uly marked 20 years since Dolly the sheep, the first mammal to be successfully cloned from an adult cell was created. Now, new research has revealed that four clones who were derived from the same cell line as Dolly recently celebrated their 8th birthdays. The study, which was published in Nature Communications, revealed that years later these clones are still in good health and are ageing well. Debbie, Denise, Dianna and Daisy were all created using a technique known as somatic-cell nuclear transfer (SCNT) in 2007 and formed part of the study that was led by Professor Kevin Sinclair, Professor of Developmental Biology at the University of Nottingham in the UK. In this study, Prof Sinclair assessed the health status of the cloned sheep over the period from their birth until last year through a variety of tests including x-rays, MRIs and even glucose tolerance tests. The sheep also underwent blood pressure and musculoskeletal investigations. The research revealed that all clones, despite their advanced age for sheep, appeared to be in good health with none of them having any detrimental health effects or signs of irregular aging. When Dolly was five years old she was diagnosed with osteoarthritis, which researchers attributed to premature ageing as a result of the cloning process using SCNT. So, in the current study Prof Sinclair assessed the bone structure of each of the clones through specific tests. The tests revealed that Debbie was the only clone who appeared to have signs

of a mild case of osteoarthritis. However, unlike in Dolly’s case, the researchers attributed Debbie’s mild osteoarthritis to the effects of normal ageing and not to the cloning procedure. This new research does offer further insight into cloning mammals using the SCNT technique, which has long been contentious topic amongst the global scientific community.
History of cloning While Dolly the sheep was by no means the first clone to be created, this famous sheep from Scotland was the first mammal to be successfully cloned using SCNT and grew to become one of the most talked about topics of the 20th century. Dolly was the creation of British Developmental Biologist, Sir Ian Wilmut and his team of researchers at the Roslin Institute at the University of Edinburgh. On the 5th July 1996, Dolly, who would become one of the most famous sheep in history, was born. The sheep was named after Dolly Parton, the famous country and western singer, because she was created from a mammary gland.

How Dolly was created Dolly was created by scientists at Roslin Institute through a process known in developmental biology as SCNT using a cell from the mammary gland of a six year old ewe. In SCNT, the nucleus of a somatic cell, which contains the genetic material of any cell, is removed from the rest of the cell. Simultaneously, the nucleus of an egg cell is removed. The nucleus of the somatic cell is then implanted into the enucleated egg cell. After the nucleus is inserted into the egg cell, electric pulses are applied to the cell to stimulate the nucleus to start dividing. After a few divisions, the cell forms an embryonic structure known as a blastocyst, which contains DNA that should be genetically identical to that of the original organism. After approximately six days, once the researchers have determined that embryo is developing normally, it is then implanted into a surrogate mother who will carry the developing embryo to full term and give birth to the clone.
20 years later… It has been two decades since the birth of Dolly and while research regarding cloning and the SCNT technique in particular, still continues, there have been no remarkable breakthroughs in this branch of science during this time. However, the ethical and moral debates surrounding cloning and its methods still persist. Nevertheless, Dollies four clones do offer some key insight into better understanding the cloning process and its consequences. The famous sheep, Dolly was put down in 2003 at the age of six as a result of a progressive disease, which caused tumours to develop on her lungs. However Debbie, Denise, Dianna and Daisy have already outlived the famed sheep by a year and could be the key to unlocking the mysteries around cloning and its consequences.
In the spirit of Women’s Month, we are celebrating the amazing women behind LMS, both the new and old faces. I
SO/IEC 17025 states in para 5.9.1, “The laboratory shall have quality control procedures for monitoring the validity of tests and calibrations undertaken. The resulting data shall be recorded in such a way that trends are detectable and, where practicable, statistical techniques shall be applied to the reviewing of the results. This monitoring shall be planned and reviewed and may include, but not be limited to, the following: b) participation in interlaboratory comparison (ILC) or proficiencytesting (PT) programmes”. Furthermore, SANAS requires that calibration laboratories comply with the requirements of document R-48 “Proficiency Testing and other Comparison Programme requirements for Calibration Laboratories”. The first part of this 2-part article, will discuss the reasons for participation in PT programmes and the considerations when planning PT activity. All test and calibration laboratories are required to continuously assure the quality of the measurement results they report. The degree of rigour employed by a laboratory to do this is directly related to the management of the risk. What is the risk associated with the reporting of a technically invalid measurement result? Obviously, the risk associated with the testing or calibration of one instrument per month, which is used to measure the colour of paint, may be very different from the risk associated with the testing of a thousand blood samples a day, the results of which will be used to determine appropriate medical intervention, or the calibration of a load-cell used to measure the maximum load that can safely be lifted by a crane. In short, the risk depends on both the potential impact an erroneous measurement result could have, as well as the potential additional workload and financial liability the laboratory would have if the tests or calibrations had to be repeated. PT is one tool available to laboratories to manage this risk to an acceptable level and
continuously assure the quality of the measurement results being reported. Unfortunately, all too often laboratories participate in PT at great expense, time and for the wrong reasons. This results in the objective of participation – to assure the quality of results, not being adequately met. SANAS Technical Assessor and metrology consultant EP Tarnow has 30 years of experience as a metrologist and has been directly involved in the planning of PT, the participation therein, the analysis and reporting of results, and the assessment thereof. This paper is intended to share these experiences and perhaps assist the laboratory community in appropriate PT activity to assuring that the quality of measurement results is met.
Planning PT activity The summarised steps of planning PT activity are as follows: Define the proficiency objective of the PT activity Appoint a pilot laboratory Define the parameter to be measured or tested Define the measurement ranges or equipment/tests to be covered Determine the Target Uncertainty of Measurement Define the reference value and how it will be determined Select suitable participants Define the means of analysing the results and evaluating performance Report the results. The most important consideration is to plan the PT activity appropriately. The following aspects need to be carefully considered. 1. The desired outcome Whenever PT activity is planned it is important to determine the objective of participation. This can be one or more of the following (the list is not exhaustive): To assure the quality of measurement results produced Validate a new or revised method Prove the competence of laboratory personnel
To prove the validity of results at the extremes of an expanded range To compare your laboratory performance relative to your peers To verify a new instrument’s performance. Since the chosen objective of participation in the PT activity will have a critical influence on all further considerations, this decision must be the first step. 2. The parameter to be measured/tested by the PT Many laboratories have the capability to perform measurements over a vast range of parameters involving the calibration of a wide range of instruments. Therefore, it is important to determine whether the objective is to test proficiency in the calibration of a particular instrument, for example the calibration of an 8.5 digit Digital Multimeter (Fig 1), or to test proficiency in measuring a specific parameter such as dc voltage (Fig 2). In some instances, it may be important to test the laboratory proficiency to perform a very specific calibration covering all the parameters associated with the calibration. An example may be the calibration of a contact type digital tachometer. While the measurement uncertainties for such a calibration would probably not be at accredited Calibration Measurement Capability (CMC) level, the test would nevertheless be useful in ascertaining whether or not the laboratory was competent in all aspects of the tachometer calibration. On the other hand, it may be important to test the laboratory proficiency at performing a measurement at the smallest uncertainty possible at the limit of one of their accredited ranges. It should be remembered that in order to satisfy the requirements of SANAS R48 [2], an accredited laboratory needs to demonstrate that they are proficient at performing measurements to within their accredited CMCs over their entire accredited range of measurements at least once every five years. 3. The ranges to be covered It is obviously impossible to participate in PT activity to cover all parameters over the entire range. The question then becomes what points in the range need to be tested for proficiency. A useful approach is to analyse what methods are employed to cover each parameter and what part of the range is used for that parameter. If only one method is used for measurements over the entire range, it is probably safe to select a point or points anywhere in the range for PT participation. It is of course always beneficial to demonstrate proficiency at the extremes of the range where performing the measurements may be a little more complex. On the other hand, if several methods are used to cover a measurement range then PT activity should be performed in the range covered by each method. In the case of the calibration of instruments, many laboratories assume that if proficiency is

demonstrated in the calibration of one model of instrument, proficiency has been adequately demonstrated for all similar instruments. This however, is not always the case. Take for example the calibration of a handheld 3.5 digit DMM, which is very different to the calibration of a laboratory 8.5 digit DMM, even though they might both fall under the broad instrument category of DMM. Consequently, several models of DMMs would be required to adequately demonstrate the laboratories ongoing proficiency to calibrate these instruments. It is not uncommon however, to find laboratories that have only one ‘generic’ method for the calibration of multimetres, but testing proficiency on a handheld DMM only would not be adequate. 4. The Target Uncertainty of Measurement In Tarnow’s experience, this is probably the biggest problem with PT activity in the calibration laboratories. In most cases it is due to ignorance but there is also a cost factor that can influence decisions in this regard. Fundamentally, the PT activity Target Uncertainty of Measurement depends heavily on whether proficiency is being tested on the calibration of specific instruments or simply on the laboratory’s ability to perform a measurement of a parameter at a particular point. In the former, the PT Activity Target Uncertainty of Measurement should, as the bare minimum, be the accuracy specification of the PT Scheme artefact (instrument). This is to
ensure that the participant’s proficiency in calibrating the particular model of instrument to within the manufacturer’s accuracy specification can be adequately demonstrated. Since a typical instrument will normally be used as the PT Scheme artefact, the Reference Value Measurement Uncertainty is unlikely to be much better than the instrument accuracy specification due to limitations in the performance and stability of the instrument. Exceptional cases are possible however, when a particular instrument has been thoroughly characterised and proven to perform way better than its accuracy specification. In the case of testing the proficiency of laboratories to perform a measurement of a parameter at a specific point, the PT Activity Target Uncertainty of Measurement should preferably be the laboratory’s accredited CMC. The PT Activity Target Uncertainty of Measurement is a critical consideration since it has a significant influence on the selection of a suitable artefact and how it is characterised. 5. The determination of the reference value Contrary to popular belief, the PT Scheme Reference Value must be defined at the start of the PT activity. It may however, be determined at the end of the PT activity. At the beginning of the PT activity, there must be agreement among the participants as to what will constitute the reference value. This is known as defining the reference value. There are various ways of doing
this and each method is related to a specific set of circumstances. The reference value can be determined as the value calibrated by a higher order (smaller uncertainty) accredited calibration laboratory. In this approach however, provision must be made for some additional uncertainty contributors due to factors such as stability, drift and influence of environmental conditions. Such a calibration is usually performed before, during and after the PT activity to provide information on the stability of the artefact. The advantage of this option is that the proficiency testing of participants’ to within their accredited CMCs is possible. The disadvantage is that suitable artefacts are scarce and therefore expensive, as is their characterisation and a significant amount of data analysis is required prior, during and after the PT activity. The reference value can be determined as the manufacturer’s accuracy specification of specific model instrument as verified through a calibration by a higher order accredited calibration laboratory (laboratory with accredited CMCs smaller than the instrument accuracy specification). In this case, very few if any additional uncertainty contributors have to be considered. This calibration is usually performed as a minimum, before and after the PT activity, but additional calibrations may be performed during the PT activity as intermediate checks. This often depends on the
number of participants and the duration of the PT activity. The advantage of this method is that the artefacts are relatively cheap as they are typical commercially available instruments and their calibration can usually be undertaken by a lower level (industrial) laboratory. In addition, the data analysis is dramatically simplified. The disadvantage of this method is that proficiency to participants’ accredited CMCs is often not possible. The reference value can be the value as determined statistically from the results submitted by all the participants. In other words, it is a calculated value usually weighted by the measurement uncertainty reported by the participants. Various structured statistical methodologies exist to do this. The advantage of this method is that it is not necessary to employ the services of a high level and often expensive calibration laboratory to assign the reference value. The disadvantage of this method however, is that while the PT scheme results may indicate good agreement between the participants, this agreement may not include the ‘true value’ therefore, indicating that all the participants are equally wrong. In Figure 3 below, the reference value is indicated by NLA 1 and NLA 2. While participants 8r56 (first measurement) and 5ht9 appear to have proven equivalence with each other, since they do not overlap the reference value they have not demonstrated proficiency. Labotec team breaks away to Soweto
T
he Labotec staff breakaway is an annual event that attempts to encourage staff to escape from their usual routine and test if they are up to a challenge and if they have nerves of steel. It also rewards those who have excelled during the year. This year was no different. Labotec’s Durban and Cape Town branches joined the team at the head office in Midrand on a trip to Soweto. The first challenge had seven teams running around Bara
Market on a treasure hunt to complete a number of tasks and purchase a list of food items. The purchases from the market were taken to the Ekukhanyeni crèche and handed over to the delighted children and teachers. Giving and receiving formed a big part of the day, and around 35 people from the township not formally employed were able to receive an income for their contribution. This included the world-renowned Kliptown
Youth Programme Dance troupe who taught Labotec staff a gumboot dance routine, and then judged the somewhat hilarious performances of the seven teams as they took to the stage. Nobody danced with a straight face. Before traversing the narrow winding dust road to Kliptown, nominated team members climbed in relay format up and down the most colourful landmark in Soweto namely, the Orlando
Towers. All members of the teams met the challenges head on and with enthusiasm, including the oldest Labotec staff member who is 75 years old.

Regaining movement after paralysis
I
n a revolutionary new study, a group of eight paralysed patients were able to regain partial movement and muscle control in their paralysed limbs using a programme that combined virtual reality and a custom-made exoskeleton. Their research featured in the August issue of the journal Scientific Reports. In their paper, the scientist followed the progression of eight individuals who had been paralysed for many years on a specific programme that they had created for them for a period of one year. This specific training programme consisted of a combination of intense immersive virtual reality together with brainmachine interfaces (BMI) using a custom-designed lower limb exoskeleton capable of delivering tactile feedback to subjects to restore movement and sensation to the areas of the body that were
paralysed. After one year of the training, the researchers found that there was an improvement in both movement and sensation in the limbs of all eight of the paraplegics. All the patients experienced improvement in sensation in their limbs. Many of them also experienced voluntary motor control of certain muscles in their lower limbs. In some cases, their muscle control changed so drastically that their doctors had to upgrade them from complete to incomplete paraplegia status. The researchers believe that movement and sensation might be due to the activation of the surviving nerves in the paralysed area, however more research is required to confirm this. While this research still has far to go, it could prove to be a very valuable way of helping people suffering from paralysis regain mobility and sensation in their limbs.

NLA supports stack emission testing community
I
n line with its mandate to nurture the interests of the local laboratory community, the National Laboratory Association of South Africa (NLA-SA) is supporting the industry in defining the way forward for stack emission testing. This support is being provided in anticipation of changes in regulations that will require accreditation of all stack emission testing houses active in the country. As part of this support, the Stack Testers of South Africa (STSA), a sub-committee created within the NLA-SA in 2012 recently hosted Managing Director of PCME Ltd and chair elect of the Source Testing Association (STA), William Averdieck, to share his knowledge. In addition, stack emission testing was a focus area of the association’s annual Test & Measurement (T&M) Conference held in July. Speaking at the STSA forum held in December 2015 Averdieck, shared the rich experiences of
the STA, which was established in 1995 in the UK as a non-profit technical trade association. With corporate membership ranging from process operators, regulators and equipment suppliers to test laboratories, the STA is committed to advancing the science and practice of emission monitoring and maintaining a high quality of service to customers. It achieves these objectives by contributing to the development of industry standards, codes, safety procedures and operating principles, and by encouraging the personal and professional development of practising source testers. Averdieck provided valuable insight into the background of worldwide emission legislation and into schemes and relationships established in the UK, focusing on the structure of the MCERTS scheme for the certification of equipment, personnel and organisations. Of particular interest was the STA’s support of quality emissions monitoring in the UK as it provides valuable pointers for the STSA to define its own role in the South African environment. While air pollution control has been the subject of legislation in SA since the 1960s, it has been given renewed focus over the past decade with the publication of the National Environmental Management: Air Quality Act (AQA) to replace the outdated Atmospheric Pollution Prevention Act of 1965. The AQA has heralded an increased stringency in emission standards by South African regulators with the next step in line with international best practice being the requirement for certification of all personnel involved in emission measurements, calibration and analysis activities. The NLA-SA is the representative association for accredited and non-accredited laboratories in SA. This includes measuring, testing, calibration, verification and other bodies as well as laboratories that operate in well-defined areas of R&D in the natural and applied sciences.

Zika virus is transmitted to people through the bite of an infected mosquito from the Aedes genus, mainly Aedes aegypti in tropical regions. This is the same mosquito that transmits dengue, chikungunya and yellow fever. Doctors in SA may encounter cases in travellers returning from affected countries.

Kroonstad NHLS laboratory gets 5 stars
A
National Health Laboratory Service (NHLS) laboratory in Kroonstad in the Free State recently obtained a 5-star rating through the Africa Society for Laboratory Medicine (ASLM) external audits. This achievement makes South Africa the first and only country in Africa to have two laboratories with the highest rating according to the World Health Organisation’s, Africa Stepwise Laboratory Improvement Process towards Accreditation’s (WHO AFRO SLIPTA). There were a total of seven laboratories from the NHLS who participated in the audit of the laboratories quality management systems this year, and the Kroonstad laboratory topped the list with an historic 5-star rating. This laboratory joins Northdale laboratory in Pietermaritzburg, Kwa-Zulu Natal as one of only two
South African laboraties to have achieved this rating. The other laboratories that participated in the audit included the NHLS laboratories in Edendale, Kalafong, Leratong, Pelonomi, RK Khan and Tembisa. All of these facilities form a part of the 18 laboratory
cohort in the Strengthening Laboratory Management Towards Accreditation (SLMTA) programme, which is a programme aimed at enhancing the quality management systems of laboratories. The programme involves baseline internal audits, three workshops and three cycles
of two, one week long site visits after each workshop, and the sharing of an improvement plan and an internal exit audit before the ASLM audit. The programme was implemented with funding by the Laboratory Branch of Centre for Communicable Disease Control South Africa as a partner to the National Department of Health and the NHLS. According to the SLIPTA, some of the criteria that are required for a laboratory to achieve a 5-star rating include having a good infrastructure, staff commitment, experts on hand for advice and quality processes in place. All of which the NHLS laboratory in Kroonstad have fulfilled. This is a major achievement for South Africa and particularly the NHLS, and there is much that can be learnt from the example set by this laboratory.

Combining the two polymers helped to created a material that is printable, can maintain its shape
after being immersed in nutrients and is also able to support the growth of the stem cells within it. In their experiment, the researchers used a benchtop 3D printer to produce the bio-ink liquid, which they then turned into gel by heating the liquid to 37°C. Over five weeks, Perriman and his colleagues worked with the bio-ink and successfully differentiated the stem cells in the 3D printed structures in osteoblasts, which are the cells responsible for secreting bone in the body. During this period the scientists were able to engineer various 3D printed tissue structures including a tracheal cartilage ring. The development of this new ink for 3D printing could revolutionise the medical industry by creating a way in which to create surgical implants using patients own stem cells. A bio-ink with a difference
I
n the last few years the 3D printer has become increasingly popular around the world. These novel devices have been used to create concept cars, toys and even musical instruments. Now, a group of scientists from the University of Bristol in the UK have created a new stem-cell containing bio-ink that could be used to print living tissue using 3D printers. This novel substance could revolutionise the surgical implants industry. The researchers findings were featured in the journal Advanced Healthcare Materials. 3D bioprinting is a process of printing cells using a 3D printer. With their new discovery, the group of researchers from the British University have found a way to make this process easier and more effective. Lead researcher Dr Adam Perriman from the School of Cellular and Molecular Medicine at the University of
Bristol and his fellow researchers successfully developed a special ink that is compatible with 3D printing and tissue engineering. The special formulation is made up of two different polymer components that each have an important role to play. The first of these components is a sacrificial synthetic polymer that is used in the medical industry. This

Novel lab-on-a-chip technology
A
team from IBM has created a new lab-on-chip technology with the potential to revolutionise the medical industry. According to the report, which was published in the August edition of the journal Nature Nanotechnology, a team at the popular IT company have created a way to separate biological particles at a nanoscale, which could give researchers the ability to detect certain diseases early. IBM’s novel lab-on-a-chip technology allows researchers to separate bioparticles down to 20nm in diameter. At this size, researchers would have access to a number of particles including, DNA, viruses and even exosomes, which are present in most bodily fluids and are biomarkers for certain
medical conditions. Exosomes range in size from 20-140nm and contain information about the health of the originating cell that they are shed from. Separating these particles at this scale would
then allow medical researchers to analyse them for signs of illnesses before they even start to show any physical symptoms. Until recently, the smallest bioparticle that could be separated by size with on-chip
technologies was about 50 times or larger. The team at IBM is currently working together with researchers at the Icahn School of Medicine at Mount Sinai in New York to continue the development of this chip technology. The collaboration of researchers plan to test the device on prostate cancer. Men in South Africa have a 1 in 8 chance of developing prostate cancer, with many of these men succumbing to the condition because of late detection. Therefore, development of this lab-on-chip-technology could create a away in which to detect medical conditions at early stages so that treatment can be started immediately. Source: IBM

In their research, the Harvard scientists sought to find a way to discover blue-light emitting organic molecules, and have succeeded in doing just that. The group developed a computer-driven screening process, called the Molecular Space Shuttle. This process incorporates theoretical and experimental chemistry, machine learning and cheminformatics to quickly identify new OLED molecules that perform as well as
or better than industry standards. Through the molecular space shuttle, the researchers were able to identify a large number of highperforming blue OLED materials. They began by building libraries of more than 1.6 million potential molecules. Then, to narrow the field, a team of researchers led by Ryan Adams, assistant professor of Computer Science at the Harvard John A. Paulson School of Engineering and Applied Sciences developed machine learning algorithms to predict which molecules were likely to have good outcomes, and prioritised those to be virtually tested. In this way, they were able to predict the colour and the brightness of the molecules from a simple quantum chemical calculation and about 12 hours of computing per molecule. The team then built a web application for collaborators to explore the results of more than half a million quantum chemistry simulations. Every month, the researchers selected the most promising molecules and used their software to create profiles of each molecule. This process identified 2500 molecules with potential. The team’s experimental collaborators at Samsung and MIT then voted on which molecules were most promising for the specific application. The team ended with hundreds of potential OLED molecules. This research could provide a cheaper and a more efficient way in which to develop OLED screens and bodes well for the future of OLED technology.
Improving screens
W
ith the amount of time that is spent looking at phones, televisions and tablets, finding ways to improve screens on these devices is essential. Now a group of researchers from Harvard University in Boston, have discovered over 1000 new blue-light emitting diodes, which could lead to development of better screens for all your electronic devices. Their research was featured in the journal Nature Materials. There are various types of screens currently available with liquid crystal display screens being one of the more popular options. However, organic light-emitting diodes (OLEDs) screens are fast
becoming the better choice. OLED screens are made from organic molecules that function by emitting light when an electric current runs through them. However, while these screens offer high quailty images, they are also very expensive to produce and therefore, sell at very high prices. Now, a team of Harvard researchers in collaboration with MIT and Samsung have developed a way in which to develop OLED screens cheaper and more efficiently. OLEDs rely on green, red and blue subpixels to produce all the colours found on screen. However, it is very difficult to find organic molecules that efficiently

Prevent scarring from burns
Researchers from the Tel Aviv University and Harvard University have developed a novel; non-invasive method to prevent scarring that usually accompanies second- or third- degree burns. In their research, they used microsecond-pulsed, high-voltage, non-thermal electric fields to control the body’s proliferation of collagen cells that cause permanent scarring at the burn site. The technique partially destroys cells in the wound with short, pulsed electric fields that cause irreversible damage to the collagen cells. The researchers had to strike a balance to avoid causing further damage to the burnt area. The study was recently published in the Journal of Investigative Dermatology.

Nirghin is also one of 16 global finalists for the annual awards, to be held at the Google headquarters in the US in September.
