It is our great pleasure to welcome alumni, faculty, staff, students, and friends to Penn Dental Medicine’s Research Day 2025.

We invite you to join us in celebrating the breadth and excellence of research at Penn Dental Medicine. This year, once again, our faculty, students, and research staff will come together to showcase the full spectrum of PDM’s research endeavors in a single venue. The program will feature two keynote speakers, faculty presentations, and more than 90 research posters. We look forward to a dynamic day of interdisciplinary exchange and collaboration that highlights the quality, depth, and diversity of the School’s research.

We are delighted to welcome our distinguished guest speakers, Dr. Sumita BhaduriMcIntosh and Dr. Michail Lionakis. Dr. Bhaduri-McIntosh, Professor of Pediatric Infectious Diseases in the Departments of Pediatrics and Molecular Genetics & Microbiology at the University of Florida, honors us as the PDM Research Day Keynote Lecturer. She will present “Epstein-Barr Virus and Oral Health – Mechanistic Connections.” Dr. Michail Lionakis, Senior Investigator and Acting Chief of the Laboratory of Host Immunity and Microbiome; Deputy Chief of the Laboratory of Clinical Immunology & Microbiology; and Chief of the Fungal Pathogenesis Section at the National Institute of Allergy & Infectious Diseases, NIH, will deliver the Joseph L. Rabinowitz Memorial Lecture, “AIRE Deficiency: A Treatable IFN-γ-opathy.” We are thrilled to have them with us and look forward to the insights they will share.

Another highlight of the day is the presentation of the 2025 Joseph and Josephine Rabinowitz Award for Excellence in Research. The Rabinowitz Award supports pilot research among Penn Dental Medicine faculty, with an emphasis on interdisciplinary or interdepartmental collaborations. Established in 1992 by the late Dr. Joseph Rabinowitz, a valued member of the School’s Biochemistry faculty for 29 years, and his wife, Josephine, the award aims to encourage independent research among faculty. We extend a warm welcome and our heartfelt thanks to the Rabinowitz family for joining us.

Research Day will again offer travel grants to attend the IADR/AADOCR/CADR General Session & Exhibition in San Diego, CA, from March 25-28, 2026. This initiative, sponsored by PDM, supports ongoing research and fosters leadership among students and research investigators at Penn Dental Medicine. Five DMD students will be selected to receive the 2026 AADOCR Travel Award.

Penn Dental Medicine continues to advance as a leader in generating new knowledge and training the next generation of oral health researchers. We hope today inspires new collaborations, fresh insights, and innovative discoveries. Enjoy the day!

Sumita Bhaduri-McIntosh, MD, PhD, is a physician-scientist, Professor of Pediatrics, and former Chief of Pediatric Infectious Diseases at the University of Florida. She completed her medical training at B.J. Medical College in Pune, India, and earned a PhD in Molecular and Cell Biology from Hahnemann University.

Following a Pediatric Infectious Diseases fellowship at Yale University, she pursued advanced training in virology and immunology under the mentorship of Dr. I. George Miller. Dr. Bhaduri-McIntosh previously held faculty appointments at Yale University and Stony Brook University before joining the University of Florida as a tenured professor and Children’s Miracle Network Scholar. She also serves as the Editor-in-Chief of PLOS Pathogens.

Her research focuses on Epstein-Barr virus (EBV) interactions with B cells, investigating its lytic activation, viral transcription and replication, and immune evasion in driving B cell proliferation.

Dr. Michail Lionakis is a physician-scientist and Chief of the Laboratory of Clinical Immunology & Microbiology at the National Institute of Allergy and Infectious Diseases (NIAID). His bench-to-bedside research studies the genetic and immune defects of human susceptibility to fungi and the cellular and molecular factors that regulate antifungal immunity.

Dr. Lionakis’ work has defined precise genetic, biochemical, immunologic, and cellular mechanisms that have led to immunotherapies. He identified interferonopathy as the driver of mucosal candidiasis and multiorgan autoimmunity in APECED and uncovered impaired microglial-neutrophil crosstalk as a key mechanism in brain candidiasis associated with CARD9 deficiency. Additionally, he has delineated novel inherited (CARD9 deficiency, STAT3 haploinsufficiency) and acquired (BTK and complement inhibitor-associated) immunodeficiencies that cause to fungal infections.

JOSEPH AND JOSEPHINE RABINOWITZ AWARD FOR EXCELLENCE IN RESEARCH

The Joseph and Josephine Rabinowitz Award for Excellence in Research was designed to help Penn Dental Medicine faculty undertake pilot projects that will enable them to successfully apply for extramural sources of funding. Designed through the lens of a researcher, this ongoing grant evaluates research proposals for their scholarly merit, creativity and innovation; the significance of the research in advancing scientific knowledge; the prospects for future extramural funding; the availability of alternate funding sources; and in the case of junior faculty, evidence that the applicant will be working as an independent investigator and forwarding of the School’s research objectives. It was under development as early as 1994 with the first award presented in 2002.

This award was endowed through the generosity of the late Dr. Joseph “Jose” Rabinowitz, an active member of the School’s biochemistry faculty for 29 years, and his wife, the late Dr. Josephine “Josy,” a fellow Penn alum. Josy’s PhD was in the field of education. Her strong endorsement of education and research provided a hand in the development of this grant. Jose was known for his research in lipid and steroid biochemistry, and made the seminal discovery that HMG CoA was a key intermediate in cholesterol biosynthesis. His research helped lead to the development of the important class of cholesterollowering drugs known as statins.

The Rabinowitz family continues to support the values and priorities of this award and congratulates its newest recipients.

Effect of Western Diet on Retinal Lipoprotein Metabolism and Visual Function

Over a third of adults over the age of 60 in the U.S. experience significant vision loss, which results in decreased independence and also serves as a biomarker of decreased health span. Age-related retinal degeneration is the leading cause of such vision loss, affecting almost 200 million people worldwide. The Age-related Eye Disease Study (AREDS) and follow-up AREDS2 has shown an association between foods containing high fat dietary products, fried and refined food, and those enriched with red/processed meat, often referred to as a “Western-style” diet, with such retinal degeneration. Lipids contained in the Western diet are transported in the circulation and between cells in lipoprotein particles. In these studies, we will test our hypothesis that in the eye, local beta-lipoprotein (Blp) assembly and secretion protects the retina against diet-induced lipotoxicity and degeneration.

Sex Differences in Post-Surgical Pain and Analgesic Response

Pain is one of the most common complications following surgery. While this acute pain typically resolves within days to weeks after a procedure, some patients experience chronic post-surgical pain, which is often underdiagnosed and undertreated. Inadequate pain management in the perioperative period and female sex are important risk factors for the development of chronic post-surgical pain, but the mechanisms underlying these relationships are unknown. We have observed that women are more likely than men to require supplemental opioids after third molar extraction, indicating inadequate analgesic response to nonsteroidal anti-inflammatory drugs (NSAIDs). This project will investigate the mechanisms underlying sex differences in pain and analgesic response, focusing on neuroimmune interactions in a preclinical model of post-surgical pain.

The Bitter Taste Signaling Pathway in Gingival

Macrophages Participates in Anti-inflammatory and Bone Repairing Responses

Periodontal disease affects nearly 50% of US adults over age 30. We previously discovered taste cell-like solitary chemosensory cells in the gingiva (gSCCs) and preclinically demonstrated their protective antimicrobial function against periodontal bone loss. Recently, we observed the presence of the taste signaling pathway also in gingival macrophages (gMΦs). While gSCCs participate in the gingival epithelial barrier by promoting anti-microbial peptide release, gMΦs participate as 2nd level of protection by evoking anti-inflammatory response that promotes bone repair. With this proposal, we will determine the antiinflammatory role of gMΦs in response to bitter compounds in reducing periodontal bone loss. In sum, gMΦs and gSCCs are novel targets for the treatment of periodontal disease and show great potential for a preventive approach by a daily routine of oral care that includes the use of taste receptor activators (e.g. in toothpaste or mouthwash).

THE DARK SIDE OF TRAINED IMMUNITY

A decade after the term ‘trained immunity’ (TRIM) was coined to reflect the long-lasting hyper-responsiveness of innate immune cells with an epigenetically imprinted ‘memory’ of earlier stimuli, our understanding has broadened to include the potential implications of TRIM in health and disease. Here, after summarizing the well-documented beneficial effects of TRIM against infections, I will discuss emerging evidence that TRIM is also a major underlying mechanism in chronic inflammationrelated disorders such as periodontitis, rheumatoid arthritis and cardiovascular disease.

Educational Objectives

• To learn that innate immunity can also build and retain immunological memory of past infectious or inflammatory challenges.

• To learn that inflammatory memory can aggravate osteoclastogenesis and bone loss

• To learn that inflammatory memory can link distinct inflammatory comorbidities

INTER-INDIVIDUAL VARIABILITY IN ANALGESIC RESPONSE TO NON-STEROIDAL ANTI-INFLAMMATORY DRUGS

Numerous clinical trials have shown that non-steroidal anti-inflammatory drugs (NSAIDs) are effective in the treatment of postsurgical pain following third molar extraction. However, there is substantial inter-individual variability in the analgesic response, with 20-30% of patients requiring opioids in addition to NSAIDs to achieve adequate pain relief. The use of precision medicine approaches to tailor analgesic therapy would enable oral surgeons to rationally prescribe opioids to only those patients who require them and avoid unnecessary opioid prescriptions in those patients who can achieve adequate pain relief with NSAIDs alone. This presentation will discuss ongoing research seeking to identify factors that contribute to inter-individual variability in analgesic response to NSAIDs, with the goal of optimizing post-surgical pain management while limiting unnecessary opioid prescriptions.

NEXT-GENERATION MACHINING AND SINTERING TECHNOLOGIES

FOR CERAMIC DENTAL RESTORATIONS

Preventive

With rapid advances in material development and digital technology, time efficiency of dental workflow and fit accuracy of ceramic restorations are ever improving. Zirconia and lithia glass–ceramics are at the forefront of this advance. The trend in fabrication is to move from CAD/CAM grinding partially sintered/crystallized blocks to fully sintered/crystallized materials, thereby circumventing the need for post-machining heat treatment; or to drastically reduce the ceramic sintering time. In these endeavors, a better understanding of ceramic properties and innovative ductile-regime grinding (shaping) and ultrafast sintering (densification) technologies is paramount. Challenges facing the implementation of new technologies in the efficient development and production of high quality dental ceramic prostheses will be addressed.

Educational Objectives

• A facile understanding of the composition, microstructure, and properties of various classes of ceramic restorative materials for the digital workflow.

• The emerging grind-and-seat ceramic restorative systems without the need of any post-shaping heat treatment.

• Novel fabrication methodologies such as ductile grinding and ultrafast sintering offer the prospect of more efficient and longer lifetime ceramic restorations.

Kelly Jordan-Sciutto

Professor, Department of Oral Medicine

Kelly Jordan-Sciutto is Professor of pathology at the University of Pennsylvania. She completed a postdoc in neuropathology at the University of Pittsburgh and received her PhD in biochemistry and molecular biology from Thomas Jefferson University. Her research focuses on understanding how neuroinflammation and organellar stress contribute to neuronal dysfunction and loss in neurodegenerative diseases like HIV-associated brain injury, Alzheimer’s, and Parkinson’s.

Frank Setzer

Frank C Setzer received his dental and doctoral degree from FAU Erlangen-Nuremberg, Germany in 1995 and 1998; and his Endodontic specialty certificate, MSOB and DMD from the University of Pennsylvania in 2006, 2008, and 2010, respectively. He is currently an Associate Professor of Endodontics at the University of Pennsylvania, serving as Predoctoral Director in Endodontics. His research interests are the clinical detection, prognosis and assessment of periradicular pathology, in particular apical periodontitis, utilizing CBCT imaging and Artificial Intelligence, as well as endodontic microsurgery. Dr. Setzer is a Diplomate of the American Board of Endodontics ABE. He is Associate Editor for Journal of Endodontics, and lectures at national and international conferences.
