T

he ‘reproducibility crisis’ in the life sciences continues to make headlines, with findings that show well over 50% of published studies cannot be reproduced (some experts cite numbers as high as 90%). There are many causes. Researchers perform complex tasks with limited grant funding and face ongoing pressure to publish compelling results. Despite these challenges, reproducibility and repeatability are aspects of science that cannot be compromised on. The field needs to have confidence that published results are accurate and able to stand the test of time. Instead of changing scientists, we advocate for changing the scientific system from within. Scientists are losing valuable time struggling with data management issues, when they could instead be concentrating on meaningful discoveries. Connecting instruments with each other and with the cloud - the so-called Internet of Things (IoT) can minimise these distractions, removing human error and the disconnect between scientists and study results. In our highly connected world, saturated with personal technologies, this online transition would seem inevitable. Welldesigned and well-integrated IoT systems can improve lab efficiency, data reliability and study reproducibility. But to date, the field has been reluctant to adopt the technology and currently lacks the critical mass ‘online’ to begin solving the macro challenges.

What needs to change? It’s incredible to think that we are only now moving beyond paper notebooks in the laboratory. Most scientists still write protocols by hand and upload data to local servers. Each instrument and each scientist operates independently, and with each step in the workflow a degree of precision is potentially lost. For all its flaws, manual recording does keep scientists in control. For IoT-enabled lab systems to replace them, they need to be equally responsive. The overarching technology has to be robust and reliable - yet sufficiently versatile to allow for the huge scope and complexity of modern life science experiments. Few, if any, lab equipment and service providers

can achieve this on their own. We need industry collaboration to combine our areas of expertise into a workable, cohesive system.

A system that truly works for scientists Below are some of the lessons our teams have learned through partnering with scientists as they trial cloud-connected products and services. As an industry we have the rare opportunity to strategically build something greater than any one company can achieve, helping labs around the world transition to cloud-based technologies in a mutually beneficial way. If we don’t work together, a system of closed circuits and technology stand-offs could result, impeding our ability to advance science in a reproducible, verifiable way. To compel scientists to adopt a new technology, there has to be a clear value. IoT promises this and more. Imagine if, as scientists pipetted, the data could be used to correct their technique. Bench scientists would no longer need to write down every step in a physical notebook and then type those notes into a computer. The IoTenabled instrument would take care of data and protocol logging so the scientist could focus on their science. Data stored in the cloud would be available for any researcher to verify or reproduce published experiments - even years later. In this scenario, IoT automates and standardises mundane laboratory tasks, allowing the scientist to focus their energy and resources on the decisions that matter most. Our recent market research suggests early cloud-connected lab instruments and data systems have not catered to scientists’ needs in this way. While there is growing interest in implementing systems such as electronic note books, adoption rates remain low. There is also a trend towards customised systems built in-house, suggesting the existing designs don’t translate well in the lab. This delay and lack of standardisation helps allow the reproducibility crisis to go on. Few scientists have the time to relearn systems and no one likes to troubleshoot programme quirks or deal with technology breakdowns. IoT systems shouldn’t represent a ‘new way of doing things’ - rather,

they should enhance what scientists already do. Scientists’ careers are built around specialisation and for the most part that doesn’t include training in bioinformatics. The option exists for advanced programming, but we’ve found only a small population of scientists are capable of, or interested in, writing their own code. For automated programmes to succeed, the focus should be on the real human benefits the technology enables, not the advanced features. However, no matter how complex the innovation is, it needs to simplify and streamline lab processes.

As instruments and technologies become connected, we need to ensure that scientists have the right to choose from a full range of vendors. This will be undermined if instrument and/or data management providers decide to pursue closed systems. A closed circuit, or an incompatibility stand-off, would hurt the efficiency of laboratories and the progress of science more broadly.

Purchasing decisions

Cost-effectiveness

When it comes to purchasing decisions, costs are front-of-mind for the majority of labs - certainly those in academia. Having limited budgets was cited by our survey respondents as the number one barrier to entry when it comes to ELN and cloud-connected technologies. If vendors were to take a transparent, collaborative approach, it would be possible to achieve a highly cost-effective transition. For example, opensource software represents the collective skills and innovations of everyone who uses the technology. Scientists thus become both customers and software engineers, feeding into a collective reservoir of knowledge. With this shared responsibility, labs can connect and begin storing data in an organised way, without paying for a premium service. As we embark on this new era of science, we’re aware that an entirely different scenario could unfold. For the system to work, IoT providers must develop programmes that are compatible with new and existing software and cloud-connected instruments. The same is true for hardware. If this does not become the norm, the efficiency benefits of cloudconnected technologies will not materialise. Labs around the world need access to a universal data language. It is our belief that no

An often-overlooked concern is the possible amplification of economic inequality between labs worldwide. We get excited about the immense potential of cloud-connected instrumentation streamlining lab processes and enabling unprecedented collaboration. But if this is not done in a cost-effective way, we risk forming a separation between ‘online’ modern labs and labs in the developing world that remain offline. Clearly, large social and scientific costs are associated with marginalising any part of science’s diverse global research team. Where possible, simple open source systems should prevail, to keep costs low and accessible to all. We’re on the cusp of a fundamental transformation in how research and development is performed. It’s born out of a need to make science reproducible. With funding agencies calling for strong measures to incentivise and/or penalise academics whose findings cannot be substantiated, scientists will search for best practices to make that happen. We need to make those tools accessible and intuitive, for a critical mass to change how they programme and record experiment data. We believe cloud-connected tools are part of the infrastructure needed to solve these systemic issues.

O

ne of the biggest problems that the SA Bureau of Standards (SABS) faces is that many young people are simply not aware of the SABS and other technical infrastructure institutions, standards or benefits or the possible application of standards in their respective fields of interest. SABS launched its academic engagement strategy toolbox in 2012 to create awareness of standards and raise the overall quality levels of production in South Africa with the help of academic institutions. Several articles in the popular press as well as conferences, seminars, workshops and symposiums all point to the fact that there is a need to raise awareness in the community of standards and standardisation. These publications have reiterated that standards offer several advantages including reducing costs, facilitating communication, improving safety and providing assurance to reduce uncertainty.

Reducing our impact on the environment It follows that if we want to reduce our impact on the environment we will need to develop better products, better packaging better logistics and better processes to reduce waste, reduce emissions and manage all our other environmental aspects and the benefits that standards offer would certainly contribute to these goals. It is hoped through this initiative to unblock and expose this resource to the innovators currently associated with the teaching organisations in SA. An extension of the lack of awareness of standards is the lack of awareness (not only in

the academic community but in industry too) of the benefits and strategic advantage of participating in the development of standards nationally and internationally. The SABS has committed to improve the level of knowledge of the standards development process and to develop skills in the area and a pool of knowledge in the field of standardisation skills towards developing a cadre of standards professionals working within the SABS and in industry to develop increasingly relevant standards.

Mystery Despite the importance of standards, the business of developing and implementing standards seems to remain somewhat of a mystery to most. The SABS feels strongly that introducing standards in the ‘classroom’ would augment the learning experience by pointing learners to available design tools and best industry practices. As a result, those entering the workplace would be better placed to transition from classroom to workplace by aligning educational concepts with realworld applications and market constraints. Added to this are the much needed skills associated with understanding, developing and using standards in South African industry in order to gain not only a technical but also a competitive strategic advantage. Further, standards provide a fruitful ground for research, as has already been shown by some South African researchers.

Academic engagement strategy SABS launched its academic engagement strategy on 1 March

2012 and through this seeks to target various academic institutions around the country to increase: The level of understanding, the degree of utilisation and the intensity of participation in the development of standards by students and academics. The SABS programme to reach academic institutions is proposed to: Raise levels of awareness among students of the benefits and application of standards. Improve levels of research into standards. Improve competence in the field of standardisation. Improve knowledge on quality and conformity testing. Increase awareness of the need for quality and basics of how to drive quality improvement. Create a basic awareness of standards and how to find out more. Training on how to gain access to standards that provide knowledge resources. Raise the overall quality levels of production in South Africa.

Standards and the environment By following a standard for product safety we can be assured that our products will be safe, even if we are not an expert in the safety of that product. This assumption is based on the fact that the safety requirements were developed by experts in that field codified in standards in a coherent and consistent manner so that they can be followed by a competent person. Similarly, we do not need to be environmental experts to implement environmental standards and for us to reduce our environmental impact. Young

people are increasingly aware of the need to behave more sustainably and consequently the topic has become an important component of nearly all curricula. The use of technology in the pursuit of profit is balanced against the need to positively impact on society and reduce the impact on the environment. While not all students will be expected to be expert in environmental matters or societal engagement, they will be expected to be cognisant of the fact that they have an impact which they will need to manage. Standards are the toolbox by which this can be done. Factors such as noise, emissions, waste and effluent are common across many industries and are impacts on the environment that will need to be managed, the measurement of these impacts as well as guidance and frameworks on how to manage them are contained in standards.

The benefits of raising awareness of standards Awareness of these standards is an important skill that should be taught to young students to equip them to establish and design processes efficiently rather than facing possible litigation, penalties or waste of resources if processes need to be redesigned and deployed. In this context, it is important that students are made aware of the existence of applicable standards but also how they affect their situation, how they should be applied, how they are developed and how they can contribute to developing standards that better include their interests resulting in increasingly more relevant standards. Optimal heating applications

W

hether pre- or postheating, flame straightening, flame hardening, flame brazing, fusing or hot forming, Lindoflamm flame solutions from Afrox ensure optimum performance at the lowest processing costs for precision manufacturing processes. Unlike slow burning fuel gases, where main heat is in the secondary zone of the flame and a large amount of heat deflects from the material, resulting in gas losses, preheating time and loss of production, the unique Lindoflamm burners operate with acetylene and compressed air and the main heat contribution sits in the primary zone of the flame. This helps to direct the heat to the actual area to be preheated where very little heat is deflected off the plate. Preheating can prevent cracking and ensure specific mechanical properties and must be used whenever specified by applicable codes. Applications in the industry where Lindoflamm offers quicker preheating times, saving on production costs and labour include stationary preheating before welding and cutting (handheld pre-heating), maintaining interpass temperatures, preheat required where low alloy steels are used from minimum plate thickness of 30mm upwards, and flame straightening or flame hardening. L

ike magnets with the same polls, there is clearly a misunderstanding at opposite ends of the spectrum. We all understand whilst SA is experiencing harsh economic times, much the same as the rest of the world, the importance of securing reliable cooling appliances, is largely overlooked. Whilst the decision is being taken in procurement to provide appliances as the need arises, at the final source, either in a laboratory, or blood bank and so forth, either due to poor management or unreliable fridges and freezers, R100 000s of inventory is being disposed of annually. I would go so far as to suggest, one wouldn’t expect to find unsuitable equipment in an operating theatre, yet a similar stance does not seem to filter its way into cold chain management? Refrigerators and freezers for use in the laboratory and in research, have to meet stringent standards, especially relating to safety and temperature consistency, not only dependability. The days of cooling appliances, being merely a ‘fridge or a freezer’, should be at the very least, forgotten, currently a whole bouquet of features equip modern

laboratory cooling appliances, monitoring, temperatures, safeguarding samples, chemicals and material. Precision electronic controls enable accurate temperature settings and create optimum storage conditions in conjunction with highly effective insulation and a dynamic cooling system. Appliances for laboratory and research should provide consistent performance, even in extreme climate conditions. In many instances appliances can be purchased with a climate rating of ‘T’, or Tropicalised, therefore safely operating with an ambient temperature of 43°C. The use of ultra-modern components, efficient and environmentally-friendly refrigerants, as well as precision controls, ensure that samples, chemicals, and research materials, are stored optimally. Optional documentation software continuously logs the temperature profile, and if required, warning systems alert the user, when pre-defined temperature limits are exceeded. Integrated visual and audible alarm systems warn the user of undesired temperature deviations. Safety is essential in the laboratory and the research sector 24 hours a day, 365 days

a year. In some instances, appliances can be connected via a volt-free contact to an external remote warning system, depending on the brand. As many as 20 individual appliances can be connected in series and may even be monitored externally via a serial interface. Previously, when electricity supply was not guaranteed, resulting in numerous power failures, battery backup of up to 72 hours could be secured, therefore preventing any data loss. The calibration option allows precise temperature control, adapted to specific applications. If external alarm, and monitoring systems have been connected, then data transfer to these is also available. Several laboratory appliances have an access port in the ceiling of the appliance, therefore allowing a temperature sensor to be positioned in the interior. All reputable laboratory appliances are designed according to ISO60068-3 standard with regard to maximum temperature stability. With the competitive nature of the appliance industry, appliances are specially constructed for intensive professional use, securing the highest

quality materials, and perfect workmanship, to the finest detail. Durability and reliability are given top priority in material selection for laboratory appliances. Quality and ease of use are paramount in the design of appliances, in some instances components are integrated into the ceiling area, allowing for ease of access in the unlikely event of product failure. Although not supported by all manufacturers, and in order not to inconvenience the operators in the labs when appliances are defrosting, a ‘hot gas method’ is used, versus electric defrosting, the net result being less often and faster defrosting intervals. The controller calculates the optimum defrost cycles, based on the compressor run time. Refrigerators now defrost within eight minutes, and freezers in 12 minutes, prior to defrosting the internal temperature is lowered, as result, the temperature remains constant during the defrost cycle. In closing, we continue to hear about shortages of blood, or the lack of medicines, however if the chain clearly understood one anothers challenges, the larger investment in advance, would be vastly out-weighed by the savings in the future.

Microbial identification: Know your risk

A

number of problems associated with the microbiological contamination of pharmaceutical products have been seen. Pharmaceutical companies spend a lot of time and money reworking product due to microbial contamination. The Pharmacopoeias does not give very specific guidance to identification of organisms, but they make a big statement when they say: “The significance of microorganisms in pharmaceutical products should be evaluated in terms of the use of the product, the nature of the product, and the potential hazard to the user”. With this statement, they fully place the responsibility on the manufacturing company to deal with any potential microbial risks.

Do you know which organisms pose a risk in your facility and final product? The Pharmacopoeias recommends that certain microbial categories be routinely tested for total counts and specified indicator microbial contaminants, such as Salmonella, E. coli, P. aeruginosa and S. aureus and in some cases for yeasts and moulds. An article published in the American Pharmaceutical Review in 2012, indicated that 87% of all product recalls were due to microbial contamination of which 72% of these recalls did not fall within the scope of the indicator organisms tested for on a routine basis. What is even more alarming was that 21% of the objectionable organisms were due to yeast and moulds contamination. To know which objectionable organisms you are dealing with, you need to identify the organisms you find in the environment and final products on a regular basis.

What is the level of testing, monitoring and identification of bacteria and yeast and moulds in your facility? In the American Pharmaceutical Review, Dr Tim Sandle states: Microbial identification plays an important role in pharmaceutical processing. Microbial identification can be defined as “microbial characterisation by a limited spectrum of tests pre-chosen and appropriate to the problem being studied”. Microbiologists should understand the quantity and the types of microorganisms present in their pharmaceutical environment.

You have a microbial contamination issue, do you know where your final product contaminant comes from and what is the most effective way to get rid of it or prevent it in future? The results from environmental monitoring are used to make ‘risk-based’ decisions about the status of an area or the likely contamination to a product. Riskbased decisions should be made on the basis of trends rather than a reaction to individual out-of-limits results. In assessing contamination risks, the direction of the trend, the level of individual counts and the microbial species are typically included as part of the assessment. The number of identifications depends on the facility, type of product, the intended user and the risk the organisms pose.

Have you done a risk assessment in your facility? To do a proper assessment of possible objectionable organisms, an accurate microbial identification is important. The most important factor when deciding on an accurate identification is considering the databases and if it suits your application. Most identification technologies have a clinical focus with their databases validated to identify clinical specimens. Another important considerations are the need to identify other organisms like yeasts and filamentous fungi and also the important ‘resident bug’ that is unique to your facility.

Are you using an ID system that suits your environment?

Cost is also an important factor and you need to take into consideration the initial investment, maintenance and running costs. The cost over a period of five years and the number of ID’s performed on a daily basis will give you an indication of the overall costs and the payback time for your ID system. Performing the actual identification is another factor. Ease-of-use (performing the ID and also interpreting results), the skills required and hands-on time when doing the ID need to be considered. Validation of the system, technology and consumables are extremely important from a pharmaceutical point of view.

What are you doing with your identification results?

From a production point of view, are the costs of doing identifications on a regular basis higher than the costs of doing no ID’s at all? Performing ID’s and writing reports and having them on file are all good from an auditor’s perspective. The real question is if you can easily access your results to perform trending and tracking of possible objectionable organisms that pose a real risk? BBI Solutions launches innovative conjugate blocking

BI Solutions has launched Morffi™, a novel conjugate blocking technology that enhances signal intensity and improves the sensitivity of lateral flow immunoassays. The new technology was launched at the Lateral Flow Test Development seminar hosted in the US in September. The technology, developed in-house by BBI Solutions scientists, improves the limit of detection of an assay, providing an increase in sensitivity of up to ten times and a faster time to result.

Lateral flow immunoassays Lateral flow immunoassays (LFIs) rely on the specific interaction between the analyte of interest and an immobilised binding partner, such as an antibody, antigen or enzyme. To improve performance, a ‘blocking’ agent is bound to unoccupied conjugation sites on the reporter label, preventing non-specific binding. However, the most commonly used conjugate blocker, BSA cross-reacts with human serum

albumin-binding antibodies, bovine and other animal-origin targets, and exhibits lot-to-lot variability. Its relatively large size can also lead to steric hindrance affecting the detection of the target molecule. The proprietary Morffi technology overcomes these issues. Due to its size in comparison to BSA, it improves the availability of analyte-specific binding partners on the surface of the reporter label, increasing the signal intensity and sensitivity of the assay.

Peter Fuller, Vice-President and Managing Director of Medtronic Africa, suggests that the lack of resources and the migration of skills from the sub-continent has a real impact on access to quality healthcare for millions of people. “We believe that through simulated training and using the latest techniques and equipment, this facility will add value to health systems across the sub-continent through improved clinical management and better patient outcomes, thereby increasing access to quality healthcare,” he said. According to Prof Ian Vlok, Head of the Division of Neurosurgery at the Faculty of Medicine and Health Sciences and collaborator on the Sunskill Laboratory project, most surgical disciplines require a combination of academic knowledge, the ability to perform manually dextrous tasks and a patient-centered ethos. This skillset can only be gained from task repetition observed under close expert supervision. He believes that proficiency gained through Sunskill’s simulated environment, which precisely mimics the real world operating environment, will expose trainees to a spectrum of procedures, and help produce more highly skilled surgeons. “The University requires that a minimum number of procedures within the various disciplines are performed in order to qualify as a specialist. The specialist training programmes will now implement the requirement for regular full theatre simulation sessions

under consultant supervision in the Sunskill Laboratory before proceeding to patients,” said Prof Vlok.

Wet and dry laboratory The dual ‘wet’ and ‘dry’ laboratory is an ideal environment for primary care practitioners, nurses and other health professionals to acquire niche skills such as placing of central venous lines, emergency air-way management and skills required in an ICU setting. The constant introduction of new technology into many fields of medicine requires that even the most experienced specialists are required to up-skill themselves to ensure the use of best-practice techniques in the management of their patients. The Sunskill Laboratory will therefore be made available to those outside of the academic sphere for post-specialist courses on new technologies and methodologies. The laboratory will be updated with new technology to ensure that it remains at the forefront of advancements made in medical science. Prof van Pittius also believes that the Sunskill Laboratory will provide new research opportunities for SU. “To the extent that a university is as good as the research it produces, this laboratory will promote increasing crossdisciplinary work, thereby enabling many previously untapped areas of research,” he noted. Arguably one of the most advanced laboratories of its kind in the region, according to Prof Vlok, the Sunskill Laboratory will attract doctors and experts from across the globe to share their excellence and expertise at dedicated courses and meetings.
