A LETTER FROM DAVID VAN ANDEL At Van Andel Institute, we’re fast becoming known – and appreciated – for challenging convention in favor of accelerating the speed of research, discovery and hope. We are proud and grateful for something else as well – a groundswell of growing support that reflects the generous hands and hearts of so many who donate their resources to our Institute. When Susan Brogger’s mother died suddenly of pancreatic cancer, she established a Purple Community® 5K walk/ run in her mother’s honor to raise awareness and funds for research in one of our labs. To date, she’s raised thousands in her beloved mother’s memory. When Dr. Willard Van Essen’s wife died unexpectedly, he decided to continue her legacy and highlight her love of science by creating a scholarship partnership with Van Andel Education Institute so that more students could participate in the “Science on Saturdays” program. Thanks to a significant donation from the Richard and Helen DeVos Foundation, and a collaboration between Van Andel Research Institute and the Spectrum Health Frederik Meijer Heart & Vascular Institute, Rich and Helen DeVos are helping the Institute support innovative research in regenerative cardiovascular therapies. We are also very grateful for the leadership and guidance of our own Dr. George Vande Woude and the Foundation for Advanced Cancer Research, whose generosity helped establish the Advanced Cancer Studies Fund at Van Andel Institute, representing the largest ever one-time donation to Van Andel Institute Graduate School. Because of our dedicated, mission-focused donors, the last two decades have been marked by extensive growth, transforming us into a biomedical research and educational institute that has earned admiration and respect around the world.

goes directly to the research labs and science education classrooms where we are shaping the future of disease treatment and therapies. In the last year, we’ve moved forward in ways that have exceeded our expectations. Leading a talented crop of new research personnel is our Research Director, Dr. Peter Jones, whose strategic plan aims to establish a national hub for epigenetic cancer and neurodegenerative disease research. Jones and his team have recruited worldclass scientists to the Institute and are working with leading research and clinical organizations to move promising epigenetic therapies into clinical trials. To further this mission, the Institute recently partnered with Stand Up To Cancer, a program of the Entertainment Industry Foundation, and committed $7.5 million over three years in support. On the educational front, the fully accredited Van Andel Institute Graduate School continues to catapult students on to promising career paths. We’ve also expanded the reach of our undergraduate internship program, which draws from a broader geography than ever before. Our Science Academy, for children as young as five, inspires thousands of students each year, and plans are under way to expand our unique science program nationally. Our corporate partnerships include dynamic entities such as iHeart Media’s B-93.7 FM, where “Country Kicks Cancer” is not only a rallying cry but a focal

point of on-air fundraising drives. Its annual Birthday Bash, to which more than 60,000 country and western music fans flock each summer, has brought our mission to an enthusiastic new audience. Singer Sheryl Crow has also become our ally and plans to perform a 2015 Nashville concert to benefit our mission. We must also thank the scores of men, women and even children who sponsor events as part of our Purple Community, raising funds to champion and commemorate those with cancer and other diseases. Through thoughtful collaborations – and the generosity of so many donors – we will keep making a difference in the lives of others: people who can’t bank on another month, week, or even day. To them, minutes count, and with your help and our efforts, we may positively affect their futures. Warmly,

Dr. Jones and his team need dynamic philanthropic efforts to move their initiatives forward. Because 100% of all donations go directly to the Institute’s labs, individual donors can make a direct impact on the Center’s continued success.

P OW E R F U L PA R T N E R S In 2014, VARI partnered with Stand Up To Cancer (SU2C), a program of the Entertainment Industry Foundation, and committed $7.5 million over three years to support the continued work of the SU2C Epigenetics Dream Team that was established in 2009. Led by Dr. Peter Jones and Dr. Stephen Baylin, the Virginia and D.K. Ludwig Professor for Cancer Research in the Oncology Department and co-head of Cancer Biology at the Sidney Kimmel Comprehensive Cancer Center at Johns Hopkins University, the VARI-SU2C Epigenetics Dream Team focuses on developing epigenetic therapies that can be applied to a variety of cancers.

R E G E N E R AT I N G T H E H E A R T Dr. Stefan Jovinge, head of the Laboratory of Cardiovascular Research, is a pioneer in the fields of cardiovascular research and regenerative medicine. Jovinge is also a professor in VARI’s Center for Epigenetics, director of the DeVos Cardiovascular Research Program (a joint effort between VARI and Spectrum Health) and medical director of research at the Spectrum Health Frederik Meijer Heart & Vascular Institute. At the center of Jovinge’s research is the regenerative potential found in the heart’s cardiac muscle cells and progenitor cells. By unlocking these codes, Jovinge aims to use the heart’s own mechanics to repair itself as well as generating new heart muscle cells from non-heart cells. Jovinge’s work is made possible through Van Andel Institute’s partnership with Spectrum Health and generous funding from the Richard and Helen DeVos Foundation. Jovinge’s work has far-reaching implications regarding therapies for those living with cardiovascular disease. His research could one day lead to treatment options that repair damaged heart tissue.

The Power of Giving Thank you, Rich and Helen DeVos The collaboration between VARI and the Spectrum Health Frederik Meijer Heart & Vascular Institute was made possible by a significant donation from the Richard and Helen DeVos Foundation. As a co-founder of Amway, Rich is a legendary entrepreneur, and the DeVoses are among the foremost philanthropists in the United States. CENTER FOR NEURODEGENERATIVE SCIENCE UNLOCKING THE POTENTIAL FOR NEW THERAPIES Moving a new Parkinson’s disease therapy from the lab to the patient costs billions of dollars and takes more than a decade, which represents two of the biggest obstacles facing potential new treatments. Repositioning medications approved to treat other diseases, which have shown promise in slowing or reversing Parkinson’s in preclinical experiments, is one way scientists and Parkinson’s disease advocates are working to cut the time and costs associated with therapy development. Van Andel Research Institute’s (VARI) Center for Neurodegenerative Science partnered with UK-based research charity The Cure Parkinson’s Trust on the Linked Clinical Trials (LCT) initiative, which aims to reposition drugs approved to treat other diseases and evaluate their potential as Parkinson’s disease therapies in clinical trials.

“We are not interested in the status quo. We are interested in urgently pursuing novel therapies that don’t just manage symptoms but that actually slow or reverse Parkinson’s and significantly improve people’s lives.” – Dr. Patrik Brundin LCT was founded on the principle that conducting smaller, more costeffective clinical trials gives scientists the opportunity to better identify which therapy is most effective while maximizing the number of clinical trials conducted. The LCT committee is chaired by Dr. Patrik Brundin, VARI’s associate research director and director of the Center for Neurodegenerative Science. The committee has already created an initial list of medications that hold promise for treating Parkinson’s disease. This unique and powerful partnership leverages innovation, existing knowledge and collaborative networks to provide better treatments and improved quality of life for people with Parkinson’s disease. 9

GRAND CHALLENGES RALLIES THE PARKINSON’S COMMUNITY More than 350 Parkinson’s disease scientists, clinicians, people with Parkinson’s and caregivers visited Van Andel Institute on September 24 and 25, 2014, for the third annual Grand Challenges in Parkinson’s Disease symposium. Hosted by VARI’s Center for Neurodegenerative Science, Grand Challenges brought together global leaders in Parkinson’s disease research and advocacy and provided a forum for people with Parkinson’s to interact with experts in the scientific community. Rallying to the Challenge, a workshop held in conjunction with Grand Challenges, highlighted the many ways people with Parkinson’s can impact the clinical trial process and accelerate access to new treatments. The event was a rare opportunity for people involved in all aspects of Parkinson’s disease to share ideas, strategize and work to improve the clinical trial process.

The Power of Giving Thank You, Ron Rutkowski Ron Rutkowski, a husband, father, businessman and philanthropist, was diagnosed with Parkinson’s disease a decade ago. Dissatisfied with the lack of available Parkinson’s clinical trials and the access to innovative therapies, Rutkowski raised more than $15,000 in three days to support Rallying to the Challenge at Van Andel Institute. Thank you, Ron, for your dedication to bolstering Parkinson’s disease research. Your support and fervor for discovery inspires others to join the fight.

CONNECTING PARKINSON’S AND DEPRESSION

BUILDING CRITICAL BRAIN MASS

When you think of Parkinson’s disease, you may think of its physical symptoms, but according to Dr. Lena Brundin, the psychiatric symptoms can be the most debilitating.

The Institute continues to expand its research portfolio and increase its impact in the field of Parkinson’s disease research by recruiting talented scientists.

Brundin, head of VARI’s Laboratory of Behavioral Medicine and associate professor at Michigan State University College of Human Medicine, studies how inflammation might play a role in depression in people with Parkinson’s.

In 2014, the Center for Neurodegenerative Science welcomed two accomplished scientists who are focused on specific areas of Parkinson’s disease research—one is an expert in inherited forms of the disease and the other focuses on how a misfolded protein might contribute to disease progression. These additions provide fertile ground for the development of new, innovative therapeutics for Parkinson’s disease.

Brundin believes lifting the stigma of depression among Parkinson’s patients and funding research on the psychiatric effects of anti-inflammatory medications might bring about a shift in how depression in Parkinson’s disease is approached.

CENTER FOR CANCER AND CELL BIOLOGY A N A R E A O F G R E AT P R O M I S E Certain types of cancer readily spread to other organs of the body in a process called metastasis, which makes treatment exceedingly difficult. This spread is responsible for the most cancer-related deaths per year in the United States, despite advances in treatment. The area surrounding a tumor, called the tumor microenvironment, plays a significant role in how a cancer spreads and responds to treatment. By better understanding the microenvironment and the mechanisms that underlie metastasis, Van Andel Research Institute (VARI) scientists aim to improve prognostics and treatment options for cancer patients. Cancer that originates in the prostate, breast or lung often metastasizes to the bone. Once there, the development of lesions can cause unbearable bone pain. Current treatments are able to relieve some symptoms but do not increase survival. To find out why these types of cancer are drawn to bone and how they can be stopped, scientists in the Laboratory of Tumor Microenvironment and Metastasis, led by Dr. Xiaohong Li, study the bone microenvironment and the processes that promote prostate cancer metastasis.

“If we can better understand the tumor microenvironment and the mechanisms that cause metastasis, we can improve prognostic and treatment options for cancer.” – Dr. Xiaohong Li The laboratory’s long-term goal is to determine the mechanisms through which the microenvironment contributes to bone metastasis in order to develop innovative, early-prognosis methods and targeted therapeutic strategies for cancer patients. At VARI, studying the tumor microenvironment and its role in metastasis are vital elements in a diverse research portfolio that is decisively focused on the future of human health.

> 67% More than two-thirds of breast and prostate cancer metastases are found in bone. Individual donors like you allow the Institute to study unique aspects of cancer metastasis, such as the tumor microenvironment, in order to develop improved prognostic methods and therapeutic strategies.

TUBEROUS SCLEROSIS COMPLEX RESEARCH ADVANCES Since launching the Pathway of Hope research initiative in 2013 with the goal of developing new, effective treatments for individuals with tuberous sclerosis complex (TSC), the VARI team has made tremendous progress. TSC causes noncancerous tumors in major organs throughout the body, and patients afflicted with the disease often suffer from epilepsy, learning disabilities and other complications. Under the direction of Dr. Jeffrey MacKeigan, associate professor and head of the Laboratory of Systems Biology, the TSC research team has screened multiple disease-relevant cell lines searching for novel genes that may be linked to TSC. The team uncovered several intriguing genes that it is investigating as potential therapeutic targets. Through collaborations with Cincinnati Children’s Hospital Medical Center and Helen DeVos Children’s Hospital, MacKeigan’s group opened a personalized medicine feasibility study, utilizing genomic data to evaluate the potential for personalized treatment plans for those living with TSC. MacKeigan is encouraged by the pace of the project and hopeful that his team’s research into TSC will help uncover information about other tumor-related neurological diseases and cancer.

The Power of Giving Thank You, Susan Brogger When Susan Brogger’s mother was diagnosed with pancreatic cancer and told there was no cure, Susan decided to take action and raise funds to support research into this devastating illness. Thank you, Susan, for raising more than $28,000 through your annual Mari J. Meyer Pancreatic Cancer Trail Run for Hope. Your funds support the creation of improved pancreatic cancer diagnostic tools that give others the hope of a brighter future.

VARI SCIENTIST AWARDED FELLOWSHIP Dr. Matthew Steensma, assistant professor and head of VARI’s Laboratory of Musculoskeletal Oncology, was named as a cowinner of the inaugural Francis S. Collins Scholars Program in Neurofibromatosis Clinical and Translational Research by Johns Hopkins University’s Neurofibromatosis Therapeutic Acceleration Program. Steensma, who is also an orthopedic oncology surgeon at Spectrum Health and holds a faculty appointment at Michigan State University College of Human Medicine, will focus his research on new treatments for neurofibromatosis type 1 (NF-1). NF-1 is a rare syndrome that causes tumors to grow throughout the body and has no unifying therapy or cure. Steensma’s clinical practice includes surgical treatment of tumors in the musculoskeletal system with a focus on NF-1, while his laboratory focuses on developing new treatment strategies. Thanks to the award, Steensma will collaborate with researchers across the globe with the hope of making great scientific strides in the development of therapies for NF-1. INQUIRY-BASED SCIENCE EDUC ATION EXPANDS

The Science Academy continued its development of the innovative QPOE2® product line in 2014 with the introduction of a step book that outlines processes for inquiry-based learning. Focused on kindergarten through second grade students, this new step book incorporates visuals and language designed for beginning readers. The book provides teachers with a framework for engaging young students in the process of thinking and acting like scientists.

A unique partnership between Van Andel Education Institute’s (VAEI) Science Academy and Ottawa Area Intermediate School District (OAISD) is empowering teachers to work together to implement progressive methods in science education for more than 40,000 students. The creation of a regional teacher network engaged more than 120 teachers from 21 middle schools and high schools to connect, share and collaborate on ways to deliver an innovative method of science instruction in their classrooms, known as QPOE2® (Question, Prediction, Observation, Explanation and Evaluation). QPOE2 is a model developed by VAEI that gives teachers the tools to implement scientific practice in the classroom. It encourages students to ask questions, take risks and acquire new knowledge. QPOE2 resources are available to educators and students as step books or as an iPad application.

“We are working to transform instructional practice and increase student engagement across science program offerings.” – Dr. Steven Triezenberg VAEI’s instructional method doesn’t change what is being taught but rather how it is taught by helping educators teach students to think and act like scientists. Instead of memorizing facts out of a book, it requires students to discover the answers to scientific questions through their own curiosity, inquiry, research and critique.

PURPLE COMMUNITY ® PARTNERS IN HOPE Community engagement is a powerful cornerstone of Van Andel Institute (VAI). Biomedical research and science education have the potential to bring about discoveries that can impact the course of human health. Because of the impactful nature of the Institute’s mission, business and community organizations of all sizes have continued to lend their generous support.

“We were inspired by the work being done at VAI, and we’ve spent the last year raising funds to support its research and education programs. Nearly everyone is affected by cancer or disease in some way, and we want to do our part. ” – Carol Smith, owner of Great Lakes Scrip Center Employees at Great Lakes Scrip Center (GLSC) completed a yearlong fundraising competition in 2014 to benefit VAI. Employees had the opportunity to meet with research staff, take a tour of the Institute and learn about the Institute’s history. Following this meeting, GLSC employees formed fundraising teams in order to raise the most funds possible. Over the course of a year, the teams raised funds by holding company-wide events and promotions. Their efforts totaled more than $51,000 at the end of the year. The experience united the organization in the service of a common cause and provided important funding to support VAI’s biomedical research and science education.

“Hops for Hope is a clever way for breweries and patrons to support Purple Community because it raises awareness and funds for Van Andel Institute in a fun atmosphere where you wouldn’t normally expect it. We’re proud to participate in this program.” - Tapistry Brewing

HOPS FOR HOPE TAPS MICHIGAN BREWERIES The rise of craft beer breweries provided promising opportunities to Van Andel Institute’s Purple Community in 2014 with the launch of Hops for Hope – a collaborative effort among breweries to raise funds for cancer research through the sale of beer. Hops for Hope included seven breweries in its inaugural year, with locations ranging across a wide landscape, including Tapistry Brewery and Saugatuck Brewing Company, plus 11 additional restaurants and pubs. Proceeds were generated through the sale of beer growlers and select pints of beer, totaling more than $10,000 over a two-month period from September through October. Bill and Jenn White, owners of White Flame Brewing Company, have a close tie to the cause. The couple helped establish Hops for Hope at the Institute – where Jenn has worked in the Laboratory for Cancer and Developmental Cell Biology for 12 years – after her father lost his fight with lung cancer. “My father was the healthiest person I knew,” said White. “For him to be diagnosed with metastatic lung cancer was shocking and incomprehensible, but it taught me a valuable lesson: no one is immune from its devastation.” Thanks to the Whites’ vision and close connection with Michigan brewers, Hops for Hope will continue to raise important funds for the Institute’s research initiatives in 2015.
