Treating Rare Diseases with Human Blood Plasma Proteins Did you know that your blood contains over 200 plasma proteins that are vital to your health? Key proteins like albumin and immune globulins have been used as medicinal products since the 1940’s and later a number of coagulation factors used to treat haemophilia. Products are isolated using a variety of processes that collectively are known as “fractionation”. With advances in protein separation other proteins are being isolated and studied clinically to treat other rare diseases. An example, plasminogen, a coagulation associated protein, is vital for prevention of excessive clotting in the body. Patients with low concentrations of this protein form clots throughout their bodies. It is classified as a rare disease with only 1.6 cases/million population. In the past, surgery has been used to remove clots or in some cases injection of plasma which contains plasminogen both treatments with limited efficacy. Prometic Life Sciences has completed advanced clinical trials to demonstrate that a purified, stabilized plasminogen product is both safe and effective to dissolve these clots and prevent their reformation. Product availability was enabled by Prometic’s proprietary plasma separation technology where binding ligands have been developed that can pull plasminogen from human plasma using a series of purification resins with a cascade process. Human plasma is a vital starting material for the manufacture of many life-saving drugs. It is the clear, straw-colored liquid portion of blood that remains after red blood cells, white blood cells, platelets and other cellular components are removed. Plasma is collected from healthy donors using a process called plasmapheresis where the donor’s blood is separated using a spinning membrane to separate the liquid portion (plasma) from the blood cells like red blood cells, lymphocytes and platelets. Donor’s can replace the donated proteins within 1 day so they can safely donate up to twice a week. The cellular parts are returned to the donors and the plasma is collected, tested and shipped for fractionation into drugs. Prometic Plasma Resources currently collects plasma at the University of Manitoba’s Smart Park campus https://plasma. prometic.com/. There are also three other licensed companies that can collect human plasma in Canada, CBS, HemaQuebec and Canadian Plasma Resources. Presently only 17% of Canada’s plasma requirements are collected in Canada, with the balance collected in the USA. Plans are underway to increase the amount of plasma from healthy Canadians to continue to assure supply of these vital life saving proteins for rare disease patients into the future. Ideally healthy Canadians can roll up their sleeves to help patients with these rare diseases. Bill Bees, VP Plasma Technologies, Prometic Plasma Resources Inc.
The key element in this story is the production of starting material, Human Source Plasma. Plasminogen can not be made synthetically and must be isolated from plasma from healthy donors.
