RI Performance Tested Methods certification that enables its SureTect assays to be run on the Applied Biosystems 7500 Fast 96-well real-Time PCR instrument.

This is in addition to the option of running the assays on the Thermo Scientific SureTect PikoReal 24-well real-Time PCR instrument for which the assays were originally validated. With SureTect assay methods, molecular pathogen testing is easy, fast and accurate.

Precise, validated results are available within 8-26 hours of sample receipt. While the SureTect PikoReal instrument is a compact 24-well PCR platform with a small footprint that suits laboratories where space is limited, the 7500 Fast instrument enables microbiology laboratories to expand their in-house PCR capability

beyond day-to-day pathogen testing with a versatile, 96-well system that is ideal for batch processing several hundred samples a day. SureTect assay methods combined with the 7500 Fast instrument enables higher throughout and the flexibility to run a broader range of tests. With the SureTect System, food testing laboratories get easy set-up and result-reading, and flexible throughput, from fewer than 10 to several hundred tests per day.

STREAMLINED AND RAPID WORKFLOWS Customers’ needs develop as they strive to meet increasing consumer and legislative demands, while continuing to gain cost efficiencies. For maximum flexibility, the SureTect Assay workflows are validated to run on a choice of two realtime PCR instruments. The Thermo Scientific SureTect PikoReal Instrument: a compact, 24-well instrument ideal for low to medium test throughput and the Applied Biosystems 7500 Fast, a 96-well instrument that can be used to process several hundred samples a day.  Enrichment: Enrich samples in accordance with validated protocols. Single enrichment step of 20 or 22 hours for the majority of sample types.  Lysis : Transfer enriched samples to SureTect lysis tubes. Heat lysis tubes for 10 minutes at 37°C followed by five minutes at 95°C. Transfer lysed samples to SureTect PCR tubes and seal tubes.  Run & Read: Load the PCR tubes into the SureTect PikoReal or 7500 Fast Instrument, start PCR run and read results.  Report or Confirm: Report negative results and confirm any positive results according to validated protocol.  Service and Support: Service and support teams are comprised of highly-experienced scientists who are devoted to offering rapid response, technical support, focusing on providing quality products and ontime delivery. Listeria spp. are gram-positive, non-spore forming, catalase-positive bacteria, shaped like rods that often cluster together to form short chains. Some members of the genus use flagella for motility. They are found widely in the natural environment and thus are potential contaminants in food preparation facilities. Of the different species previously identified, only Listeria monocytogenes is pathogenic in humans, causing a febrile gastroenteritis that is usually mild and self-limiting. Infection rates are low but the disease has a 20% fatality rate in those susceptible, including immunocompromised individuals, the elderly and during pregnancy (mother and fetus). L. monocytogenes causes meningitis in newborns (acquired transvaginally).

It is the third-most-common cause of meningitis in newborns. It can infect the brain, spinal cord membranes and/or the bloodstream of the host through the ingestion of contaminated food such as unpasteurized dairy or raw foods.

Contamination of soil and waterborne bacteria is often Listeria spp. It is found in some animals including poultry and dairy cows, vegetable and fruit, cheese, sausages, or even salmon. In addition, Listeria spp. can grow at low temperature in refrigerators. L. monocytogenes is the important pathogen in food

industry. However, it is terminated by cooking and pasteurization.

ISO STANDARDS FOR LISTERIA To achieve consumer safety, the ISO 11290 series is established. It describes standards of detection and enumeration of L. monocytogenes and Listeria spp.

The standard is applicable to products intended for human consumption, animal feeds, and environmental samples from food production and handling area.

It was updated to current 2017 version. ISO 11290 consists of two sections; ISO 11290-1 and ISO 11290-2.

ISO 11290-1 is the standard of detection of L. monocytogenes and Listeria spp., definition of detected/non-detected in given mass or volume of product or specified surface, when tests are carried out in accordance with the document. Many issues have been resolved in the 2017 version, for example;  Detection method of L. monocytogenes has been modified  Primary enrichment in half-Fraser broth: incubation for 25±1 hr  Secondary enrichment in Fraser broth: incubation for 24±2 hr  CAMP test and catalase test are optional

Five new species of the genus Listeria were discovered, thus bringing the total number of members to 15. Bakker et al. (2014) have established the genomic identity of the isolates from Ion Torrent Personal Genome Machine (PGM) reads and then proceeded with traditional phenotyping to establish classification. L. floridensis sp. nov., L. aquatica sp. nov., L. cornellensis sp. nov., L. riparia sp. nov. and L. grandensis sp. nov., were identified from agricultural and natural environment samples in two US states (Colorado and Florida).

The study has economic implications, since tests to identify Listeria cost about 25 usd each, and large food plants run thousands of tests, spending up to half a million per year. To identify L. monocytogenes, food producers and researchers first look for the presence of more common Listeria spp. But tests may only work to identify Listeria spp. that are very similar to L. monocytogenes.
