Cover Story

Tips to make your laboratory Clean Cleaning equipment and supplies are essential for the efficient operation of the modern research lab leaning equipment and supplies are essential for the efficient operation of the modern research lab. Other than the after-hours work done by the normal cleaning crew, cleaning is done on a regular basis by the researchers to maintain the safety of the lab environment, restore the lab after accidental spills, protect the lab equipment and instruments, and to prevent the contamination of on-going experiments from each other. Clean lab glasswares

C

Cleaning Always wash glass labware immediately after use. If a thorough cleaning is not immediately possible, always allow the glassware to soak. If not cleaned immediately some residues may be impossible to remove. Most new glass is slightly alkaline and should be washed upon receipt and generally can be soaked in a 1% HCL or HNO3 solution before wash and DI rinse. Never soak for long periods in strong alkaline solutions as it will damage the glass.

16

Always follow up a soap or acid wash with a good DI water rinse. Always use soft brushes with a wooden or soft plastic handle to avoid abrasion. Do not use wire brushes or brushes with a wire core as it can abrade the glass.

Glass cleaners Alconox is the best as it’s not abrasive. In fact they have a full line of detergents for soaking, hand washing and automatic washers. A detergent such as non-abrasive dishwasher soap will also work well. Always use soft brushes. Always rinse glass well and do a final DI rinse. If you need to do an acid wash, always rinse the soap off the glass completely or it may cause a reaction and leave a film on the glass. There are many lab detergents available commercially such as; Mallinckrodt’s KleanAR and Chem-Solv. Texwipe and EM Science also make good cleaning detergents

Clean Lab Coat and Masks

We carry many cleanroom lab coat and frock styles in different materials for your controlled environment. These lint-free lab coats are the perfect option when you want to cover most of your clothing. Our clean room lab coats and frocks vary in length from the waist to below the knee. Choose from a variety of clean processed, sterile and lint free lab coats from top manufacturers. For aseptic cleanrooms, we offer sterilized lint-free clean room lab coats and frocks. A cleanroom lab coat is generally worn in industrial clean rooms or laboratories where a coverall is not necessary. Class 1,000 ISO6+ cleanrooms generally require only a lab coat be worn. When working in a Class 10-100 clean room facility you would be required to wear a coverall to maintain the cleanliness of the room. Coveralls cover much more of your body and clothing since they have attached legs and sometimes hood and boot as well. Choosing the right cleanroom clothing and then figuring out how to put it on and take it off can be confusing. The main focus of a clean room is to eliminate any outside particles and contamination. One of the main ways

Cover Story this is done is by wearing non-shedding, clean, disposable apparel over your street clothes. Depending on the cleanliness level of your clean room, you may have to wear a full bunny suit coverall with attached hood and shoe

covers or as little as a lab coat, bouffant cap and some shoe covers. The most common supplies used for clean-up are paper towels, gloves, and detergents and soaps (about 80% of the survey respondents indicated using these materials). Depending upon the type of ongoing research in the lab, appropriate clean-up supplies should be readily available and regular training provided to maintain the integrity and safety of the lab environment and personnel. Commercially available spill control stations are available from suppliers,

Organize bench tops with large equipment in the back and progressively smaller equipment toward the front. Leave sufficient space to perform your work safely and to avoid reaching over items that can easily get knocked over, such as graduated cylinders or flasks (Figure 2). Keep all containers and glassware at least 2 inches away from the front of the lab bench. Put away any clean glassware that is not being used. Avoid accumulating large amounts of dirty dishes on lab benches and in sinks.Clean them when your experiment is done. Never leave glassware in the sink because it can easily break. Regularly check glassware for star cracks, chips, or cracks, and

promptly discard or repair any unsafe glassware. Discard disposable pipettes and pipette tips immediately after use. Properly secure and label all containers of chemicals/experimental intermediates. Keep drawers and cabinets closed when not in use. Promptly clean up spilled chemicals, silica gel, and any other powdered materials to eliminate respiratory hazards Remove any equipment or clutter that interferes with access to emergency equipment such as eyewash stations, safety showers, and fire extinguishers.

Antistatic Silicones: Playing Vital Role in Food and Pharma Industries ow a days, Food and Pharmaceutical industries are facing many issues regarding safe processing operations. Major processing steps involved in drug manufacturing and delivery system are of having dynamic mechanical movements. Dynamic movement of machines generate static electricity charge on the surface of components.

N

Silicone is most widely used polymer in pharma industries for fluid delivery and sealing application. Naturally, Silicone rubber is having insulating properties in nature. Developed static electric charges on polymer surface can be reason of fire while contact with flammable processing solvents. Many fire incidents happened in last few years because of this reason.

Featured Article Silicones and other polymers fall under Insulative category. There are certain additives available in market which imparts conductivity in silicones. Extent of conductivity depends upon concentration of additives in silicone matrix. Though, it is a very challenging job to develop as these additives may affect the chemical properties of

silicone. Like food grade and Pharma grade properties may alter. Let’s understand the case through below example: Consider a fluid DCM is flowing though silicone tubing with peristaltic pump RPM of 300. Over a period of

time, static electric charge gets developed on inner surface of tubing. Boiling point of DCM is 40°C and it is flammable in nature which can generate fire on exposure of static electricity charge.

TYPES OF FOOD AND PHARMA GRADE APPLICATIONS

Polymer industries are developing colored antistatic and static dissipative silicones. Generally black colored Polymer products are rarely in demand for pharma industry. It is all about art of compounding to achieve electrical properties with variety of colors possible like green, blue, yellow, grey etc.

It is expected that within coming years, antistatic silicone products will replace the market of general silicone products for dynamic applications. By: Vinay Pandey (B- Tech Rubber Technologist) Ami Polymer Pvt. Ltd. Mob: +91 8691013936 Email ID: tech@amipolymer.com

Switching off enzymes to block cell division in cancer Researchers from Sweden and the U.K. have devised a new method to curb cancer growth by shutting down a single enzyme and stopping cells from dividing.

The companies will jointly develop and make the drugs known as biosimilars and obtain regulatory approvals for them globally, apart from sharing costs and earnings, Biocon said in a statement on Thursday. Biosimilars are identical copies of highly complexbiologic drugs that are said to have the same efficacy. Biologic drugs are made through biotechnology, derived from natural sources or produced synthetically. The companies will split up global sales, with North America and the European Union going to Sandoz and Biocon looking after the rest of the world. "I think both Sandoz and Biocon have good shared values and synergetic capabilities," Kiran Mazumdar-Shaw, MD of Biocon, told ET. She said Biocon will continue its partnership with Mylan and they will

"This acquisition is an important step for increasing Torrent's presence in the United States, is consistent with our strategy of dosage form diversification, and provides us with new capabilities including manufacturing and R&D presence in the USA," the Ahmedabad-based company said on Thursday. Bio-Pharm, Inc. (BPI), which has a 75,000 sqft manufacturing facility, boasts of 10 abbreviated new drug applications (ANDAs) already approved by the US Food and Drug Administration (FDA). It has another 10 ANDAs — or applications for approval of a generic drug product — under review at FDA while its partners have an additional 17 products under development.

While switching off certain enzymes is an attractive approach to treat cancer, it can be difficult to create a drug that targets just one enzyme out of the thousands in the human body, the researchers from Uppsala University, Karolinska Institutet, and the University of Oxford said in a statement. The team zeroed in on the enzyme dihydroorotate dehydrogenase (DHODH), a membrane protein and a new cancer target. They used a method called nondenaturing mass spectrometry, alongside molecular dynamics simulations, to study how lipids and inhibitors bind to DHODH.

30

Market Research "To our surprise, we saw that one drug seemed to bind better to the enzyme when lipid-like molecules were present," said assistant professor Michael Landreh of Karolinska Institutet. DHODH also binds a lipid in the cell’s mitochondrial respiratory chain complex, suggesting the enzyme might use special lipids to locate the correct binding site on the cell membrane, Landreh said.

The gene-editing method CRISPR may also prove useful in keeping cancer growth in check. A University of Rochester team used CRISPR to remove Tudor-SN, a protein that plays a role in a preparatory phase of cell division. Cells without the protein took much longer to prepare for division, suggesting that the approach could be

GlaxoSmithKline already partners with Vectura on respiratory therapies, but some say the British pharma giant could be looking to take the pair’s relationship one step further.

Glaxo is preparing a 175-pence-pershare bid for the Chippenham, U.K.based company, sources told The Telegraph. That offer would value Vectura at close to £1.2 billion. If GSK does strike, it’ll be taking advantage of Vectura’s 2017 share price drop. In May, the FDA turned down a generic of GlaxoSmithKline blockbuster Advair from Vectura and partner Hikma, and investors have continued to pressure the stock as they await a final verdict from the regulatory

"Our simulations show that the enzyme uses a few lipids as anchors in the membrane,” said Erik Marklund of Uppsala University. “When binding to these lipids, a small part of the enzyme folds into an adapter that allows the enzyme to lift its natural substrate out of the membrane. It seems that the drug, since it binds in the same place, takes advantage of the same mechanism.” Attacking cell division to slow cancer growth is a popular idea, for the simple reason that the abnormal growth of cells is the very definition of cancer. Scientists at the Oxford Institute for Radiation Oncology and Cancer Research UK created radiation-carrying gold nanoparticles that target cell division by entering cells and shutting down telomerase. Found at the ends of chromosomes, telomerase allows cells to divide but degrades over time. In cancer, this breakdown is sabotaged, allowing the cells to multiply freely.

In it, Vectura said it would increase its focus on lower-risk pipeline development opportunities and “cease” investment in “relatively higher risk, novel molecule, early stage programs,” instead looking for partnerships in that arena. While CEO Emma Walmsley has made clear that reviving Glaxo’s pharma portfolio is a top priority,, right now all eyes are on the company’s consumer unit—at least when it comes to M&A. Canon BioMedical Launches Assays for Spinal Muscular Atrophy Canon BioMedical, Melville, NY, has introduced the Novallele copy number assay, a simplified method to investigate deletions and duplications of individual genes. Determining SMN1 and SMN2 copy number, the polymerase chain reactionbased method requires only a thermocycler capable of high-resolution melting (HRM) and a single sample replicate. The assays are bench-tested and optimized to work with multiple HRM-capable thermocyclers and numerous DNA extraction methods. With some thermocyclers, results can be generated in less than an hour. Most cases of spinal muscular atrophy are related to the deletion of a specific gene, SMN1. However, determining disease severity is a multifactor assessment relating to the copy number of a highly homologous gene, SMN2. Often, copy number variation studies of SMN1 and SMN2 require a combination of specialized instrument systems, laborious workflows, and expensive reagents.

New QuickQuant Feature in GeneSys Software Saves Time with Easy-to-Use Analysis alongside Image Capture Syngene, a world-leading manufacturer of image analysis solutions, is delighted to announce its GeneSys software has been upgraded to include QuickQuant, a new feature for rapid band quantification analysis on blots or gels. This software, available to download free of charge to existing users of specified Syngene imaging systems, saves scientists time by allowing them to accurately analyse gels and blots at the point of image capture.

For use in Syngene’sG:BOX and GeneGnome systems, QuickQuant is ideal for scientists who need to perform rapid band quantification while they are using their imager. QuickQuant makes it easy for scientists to analyse protein or DNA down to nanogram levels as soon as they have captured their gel or Western blot images, saving them the time and effort of transferring images to other analysis packages. QuickQuant can measure relative and actual amounts (in the presence of a known standard) of DNA and protein and up/down regulation of bands, all while using their Syngene imaging system. QuickQuant can also be utilised to show incidence values of bands, indicating samples that are “greater or less than” a specified value. All standard

data is displayed on a clear calibration curve. The new GeneSys software with QuickQuant retains all its user-friendly features such as guiding scientists through quick set-up using its vast database of lighting and filter conditions, which means capturing images of gels and blots stained with all commercial visual, fluorescent and chemiluminescent dyes is an effortless task.

As the name implies, flow meters measure the rate of flow of a product, usually through a pipeline or processing line. Gasoline pump flow meters at your local filling station are another common example. Standards may apply to periodic certification by calibrating flow meters to confirm that you are getting the fuel you pay for. Heating oil, natural or propane gas are also delivered via flow meters that vendors use to calculate your bill.

“Scientists often need quick results and a simple to use tool for quantifying band values,” explains Dr Martin Biggs, Divisional Manager at Syngene. “We’re excited to offer QuickQuant as an ideal addition to our GeneSys image capture package because scientists can now access this great feature, free of charge, to rapidly analyse their gels and blots, without having to leave their Syngene systems.” Story source: Syngene

How to Calibrate Flow Meters for HPLC Pumping Systems Calibrating a flow meter for HPLC pumping systems is best left to the experts. Do not try it at home. But since you landed on this post, we’ll tell you how it is done by the experts. Experts are required to calibrate flow meters for high performance liquid chromatography (HPLC) pumping systems because it is an exacting exercise involving a high-precision measuring instrument. We’ll explain why in a bit.

Other flow meters are used to measure flow rates in thousands of gallons per minute. Why Calibrating Flow Meters for HPLC Pumping Systems is Different The answer in a few words: Instead of measuring flow rates in gallons per minute, flow meters for HPLC pump systems measure rates in fractions of milliliters per minute.

But first, a brief introduction to flow meters in general. If you are familiar with flow meters you can skip to how they are calibrated for HPLC systems. What do Flow Meters Do?

and biopharmaceutical industries as well as other analytical labs. The process demands accurate, reproducible column retention time as an analyte (what is being analyzed) is pumped through the HPLC system. Consistent and accurate performance is an absolute necessity. But complicating things is that HPLC pumping systems are composed of parts subject to leaks and wear when particulates settle in their components. And unlike the gas pump or oil delivery truck HPLC pumping systems must accommodate flow rate accuracy such as 0.05 to 25 milliliters per minute. This demands care to avoid or at least minimize wear on HPLC pump system parts. But equipment wear does occur and accuracy impacted meaning HPLC pump system testing is required.

Self-adhesive strip plays a crucial part in sealing and spacing throughout a multitude of different industrial sectors. Strips that are manufactured from EPDM make ideal seals against any ingress of water or air. Self-adhesive strip has a closed cell structure and therefore don’t allow any liquid, gases or air to pass through them, creating a perfect airtight seal on your products. The compression of the EPDM base material allows for uneven surfaces to be securely sealed once clamped down onto. EPDM has excellent resistance to

UV and ozone and therefore is widely used in external applications when parts will be subject to weathering. EPDM is a relatively cheap sponge material choice making it a cost effective solution to most sealing application.

Advantage of EPDM self adhesive strip: Easy to install, just peel off the self-adhesive backing. The adhesive backing tape used is not fully resistance to water, therefore where there is a large amount of moisture or water present; it is advisable to use plain expanded EPDM and a good conduct adhesive. EPDM provide excellent resistance against oil, acid, alkalis and has good weathering against heat and ozone. Ami Polymer Pvt.
