Stage 10 Waiting and monitoring. The mycelial colonisation will take between 4-6 weeks. During this time the temperature of the room will need to be monitored as well as checking for mould. The bags should be sterilised once a week to reduce infection.

Stage 9 Formwork of the unrolled weave geometry of Inoculation 2 is cut and assembled. The process of placing the substrate and grain spawn in the moulds is repeated as specified in Stage 8.

Stage 8 Mycelial growth happens in high humidity, low light and low air change environments. For this reason all models will be covered in polythene sheets to which small incisions will be made to allow adequate oxygenation. All participants must be wearing hair nets, plastic gloves up to the elbow and paper mouth masks. All surfaces to be washed with isopropyl alcohol. Alternative 50mm sterlised substrate and spawn grain layers of a 60/80 ratio is to be placed in moulds until full. Entire mixture to be placed in moulds until full. Large gaps are difficult for the mycelium to bridge and will delay the colonisation process.

Stage 7 Preparation of the polythene sheets that will be used to seal the growing component. These are cut to size and sterilised with isopropyl alcohol, applied with a pulverising spray bottle. Formwork for inoculation is cut to size and assembled. This consisted of four small boxes of 200x250x200mm and two boxes of 300x500x200mm.

Stage 6 During this stage all work needs to be done on sterilised surfaces, operative must wear mask, hair net as well as sleeved gloves to avoid contamination. Substrate is carefully removed from the steel container to the drying surface for any excess water to be drained and allow for cooking for 2 hours.

Stage 5 Substrate pasteurisation. Substrate material will need to be pasteurised prior to being pt in contact with the spawn grain. Substrate and woodchip to be placed in a heavy duty container with 60-80C water. Process to last between 1-1.5 hours. Container to be topped up with hot water throughout process to maintain temperature. Stage 4 Mycelium is mixed with rye-berries and sealed for the purpose of better preservation until the inocluation. This will be kept at a low temperature between 2C and 4C until use. Substrate materials are sourced. Stage 3 Glass grain spawn jars are filled with a high nitrogen organic material. This stage of the process is one of the most likely times for contamination. To avoid contamination grain spawn is washed thoroughly and soaked in water until grain is saturated without excess water. Jars are sterilised in a pressure cooker for one and a half hours at 15Psi.

Stage 2 Place a small piece of flesh in antibiotic agar medium and seal edges. Rhizomorphic mycelium growth should be apparent in 10 days. Again, a small piece of rhizomorphic mycelium is selected from the strongest petri dish. Only the more developed feathery root structure is useful for cultivation. The selected mycelium is then placed in more nutrient rich solution for the final part of the agar cultivation process.

Stage 1 A 3mm section is removed from the inner most part of the fruiting body using a sterile saclple. Work at this condition must be undertaken using a High Effieciency particulate air filter and in completely disenfected conditions.

6a / Inoculation

After the substrate bags have been given adequate time to cool they are mixed with the grain spawn of G. Lucidum. Moulds are then either sealed with a polythene sheet or else placed in a sealed polythene bad. During the mixing a 3% Hydrogen peroxide solution is added to reduce contamination. The bag is also cleaned with alcohol for the same reason.

4 / Substrate Prep 5 / Pasteurisation A substrate with ratio 4:1:.5:.5 of Oak sawdust, straw, calcium carbonate and oat-bran are mixed by hand and hydrated in nylon roasting bags in preparation for pasteurisation.

The plastic bags are placed in a pottery kiln for 3 hours at 100 degrees to kill any competing spores or contaminating germs. The internal temperature of the bags is maintained at a temperature between 60-80 to stop the degradation of cells.

The samples are allowed to ow in their sealed containers -3 weeks until a visible layer mycelium has formed on the top. The pieces are then moved from the moulds and placed in a larger bag to facilitate the aeration of the bstrate and increase growth.

Each of the samples is allowed to air dry. In previous experiments it was found that the rapid dehydration of material can cause unwanted shrinkage and deformation

3 / Mould Prep

For this experiment, a positive for the mould is created by layering cnc MDF. The positive is then formed in plastic using high impact polystyrene in a vacuum former. We were limited in geometry size as the former bed was only 380x380mm.
