Introduction of New SAC Director - Richard Grudeski Hi, my name is Rick Grudeski. My wife Gillian and I live at Vista, Manitoba. I have been breeding Salers cattle since 1979. I was first introduced to Salers while attending Olds College in 1978. I came home from college and started using A.I. on my commercial cows and in 1981 purchased my first purebred cow from Gerald Dixon at Agribition. As they say the rest is history. At Olds College I met my wife Gillian and together we have raised five children who have grown and have now left home and pursuing their own careers with plans of possibly returning home to the family farm. At present we have a mixed farm consisting of 60 purebred Salers cows, 40 Percheron heavy horse mares and a flock of 80 breeding ewes. We love, love, love the rural farm life and our livestock. Our Salers cattle have pulled us through many hard times: BSE, TB in Manitoba, droughts and floods. I consider it an honor and a privilege to be asked to serve as a director on the board for the Canadian Salers Association. Our door is always open and coffee always available, so stop by for a visit or give us a call. Everybody has a story to tell which will usually bring a smile to ones face which is as healthy as fresh country air. Take care and talk to you soon. Rick Grudeski

President - Peter Watkins What exciting times in the Canadian Salers Association! We as an association are working on the advancement of our EPD system. The technology now exists to merge our EPD’s with our genomic data bank. The American Salers Association has ask for our participation in this initiative. International genetic solutions internationalgeneticsolutions.com has already created similar platforms for many other beef breeds in North America. This will be an important stepping stone in how we will select and adjudicate breeding stock in the future.

DNA:
We have funding available from CBBC. The board of directors decided to have all walking sires tested on a GGP 50K SNP panel. This will allow to generate a greater training population with DNA results added to the enhanced EPD calculations. The GGP 50k SNP panel will cost our members $10 more than a low density SNP panel.
It was an honour to be selected as President of the Association at the past AGM. Our past president cast a long shadow, and I look forward to his mentorship, as well as collaboration with our great group of directors. The Salers breed possesses characteristics that other breeds are constantly trying to attain. It is all our responsibility to continue to champion the virtues of our breed to anyone who will listen. Walking Sires:
Please remember to pull hair on all your walking sires. Store in a paper envelope, clearly labeled in a filing cabinet. DNA reconstruction is very costly (see page 14&15). With the new registry system implemented in the fall of 2016, it looks like there was a glitch in the system not warning when there is no DNA on file on a walking sire. This has been corrected now. Once in a while the updated program catches a walking sire with no DNA on file when a new offspring is recorded. Fullbloods and ET calves need parentage verification, this means DNA on dam, sire and calf to be registered.

DNA Reconstruction Michelle Miller, CEO Delta Genomics, Edmonton Many of you can appreciate that feeling when you get a notice from the association office that a sample is required for DNA testing, and your stomach drops, because that animal is long gone and there’s no possible way to get a sample from it. Upon speaking with the association office you hear about this thing called a “reconstruction” that could potentially solve the problem of not having a sample from the animal, but the whole process sounds confusing and expensive. That’s because it is. In this article my goal is to explain what a reconstruction is, why it’s expensive, and hopefully convince you to take samples from animals even if you don’t plan to test them right away (or ever). Starting with why this whole process is confusing is because a reconstruction is essentially a complicated math problem. If we want to reconstruct a genotype for a bull, we will compare the genotypes from the cows he bred and each of their calves. From each cow-calf pair, we work backwards to determine the genotype of the bull in question. Each cow and calf will have their own genotype which is made up of a number of markers, and markers have two pieces of information: one was inherited from the animal’s dam and the other from that animal’s sire. Once we have the reconstructed genotype, it is used like a normal genotype for parentage verification.

For example, if Marker #1 in the cow is A/A and the same marker in the calf is A/B, we know that the calf must have inherited the A from the cow (because she can only give an A) and the B from the bull, so now we know that Marker #1 in the bull is B/?. In the example in the table below, all three cow-calf pairs have the same combination for Marker #1, so we cannot determine with certainty what the other half of Marker #1 is for the bull. For markers in the cow-calf pairs where the cow and the calf are the same, that marker cannot be used to work backwards and reconstruct the bull’s genotype. For example, if Marker #2 in both the cow and the calf is A/B, we cannot tell if the cow passed on the A or the B component of Marker #2. So Marker #2 would be removed from the math problem because it doesn’t provide any certain information. In the example below, all three cow-calf pairs are all exactly the same combination for Marker #2 and it is not possible to work backwards to the bull. The ideal reconstruction markers are those that indicate that the bull is A/B. In the example in the table, Marker #3 is a perfect marker for a reconstruction. Cow-calf pair 1 showed that the cow can only pass down the A, so the bull must have provided the B. Cow-calf pair 2 confirms that the bull must have provided the calf with a B because the cow passed down one B and the bull must have passed down the other B. Cow-calf pair 3 however shows that the cow must have passed down a B so the bull must have provided the A to the calf. So the bull’s Marker #3 is A/B. calf 2

DNA Reconstruction For those of you who really enjoy math and are digging into this, you may have noticed that in a reconstruction we only use information that we’re certain is true. If we look at Marker #1, one could argue that the most probable marker for the bull is B/B. Although it is true that B/B is highly probable for Marker #1, we do not know for certain so we only consider what we know: that Marker #1 in the reconstructed bull is B/?. One final note on the complexity of reconstructions is that they are the unique situation where the older microsatellite technology works better than the newer SNP technology. The example in the chart above is an example of a SNP reconstruction. SNPs are binary pieces of information, which means there are only two options: A or B. Microsatellite markers however are much more complex and can include A, B, C, D, E, etc… With more possibilities to compare using microsatellites, fewer cow-calf pairs are required to complete a bull reconstruction. So how many cow-calf pairs are required for the best chance at a successful reconstruction?
When we are using microsatellites, the answer is at least 5 cow-calf pairs or 10 individuals. When we are using SNP markers, we need at least 10 cow-calf pairs or 20 individuals with high density (ie: 50K) SNP genotypes. We want to avoid including the same cow with multiple calves in reconstructions to get the best chance of finding those ideal markers. At this point, you’re probably seeing how this can get expensive. To get one reconstructed genotype for a bull, the cheapest option is to pay for 10 microsatellite profiles at $50 each. That’s a $500 math problem! So how can we avoid reconstructions? The answer is simple: pull samples from animals even if you don’t plan on testing them. Even though we all do our best to avoid these situations, you never know if a bull will get in with cows he’s not supposed to breed before he gets on the truck or if one of your walking bulls ends up with the wolves. Having a box of hair samples that you may never use is much cheaper on both your wallet and your sanity.

Guidelines
for collecting hair root samples for DNA and SNP Genotyping
COLLECT HAIR FROM THE TIP OF THE TAIL (SWITCH). THE ROOT ENDS CONTAIN THE DNA. SAMPLES ARE TO BE PULLED NOT CLIPPED. 1. Clean the tail switch to remove any foreign material. Comb or brush the tail to remove any dead hair. If needed, wash clean and rinse with water. Wait for the tail to be completely dry. The sample must be free of urine or manure. Dirty samples will not be processed. Contamination will make the sample unfit for testing. 2. Wrap approximately 5-10 strands of hair around a finger, about 2 inches away from the skin, and give a sharp pull. Inspect the hair to ensure that the follicles are attached. Hair strands without follicles do not contain DNA and cannot be tested. 3. Repeat step #2 until you have obtained approximately 40-50 hair roots 4. Place all of the hair roots at one end with the long strands pointing straight down. Secure the hairs together with adhesive tape wrapped approximately 1 inch from the follicles. Place the sample in a labelled envelope and immediately seal to minimize contamination. Hair from only one animal is to be placed in each envelope. 5. If doing more than one animal, WASH HANDS before starting on the next animal or use a clean pair of surgical gloves. This will reduce the risk of cross-contamination of the samples. Repeat steps 1-4 for each animal. 6. Place the identified hair sample envelope in a larger envelope an mail to the Salers office in Calgary.
