one of the other breeds we own and has done equally well

- Diana Curtis, youth committee

with his project” said Altmeter. Xavier’s responsibilities include the daily tasks of running the farm when he’s not at college. His favorite task is mixing the TMR ration for the various groups. “I like the responsibility of making the right mix for the milking herd, versus the dry cows or heifers. Xavier also has learned how to breed the cows artificially and he “feels accomplished when pregnancies are confirmed to matings that I feel will improve the next generation.” Xavier and his family show at various shows. He has showed at the Wyoming County Fair as a 4-H member for 10 years where he has been awarded various championships. His goal is to win Supreme Champion. Xavier also has showed at the New York State Fair for several years. Xavier explained that his busy schedule has limited his show schedule. He has attended the Royal, which he truly enjoyed, and hopes to attend World Dairy Expo in the near future. Xavier has been a very active member of both the New York Junior Ayrshire Club and the Junior Ayrshire Breeders’ Association. He has attended four national conventions, with his favorite being hosted by Maryland, where he had the opportunity to meet breeders from all over the world. On a national level, Xavier has participated in the photography contest, folding display contest, dairy bowl and dairy jeopardy. He also has been a participant in

interactions I would have would be very formal and professional. But I have been able to really get to know some of the breeders, and hear all about their own passions. From pretty cows, to special cows, or ones who milk like crazy, everyone has their preferences, and everyone’s story is truly unique. I can honestly say this year has made me so proud to be a part of the breed, and as my months tick away I am excited for convention but sad to put my crown and sash away. Hopefully, I will be able to give back to the breed as much as they have given to me. Have a lovely spring, and I hope the sun is shining wherever you are! ~Ashlie the National Youth Book Contest and was the winner in the senior division, which led him to being named 2013 National Outstanding Ayrshire Youth. Xavier has been a member of the New York Spring Sale work crew and an active member during the state annual meetings; fund raising and breed promotion at the New York State Fair. He promotes the Ayrshire on all levels and at a variety of agricultural related events and helps younger 4-H members with their projects. Xavier was a very active member of the Pioneer Chapter of FFA where he competed at all levels of competition. He is a Empire Degree recipient; proficiency award winner for ag mechanics and was the Ayrshire dairy breed winner. He also placed second in the dairy proficiency. Xavier competed in agricultural business competition at the national level and received the silver award. As a 10 year member of Wyoming County 4-H, Xavier has participated in all the 4-H dairy events but states showing his Ayrshires has been his favorite. Altmeter would like to work for an agricultural firm on the corporate level designing new prototypes of equipment and eventually have his own Ayrshire dairy farm. On his farm, Xavier would focus on quality over quantity with an emphasis on components and production.

Ayrshire high cpi Cow list for april 2014

The CPI formula is based upon six traits with the following emphasis: 25% fat, 35% protein, 25% type, 6% daughter pregnancy rate (DPR), 5% udder depth and 4% somatic cell. Milk yield is not included as a separate trait in the formula because it is highly correlated to protein and fat yields. The top 125 cows are listed below.

Brown Swiss and Ayrshire National Convention

Wednesday, June 25 - Friday, June 27 Embassy Suites, East Peoria, Illinois (All events except tours and golf outing will be held at the Embassy Suites)

Peoria Loop Tour

Convention Tours

The shopping and sightseeing tour will provide an opportunity for a leisurely, on-your-own day. The bus will leave the hotel lobby at 9:00 am and return to pick up or drop off, every hour, making a loop of the three stops. Feel free to stop or ride until the last pick up of the day. The ride back to the hotel will be along Grandview Drive, dubbed by Teddy Roosevelt as the "world's most beautiful drive." The bluff is lined with mansions built during the early 1900's overlooking the Illinois River Valley. Meals are on your own. 1) Downtown Peoria Caterpillar Visitors Center - Spend an hour or an afternoon at the Caterpillar Visitors Center where you can experience Caterpillar on a self-guided journey at your own pace. You will find interactive displays and exhibits sure to engage and entertain visitors of all ages. www.caterpillar.com/visitors-center Peoria Riverfront Museum - The museum is a one-of-a-kind experience that lets you take a peek into the past of Peoria. See how humans have changed the Illinois River, catch a movie on the giant screen, or gaze into the stars and watch a laser light show at the planetarium. The museum is also home to an expansive collection of fine art and folk art. www.peoriariverfrontmuseum.org

Riverfront Museum 2) Glen Oak Park Peoria Zoo is open all year long and home to more than 100 species of animals from around the world, including several which are endangered. The zoo is located in beautiful Glen Oak Park, located in the heart of the city, preserving ancient trees with plenty of playground

areas. www.peoriazoo.org Luthy Botanical Gardens - A gorgeous spot which features five acres of landscaped gardens and a tropical conservatory. www.peoriaparks.org/luthy-botanicalgarden

3) Peoria Heights Shopping - Stroll the quaint streets of Peoria Heights and discover the unique businesses and speciality shops lining the historically beautiful downtown area. Clothing, fun accessories, wine, gourmet food, custom-made jewelry, gifts or fine art. Restaurants and a park are also available or view the river valley from the top of the Tower. The River Valley and Mackinaw Winery Tour will also depart from the hotel at 9:00. The first stop will be River Valley Dairy. The group will then travel to the Mackinaw Winery for lunch followed by wine tasting.

River Valley Dairy Owned by the Gregg Sauder family and located near Tremont, Illinois, the feature of this stop will be the robotics used on the dairy. There will also be a chance to take a look at one of the nation’s special Jersey herds. A trip to a Wisconsin sale in 2004 and the purchase of two animals “kicked things in gear for the family.” In 2006 Greg and Cindy Sauder moved to their current location with their family of seven children and nine Jersey cows being milked with a vacuum pump and one milker. Shortly thereafter they purchased 50 cows from an Iowa herd and they were off. Since the entering the showring on the national level in 2007, they have had numerous All-American honorees and several Grand Champions at both World Dairy Expo and The All American Jersey Show in Louisville, open and/or junior shows. They strive to work with cows that are going to impact the breed both in the genomic world and the show ring. In January 2012 the larger part of the herd moved into a new robotic milked freestall barn. The barn has the capacity

to hold 300 milking cows. The four Lely Astronaut robots are equipped with activity monitors, rumination sensors and other management tools. The cows are being milked on an average 3.8 times daily on the robots. The spacious freestall barn is equipped with gel mattresses, large alleyways and a drive through feed alley. Feed is pushed up once an hour by a robotic feed pusher.

Mackinaw Valley Vineyard Following lunch at the winery, you will have the opportunity for a wine tasting with over 20 wines from which to choose. Situated on a glacial moraine with a natural high point, the winery’s tasting room provides an amazing 30 mile view of the valley and farms below, a beautiful and relaxing experience. GONADORELIN HYDROCHLORIDE For Injection For the treatment of cystic ovaries in cattle. CAUTION Federal law restricts this drug to use by or on the order of a licensed veterinarian. DESCRIPTION FACTREL (gonadorelin hydrochloride) is a sterile solution containing 50 micrograms of synthetic gonadorelin (as hydrochloride) per mL in aqueous formulation containing 0.6% sodium chloride and 2% benzyl alcohol (as a preservative). Gonadorelin is the gonadotropin releasing hormone (GnRH) which is produced by the hypothalamus and causes the release of the gonadotropin luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from the anterior pituitary. FACTREL (gonadorelin hydrochloride) has the identical amino acid sequence as endogenous gonadorelin; 5-oxo Pro-His-Trp-Ser-TyrGly-Leu-Arg-Pro-Gly-NH2 with identical physiological activities. The molecular weight of gonadorelin is 1182 with a molecular formula of C55H75N17O13. The corresponding values for gonadorelin hydrochloride are 1219 (1 HCI) expressed as C55H75N17O13HCI, or 1255 (2 HCI) expressed as C55H75N17O13 2HCI. MECHANISM OF ACTION Follicular cysts are enlarged non-ovulatory follicles resulting from a malfunction of the neuroendocrine mechanism controlling follicular maturation and ovulation. Exogenous administration of agents possessing luteinizing hormone (LH) activity, such as pituitary extracts or human chorionic gonadotropin, often causes ovulation or regression of follicular cysts. FACTREL induces release of endogenous luteinizing hormone (LH) to produce this same effect. No significant differences have been demonstrated in days from treatment to conception, frequency of cows conceiving at first or subsequent heats, or conception rates among treated or non-treated control animals. INDICATIONS FACTREL (gonadorelin hydrochloride) is indicated for the treatment of ovarian follicular cysts in cattle. The treatment effect of FACTREL when used in cattle with ovarian follicular cysts is a reduction in the number of days to first estrus. DOSAGE The recommended dosage of FACTREL is 100 mcg/ cow intramuscularly. RESIDUE WARNING Because FACTREL is identical to endogenous GnRH such that both are rapidly metabolized without detectable levels in milk or tissue, no withdrawal period is required. STORAGE CONDITIONS Store at refrigerator temperature 2° to 8°C (36° to 46°F). SAFETY AND TOXICITY In cows the intramuscular administration of up to 25 times recommended dosage (2,500 mcg/day) of FACTREL for 3 days did not affect any physiological or clinical parameter. Likewise, single intramuscular doses of 5 times recommended dosage (500 mcg) did not interfere with pregnancy. No evidence of irritation at injection site was found in any animal. HOW SUPPLIED FACTREL (gonadorelin hydrochloride) solution 50 mcg/mL is available in 20 mL multidose vials (box of one). INDICATIONS AND INSTRUCTIONS FOR USE Cattle: LUTALYSE Sterile Solution is indicated as a luteolytic agent. LUTALYSE is effective only in those cattle having a corpus luteum, i.e., those which ovulated at least five days prior to treatment. Future reproductive performance of animals that are not cycling will be unaffected by injection of LUTALYSE. 1. For Intramuscular Use for Estrous Synchronization in Beef Cattle and Non-Lactating Dairy Heifers. LUTALYSE is used to control the timing of estrus and ovulation in estrous cycling cattle that have a corpus luteum. Inject a dose of 5 mL LUTALYSE (25 mg PGF2 ) intramuscularly either once or twice at a 10 to 12 day interval. With the single injection, cattle should be bred at the usual time relative to estrus. With the two injections, cattle can be bred after the second injection either at the usual time relative to detected estrus or at about 80 hours after the second injection of LUTALYSE. Estrus is expected to occur 1 to 5 days after injection if a corpus luteum was present. Cattle that do not become pregnant to breeding at estrus on days 1 to 5 after injection will be expected to return to estrus in about 18 to 24 days. 2. For Intramuscular Use for Unobserved (Silent) Estrus in Lactating Dairy Cows with a Corpus Luteum. Inject a dose of 5 mL LUTALYSE (25 mg PGF2 ) intramuscularly. Breed cows as they are detected in estrus. If estrus has not been observed by 80 hours after injection, breed at 80 hours. If the cow returns to estrus breed at the usual time relative to estrus. Management Considerations: Many factors contribute to success and failure of reproduction management, and these factors are important also when time of breeding is to be regulated with LUTALYSE Sterile Solution. Some of these factors are: a. Cattle must be ready to breed — they must have a corpus luteum and be healthy; b. Nutritional status must be adequate as this has a direct effect on conception and the initiation of estrus in heifers or return of estrous cycles in cows following calving; c. Physical facilities must be adequate to allow cattle handling without being detrimental to the animal; d. Estrus must be detected accurately if timed Al is not employed; e. Semen of high fertility must be used; f. Semen must be inseminated properly. A successful breeding program can employ LUTALYSE effectively, but a poorly managed breeding program will continue to be poor when LUTALYSE is employed unless other management deficiencies are remedied first. Cattle expressing estrus following LUTALYSE are receptive to breeding by a bull. Using bulls to breed large numbers of cattle in heat following LUTALYSE will require proper management of bulls and cattle. 3. For Intramuscular Use for Treatment of Pyometra (chronic endometritis) in Cattle. Inject a dose of 5 mL LUTALYSE (25 mg PGF2 ) intramuscularly. In studies conducted with LUTALYSE, pyometra was defined as presence of a corpus luteum in the ovary and uterine horns containing fluid but not a conceptus based on palpation per rectum. Return to normal was defined as evacuation of fluid and return of the uterine horn size to 40 mm or less based on palpation per rectum at 14 and 28 days. Most cattle that recovered in response to LUTALYSE recovered within 14 days after injection. After 14 days, recovery rate of treated cattle was no different than that of nontreated cattle. 4. For Intramuscular Use for Abortion of Feedlot and Other Non-Lactating Cattle. LUTALYSE is indicated for its abortifacient effect in feedlot and other non-lactating cattle during the first 100 days of gestation. Inject a dose of 25 mg intramuscularly. Cattle that abort will abort within 35 days of injection. Commercial cattle were palpated per rectum for pregnancy in six feedlots. The percent of pregnant cattle in each feedlot less than 100 days of gestation ranged between 26 and 84; 80% or more of the pregnant cattle were less than 150 days of gestation. The abortion rates following injection of LUTALYSE increased with increasing doses up to about 25 mg. As examples, the abortion rates, over 7 feedlots on the dose titration study, were 22%, 50%, 71%, 90% and 78% for cattle up to 100 days of gestation when injected IM with LUTALYSE doses of 0,1 (5 mg), 2 (10 mg), 4 (20 mg) and 8 (40 mg) mL, respectively. The statistical predicted relative abortion rate based on the dose titration data, was about 93% for the 5 mL (25 mg) LUTALYSE dose for cattle injected up to 100 days of gestation. Swine: For intramuscular use for parturition induction in swine. LUTALYSE Sterile Solution is indicated for parturition induction in swine when injected within 3 days of normal predicted farrowing. The response to treatment varies by individual animals with a mean interval from administration of 2 mL LUTALYSE (10 mg dinoprost) to parturition of approximately 30 hours. This can be employed to control the time of farrowing in sows and gilts in late gestation. Management Considerations: Several factors must be considered for the successful use of LUTALYSE Sterile Solution for parturition induction in swine. The product must be administered at a relatively specific time (treatment earlier than 3 days prior to normal predicted farrowing may result in increased piglet mortality). It is important that adequate records be maintained on (1) the average length of gestation period for the animals on a specific location, and (2) the breeding and projected farrowing dates for each animal. This information is essential to determine the appropriate time for administration of LUTALYSE. Mares: LUTALYSE Sterile Solution is indicated for its luteolytic effect in mares. This lutelytic effect can be utilized to control the timing of estrus in estrous cycling and clinically anestrous mares that have a corpus luteum in the following circumstances: 1. Controlling Time of Estrus of Estrous Cycling Mares: Mares treated with LUTALYSE during diestrus (4 or more days after ovulation) will return to estrus within 2 to 4 days in most cases and ovulate 8 to 12 days after treatment. This procedure may be utilized as an aid to scheduling the use of stallions. 2. Difficult-to-Breed Mares: In extended diestrus there is failure to exhibit regular estrous cycles, which is different from true anestrus. Many mares described as anestrus during the breeding season have serum progesterone levels consistent with the presence of a functional corpus luteum. A proportion of “barren”, maiden, and lactating mares do not exhibit regular estrous cycles and may be in extended diestrus. Following abortion, early fetal death and re-sorption, or as a result of “pseudopregnancy,” there may be serum progesterone levels consistent with a functional corpus luteum. Treatment of such mares with LUTALYSE usually results in regression of the corpus luteum followed by estrus and/or ovulation. In one study with 122 Standardbred and Thoroughbred mares in clinical anestrus for an average of 58 days and treated during the breeding season, behavioral estrus was detected in 81 percent at an average time of 3.7 days after injection with 5 mg LUTALYSE; ovulation occurred an average of 7.0 days after treatment. Of those mares bred, 59% were pregnant following an average of 1.4 services during that estrus. Treatment of “anestrous” mares, which abort subsequent to 36 days of pregnancy, may not result in return to estrus due to presence of functional endometrial cups. USER SAFETY (HUMAN WARNINGS) Not for human use. Women of childbearing age, asthmatics, and persons with bronchial and other respiratory problems should exercise extreme caution when handling this product. In the early stages, women may be unaware of their pregnancies. Dinoprost tromethamine is readily absorbed through the skin and can cause abortion. Accidental spillage on the skin should be washed off immediately with soap and water. RESIDUE INFORMATION No milk discard or preslaughter drug withdrawal period is required for labeled uses in cattle. No preslaughter drug withdrawal period is required for labeled uses in swine. Use of this product in excess of the approved dose may result in drug residues. Not for horses intended for human consumption. ANIMAL SAFETY (WARNINGS) Severe localized clostridial infections associated with injection of LUTALYSE have been reported. In rare instances, such infections have resulted in death. Aggressive antibiotic therapy should be employed at the first sign of infection at the injection site whether localized or diffuse. PRECAUTIONS • Do not administer intravenously (I.V.) as this route may potentiate adverse reactions. • No vial stopper should be entered more than 20 times. For this reason, the 100 mL bottle should only be used for cattle. The 30 mL bottle may be used for cattle, swine, or mares. • As with all parenteral products, careful aseptic techniques should be used to decrease the possibility of post-injection bacterial infections. The vial stopper should be cleaned and disinfected prior to needle entry. Only sterile needles should be used and the same needle should not be used more than once. • Nonsteroidal anti-inflammatory drugs may inhibit prostaglandin synthesis; therefore this class of drugs should not be administered concurrently. Cattle: Do not administer to pregnant cattle, unless abortion is desired. Cattle administered a progestogen would be expected to have a reduced response to LUTALYSE Sterile Solution. Swine: Do not administer to sows and/or gilts prior to 3 days of normal predicted farrowing as an increased number of stillbirths and postnatal mortality may result. Mares: LUTALYSE Sterile Solution is ineffective when administered prior to day-5 after ovulation. Pregnancy status should be determined prior to treatment since LUTALYSE has been reported to induce abortion and parturition when sufficient doses were administered. Mares should not be treated if they suffer from either acute or subacute disorders of the vascular system, gastrointestinal tract, respiratory system, or reproductive tract. ADVERSE REACTIONS Cattle: Limited salivation has been reported in some instances. Swine: The most frequently observed side effects were erythema and pruritus, slight incoordination, nesting behavior, itching, urination, defecation, abdominal muscle spasms, tail movements, hyperpnea or dyspnea, increased vocalization, salivation, and at the 100 mg (10X) dose only, possible vomiting. These side effects are transitory, lasting from 10 minutes to 3 hours, and were not detrimental to the health of the animal. Mares: The most frequently observed side effects are sweating and decreased rectal temperature. However, these have been transient in all cases observed and have not been detrimental to the animal. Other reactions seen have been increase in heart rate, increase in respiration rate, some abdominal discomfort, locomotor incoordination, and lying down. These effects are usually seen within 15 minutes of injection and disappear within one hour. Mares usually continue to eat during the period of expression of side effects. One anaphylactic reaction of several hundred mares treated with LUTALYSE Sterile Solution was reported but was not confirmed. NADA #108-901, Approved by FDA
