Dear Friends, Welcome to the 32nd Carousel of Hope Ball. This is a very special evening because we are celebrating the research and advancements we have made for type 1 diabetes. Your presence here tonight helps wonderful doctors and researchers find a cure for diabetes and I thank you so much for your support. These efforts will save the 30 million people in America living with diabetes and the 415 million who are battling this disease all around the world. We have come a long way since we opened the Barbara Davis Center forty years ago and we won’t stop fighting until there is a cure. Tonight we honor a very special man who is an inspiration to the entire movie industry and his genius and philanthropy is known the world over. We are honored to present the Brass Ring Award to Robert De Niro. We are all so mindful of the great philanthropic work he has done that has even enhanced his talents as a great, great actor. We salute you Robert. We thank you for being with us. We thank you for helping us reach for the brass ring, the cure for diabetes and with your help, we will have it even sooner. The disease has doubled over the last twenty years and is projected to double yet again by 2030. Marvin and I began this effort over forty years ago when our daughter Dana was diagnosed with type 1 diabetes. Our dream was to build a clinic with the most advanced ways to treat diabetes, prevent complications, and do research. Today, the Barbara Davis Center for Diabetes has become one of the leading centers for diabetes in the world. The Barbara Davis Center treats thousands of patients from across the nation and around the world regardless of their ability to pay. We are just so grateful for all of you tonight who have joined us in the fight to overcome a disease which leads to so many terrible complications. I thank you from the bottom of my heart for your generosity and for being here to join our battle against type 1 diabetes. Again, I thank my wonderful group of friends. Children’s Diabetes C Foundation (CDF) The Children’s Diabetes Foundation was established by Barbara and Marvin Davis in 1977 in Denver as a non-profit organization dedicated solely to the support of research in diabetes and to provide the best possible clinical and educational programs for people with the disease. The Foundation’s mission is to raise funds to support the Barbara Davis Center for Diabetes, where thousands of patients of all ages from all over the world receive the finest diabetes care available.

C Barbara Davis Center (BDC) The Barbara Davis Center’s mission is to provide state-of the-art care to children and adults with type 1 diabetes and to teach their patients how to prevent or delay complications. Their research is devoted to finding prevention, cure, and most effective treatment of diabetes and associated disorders.

The Brass Ring Award was designed by artist Robert Graham (1938-2008) for the Children’s Diabetes Foundation. It consists of a small child’s hand, catching a brass ring. The award is cast in bronze, and gilded and engraved with the recipient’s name. Robert Graham was born in Mexico City and studied at San Jose State College and the San Francisco Art Institute. He is known for civic monuments, such as the Olympic Gateway 1984 in Los Angeles; Monument to Joe Louis 1986 in Detroit; Franklin Delano Roosevelt Memorial 1997 in Washington DC; Duke Ellington Memorial in 1997 in New York City; Charlie “Bird” Parker 1999 in Kansas City, and the Great Bronze Doors of the Cathedral of Our Lady of the Angeles 2002 in Los Angeles. Robert Graham also designed the National Medal Arts, presented by the President of the United States, and the California Hall of Fame, presented by the Governor. His work is included in the collection of many national and international museums.

obert De Niro has been recognized around the world for his outstanding work in film. In 2009, De Niro received the coveted Kennedy Center Honor, and President Obama presented De Niro with the Presidential Medal of Freedom in 2016, recognizing him for his outstanding contributions to the U.S. He served as the jury president of the 64th Cannes Film Festival and in 2009, received the Stanley Kubrick Award from the BAFTA Britannia Awards. AARP The Magazine gave De Niro the 2010 Movies for Grownups Lifetime Achievement Award and the following year, he was honored with the Cecil B. DeMille Award at the 2011 Golden Globe Awards. He was honored by AFI with their Life Achievement Award in 2003 and two years prior, received the Bravo Lifetime Achievement Award at the Independent Feature Projects’ Gotham Awards. De Niro and the cast of “The Good Shepherd” were awarded the Silver Berlin Bear at the 2007 Berlin International Film Festival. He is the recipient of two National Board of Review Awards, four New York Film Critics Circle Awards, two Los Angeles Film Critics Association Awards, and GLAAD’s 2016 Excellent in Media Award. Robert De Niro launched his prolific motion picture career in Brian De Palma’s “The Wedding Party” in 1969. By 1974, he had won the New York Film Critics Award for Best Supporting Actor in recognition of his critically acclaimed performance in “Bang the Drum Slowly” and from the National Society of Film Critic for Martin Scorsese’s “Mean Streets.” In 1974, De Niro won the Academy Award for Best Supporting Actor for his portrayal of the young Vito Corleone in “The Godfather, Part II.” In 1980 he won his second Oscar, as Best Actor, for his extraordinary portrayal of Jake La Motta in Scorsese’s “Raging Bull.” De Niro has earned Academy Award nominations for his work in five additional films: as Travis Bickle in Scorsese’s acclaimed “Taxi Driver,” as a Vietnam vet in Michael Cimino’s “The Deer Hunter,” as a catatonic patient brought to life in Penny Marshall’s “Awakenings,” in 1992 as Max Cady, an ex-con looking for revenge, in Scorsese’s remake of the 1962 classic “Cape Fear,” and as a father to a bi-polar son in David O. The festival, which just completed its 17th run, was conceived to foster the economic and cultural revitalization of Lower Manhattan through an annual celebration of film, music, and culture; the festival’s mission is to promote New York City as a major filmmaking center and help filmmakers reach the broadest possible audiences. ladys Knight is a seven-time Grammy winner, who has enjoyed #1 hits in Pop, Gospel, R&B and Adult Contemporary, and has triumphed in film, television and live performance. Knight’s career started with a family gospel group known as “Gladys Knight & The Pips.” The group went on to achieve icon status, having recorded some of the most memorable songs of the 60s, 70s, and 80s. Top 20 hits, like “Every Beat of My Heart,” “Letter Full of Tears,” “I Heard it Through the Grapevine,” and “If I Were Your Woman,” set the stage for an amazing run in the mid-1970s, with Top 10 goldcertified singles like “Neither One of Us (Wants to be the First to Say Goodbye),” “I’ve Got to Use My Imagination,” “Best Thing to Ever Happen to Me,” and the #1 smash “Midnight Train to Georgia.” Knight enjoyed another #1 hit in 1985 when she teamed with Stevie Wonder, Elton John and Dionne Warwick on “That’s What Friends are For.” In 1995, Knight earned her star on the Hollywood Walk of Fame, and the next year, Gladys Knight & The Pips were inducted into the Rock ‘N’ Roll Hall of Fame. Knight published an autobiography, “Between Each Line of Pain and Glory” in 1997 and the next year, she and The Pips were presented with the Lifetime Achievement Award from the Rhythm & Blues Hall of Fame. In 2004, Knight received the “Lifetime Achievement Award” at the annual BET Awards ceremony. Knight has recorded more than 38 albums over the years, including four solo albums during the past decade: “Good Woman” (1991); “Just for You” (1994); “Many Different Roads” (1999); and “At Last” (2001). In 2015, she released her first mainstream dance record, “Just A Little,” in nearly twenty years. Knight, who is a two-time Grammy winner in the gospel category, released “Where My Heart Belongs” in 2014. The album won an NAACP Image Award for “Outstanding Gospel Album.” In 2011, Knight joined such performers as Jennifer Hudson, Beyonce, and Smokey Robinson in a tribute
concert to the legendary King of Pop, “Michael Forever.” Knight was then honored with a “Legend Award” at the 3rd annual Soul Train Awards. Knight returned to the Las Vegas Strip in the late2000s for a special engagement that ran in the newly named Gladys Knight Theater, making her the first African-American performer to have a venue named after her in Las Vegas. This followed a successful fouryear show run at The Flamingo, which the Las Vegas Review-Journal praised as “the number-one show on the Strip.” In February 2011, Knight reunited with Elton John, Dionne Warwick, and Stevie Wonder for the first time in 25 years to perform their Grammy-winning song, “That’s What Friends Are For.” Knight won another Grammy for her duet with the late Ray Charles on his posthumous album, “Genius Loves Company” (2005). The duo won for Best Gospel Performance for their duet “Heaven Help Us All.” Knight’s solo album, “At Last” also won a Grammy for Best Traditional R&B Vocal Album in 2002. She is a wife, mother, grandmother, great-grandmother, performer, restaurateur, and businesswoman with a spiritual outlook on her life. enny “Babyface” Edmonds’ award–winning, multi-faceted pop/urban career includes the consecutive multi-platinum landmark albums Tender Lover, For the Cool in You (including signature hits “Never Keeping Secrets” and “When Can I See You”), and The Day (”Every Time I Close My Eyes”). As co-founder with L.A. Reid of LaFace Records in 1989, Babyface nurtured the careers of such megastars as TLC, OutKast, Usher, and Toni Braxton. An 11-time Grammy Award® winner, Babyface most recently released Return of the Tender Lover in December 2015, his first solo album in seven years. eorge Schlatter is a veteran of over 50 years in network television. Throughout his long and successful career as a producer, director and writer, George Schlatter has been responsible for hundreds of hours of television series and specials. He changed the face of television when he created and produced such breakthrough series as “Laugh-In” and “Real People.” Over the years, Mr. Schlatter has received numerous honors and awards including 25 Emmy Award nominations, five Emmys, three Image Awards, Golden Globe Awards, Television Critics Awards, the International Radio & Television “Man of the Year” Award, Directors Guild Award, Producers Guild “Man of the Year” Award, as well as many others. When the Television Academy celebrated its 15th Anniversary, they honored him for his outstanding contribution to television. In 1989, he was awarded a star on Hollywood Boulevard’s Walk of Fame. In 1992 Paul Newman and Frank Sinatra, on behalf of the Scott Newman Center, honored him with a tribute dinner for his showmanship and involvement with so many charitable causes. In 1995, the Museum of Television & Radio did a special tribute to him for his contributions in the world of television. He was also honored by his alma mater, Pepperdine University, with a three-hour gala at the Saban Theater hosted by many of the stars with whom he has worked over the past 50 years. In 1987 he created and established the “American Comedy Awards,” which aired for 15 straight years as annual network televised events designed to acknowledge the contributions and achievements of comedic actors and performers. “Of all the work I’ve done, I am perhaps most proud of my involvement in the early careers of performers like Goldie Hawn, Lily Tomlin, Roseanne, Robin Williams, Ellen DeGeneres, Flip Wilson, and others who have gone on to greater heights. Working with stars is rewarding, but helping to create stars is the most fulfilling of all accomplishments.” This year he was invited to participate in the opening of the National Comedy Center in Jamestown, New York. He produced the first 5 years of the Grammy Awards plus CBS television series with Cher and with Bill Cosby, as well as “The Judy Garland Show” and all the way back to the “Dinah Shore Chevy Show” on NBC. He produced specials starring Frank Sinatra, Judy Garland, Eddie Murphy, Elton John, Bette Midler, Goldie Hawn, Liza Minnelli, Cher, Michael Jackson, Doris Day, Jonathan Winters, Richard Pryor, Shirley MacLaine, Lucille Ball, Jackie Gleason, Diana Ross, Lena Horne, Placido Domingo, Nat King Cole, and scores of others. Mr. Schlatter and Whoopi Goldberg co-produced a special based on the life of Moms Mabley, which included interviews with Sidney Poitier, Quincy Jones, Tommy Smothers, Eddie Murphy, Arsenio Hall, Kathy Griffin and many others. He also has produced a “Laugh-In” reunion show for a PBS pledge drive, which was the most successful comedy special ever for PBS. His credits also include The American Film Institute Life Achievement Award Salutes to Dustin Hoffman and Harrison Ford, The 54th and 55th Presidential Inaugural Opening Ceremonies of George W. Bush, the Juvenile Diabetes Research Foundation Inaugural Gala: Finding a Cure, and Nancy Reagan: A Love Story. He has been working on a documentary dealing with Dr. William Rader and his successful use of Fetal Stem Cells in treating many debilitating diseases. For twenty years, he has produced all of Barbara Davis’s Carousel of Hope events, which created the Barbara Davis Center for Diabetes in Denver. He is particularly proud of the work he did in helping Barbara Sinatra create her center for the treatment of sexually abused children at the Eisenhower hospital in Palm Springs. He is married to former actress Jolene Brand who was a regular on the Ernie Kovacs Show. They have two daughters, Andrea and Maria. 17

When he’s not on TV, Leno performs more than 100 stand-up shows annually across the country and internationally, participates in numerous charity and corporate events, does voice-overs for animated series, pens children’s books, and writes a monthly column in various automotive magazines. And in his “spare” time (as if he has any), he enjoys working on his collection of classic cars and motorcycles. In fact, he has built a number of cars, including an acclaimed eco-car in his eco-friendly green garage. Leno is the recipient of many honors, including Emmy, People’s Choice, and TV Guide awards, Harris Poll selection as most popular star on television, the Hasty Pudding Award at Harvard University, The Mark Twain Prize, a star on Hollywood’s Walk of Fame, and being the first person to drive the pace car of all major NASCAR events. s the record industry’s most influential executive, Clive Davis has had a profound effect on the world of music. Successively head of Columbia Records, Arista, and the RCA Music Group, Davis now serves as Chief Creative Officer of SONY Music Entertainment. Davis has signed and nurtured such performers as Janis Joplin, Blood, Sweat and Tears, Santana, Aerosmith, Chicago, Billy Joel, Bruce Springsteen, Whitney Houston, Barry Manilow, Patti Smith, Alicia Keys and Jennifer Hudson. And over the years, he has played a pivotal role in the major revival of the careers of Aretha Franklin, Dionne Warwick and Rod Stewart. Mr. Davis’ passion for music is matched by his passion for helping others. Clive Davis began his tireless and ongoing battle against HIV and AIDS in 1985 and brought together Stevie Wonder, Elton John, and Gladys Knight with Dionne Warwick to record the single “That’s What Friends Are For,” a #1 smash hit which raised over $2 million dollars for the fight against AIDS. In March 1990, Davis transformed the “Arista 15th Anniversary Celebration” at Radio City Music Hall into an AIDS Benefit Concert raising another $2 million dollars. A decade later, the “Arista 25th Anniversary Celebration” aired on NBC in May 2000 raising AIDS awareness and funds for amfAR, the City of Hope, and the T.J. Martell Foundation. He was twice named Humanitarian of the Year by the T.J. Martell Foundation – an honor solely accomplished by Mr. Davis. In January 1997, Clive Davis became the first active record company president to receive a star on the Hollywood Walk of Fame. In the year 2000, Clive Davis was inducted into the Rock and Roll Hall of Fame and was awarded by NARAS the Grammy Trustee Award for Lifetime Achievement. The Tisch School of the Arts at New York University and Clive Davis announced his $10 million gift to the School for the creation of The Clive Davis Institute of Contemporary Music, the first program to offer a four-year, degree-granting undergraduate program of its kind. He has also been saluted by the New York Landmarks Conservatory as a “Living Landmark” and he has received the coveted NARAS Heroes Award. Born in Brooklyn, New York, Clive Davis was a Phi Beta Kappa graduate of New York University, where he received his B.A. magna cum laude and he graduated with honors from Harvard Law School. amed by Time Magazine as one of the six most influential Jazz artists of the 20th Century, Quincy Jones is an impresario in the broadest and most creative sense of the word. Over six-decades, Jones’ career has encompassed the roles of composer, record producer, artist, film producer, arranger, conductor, instrumentalist, TV producer, record company executive, television station owner, magazine founder, multi-media entrepreneur, and humanitarian. Among the multitude of awards that he has received for his contributions are an Emmy Award, seven Academy Award nominations, the Academy of Motion Picture Arts and Sciences’ Jean Hersholt Humanitarian Award, 27 Grammy Awards, and is the all-time most nominated Grammy artist with a total of 79 nominations. He is the recipient of the Republic of France’s Commandeur de la Legion d’ Honneur and in 2001, he was inducted as a Kennedy Center Honoree, the United States’ most prestigious cultural award, for his lifetime contributions to the culture of the country. He was recognized by the National Endowment for the Arts as a Jazz Master - the nation’s highest jazz honor and in 2010 was bestowed the National Medal of Arts, our nation’s highest artistic honor. In 2016, Jones received a Tony Award for Best Revival of a Musical for the Broadway production of “The Color Purple.” The award completed the rare EGOT set for Jones, an exclusive club of artists who have received an Emmy, Grammy, Oscar and Tony Award. In 2001, Quincy Jones added the title “Best Selling Author” to his list of accomplishments when his autobiography “Q: The Autobiography of Quincy Jones” entered the New York Times, Los Angeles Times, and Wall Street Journal Best-Sellers lists. Released by Doubleday Publishing, the critically acclaimed biography retells Jones’ life story from his days as an impoverished youth on the Southside of Chicago through a massively impressive career in music, film, and television where he worked beside legends such as Billie Holiday, Ray Charles, Dizzy Gillespie, Count Basie, Ella Fitzgerald, and Michael Jackson, among many others. In conjunction with the autobiography, Rhino Records released a 4-cd boxed set of Jones’ music, spanning his more than 5 decade career in the music business, entitled “Q: The Musical Biography of Quincy Jones.” In 2008, he released the coffee-table book “The Complete Quincy Jones: My Journey and Passions.” In 2010, Jones released his latest record Soul Bossa Nostra, an album featuring some of the day’s biggest recording artists and producers such as Usher, Ludacris, Akon, Jamie Foxx, Jennifer Hudson, Mary J. A longtime humanitarian and activist, in 1999, Quincy Jones joined Bono and Bob Geldof during a meeting with Pope John Paul II as a part of the Jubilee 2000 delegation to end third world debt. The delegation’s visit resulted in $27 billion in third world debt being relieved. Through his Project Q Foundation, a joint initiative with the Harvard School of Public Health, Jones is actively working with NGO’s such as UNICEF, the Clinton Hunter Development Initiative, and the Inter-American Development Bank, among others, to improve the health and well being of millions of children in developing countries such as Rwanda, South Africa, and Asia. Jones is also the founder of the Quincy Jones Music Consortium, a cooperative of musicians, music and arts educators, corporations, foundations, and philanthropists tasked with developing an innovative curriculum to be implemented in America’s schools that explores, celebrates, and preserves the history of American Popular Music so that music education can be an ongoing part of the lives of the children of the United States.

haron Stone began her career in New York, working as an internationally successful model in print and as an actor appearing in network television. The Symposium:
Challenges in Implementing Diabetes Technology and Detecting Biomarkers for Type 1 Diabetes The 2018 Carousel of Hope (COH) symposium’s first session focused on emerging therapies and diabetes technologies for improving healthcare outcomes. There is emerging data that non-insulin adjunctive therapies, especially SGLT1 & 2 inhibitors (which do not require insulin for their action profile) may become available as a possible therapeutic choice for patients with type 1 diabetes (T1D). The data presented in the literature and in the leading conferences (ADA, EASD, and ATTD) clearly highlight that these therapies improve glucose control without increasing severe hypoglycemia and are usually associated with significant weight loss. The two drugs in this category, dapagliflozin and Sotagliflozin, have been filed with the FDA and EMA for their possible approval as adjunctive therapies for treating patients with T1D. There are approximately 150-200 million people worldwide needing insulin therapy for managing their diabetes. About 90% of them are patients with type 2 diabetes (T2D). Only about 0.5–1% are using some sort of insulin pump therapy despite the approval of the first artificial pancreas by the FDA in September of 2016. This was shared by Satish Garg. The introduction of newer therapies in T1D also pose a possible risk of increased episodes of diabetic ketoacidosis (DKA). Anne Peters talked about how best to mitigate risk of DKA in T1D. The difficulty in implementing new technologies may in part be due to the cost, resources, and challenges in implementing technologies in real life as highlighted by Irl Hirsch. The landscape for continuous glucose monitoring (CGM) has completely revolutionized how these technologies will be used in real life. In March 2018, the FDA approved the first factory-calibrated CGM that could be used for 10 days. In addition, they also created a new 510K pathway for possible approval for future CGMs, if they can satisfy the strict guidelines as laid out by the FDA. These were discussed in detail by Andy Ahmann and Jake Leach. The basic science research highlighted the DNA biomarkers of beta cell stress in T1D and protein biomarkers for beta cell stress. It is also important to understand that biomarkers could be used in clinical practice as indicators of response to T1D therapies. These were discussed at length by Lori Sussel, Marian Rewers, and Tom Metz. Satish K. Garg, MD, Professor of Medicine and Pediatrics, Adult Clinic at the Barbara Davis Center for Childhood Diabetes, University of Colorado, Aurora, CO Dr. Garg’s research interests include the development of new diagnostic and therapeutic tools related to clinical diabetes management that include new insulin analogs and glucose sensors, artificial pancreas studies in subjects with type 1 diabetes, evaluating accuracy of meters at high altitude, alternate ways to deliver insulin, adjunctive therapies for T1D, etc. He has been the Director of the ATDC Keystone Diabetes Conference since 2015. Dr. Garg is an international lecturer and speaker, has published many book chapters, serves on the editorial boards for many global diabetes journals, published more than 235 original manuscripts in peerreview journals, and is the Editor-in-Chief of the Diabetes Technology and Therapeutics Journal.
Jesper Gromada, PhD, Vice President at Regeneron, Tarrytown, NY Dr. Gromada is Vice President at Regeneron Pharmaceuticals and oversees their diabetes, metabolism, and muscle research. Dr. Gromada has a long interest and contribution to the diabetes field as reflected in the development of several diabetes drug candidates, one diabetes medication, and over one hundred publications.
Irl Hirsch, MD, Professor of Medicine at University of Washington, Seattle, WA Dr. Hirsch is a Professor of Medicine and holds the Diabetes Treatment and Teaching Chair at the University of Washington. For the last 30 years, his research has focused on new treatments for type 1 diabetes, better understanding of HbA1c, inpatient diabetes management, and cost-effective use of insulin.
Jake Leach, Senior Vice President, Research and Development at Dexcom Inc., San Diego, CA Mr. Leach was promoted to his current position in 2015, where he is responsible for the leadership of scientific research, engineering, product development, and project management. Jake oversees the development of next generation products and leads a large organization of amazing people. His teams are responsible for delivering best-in-class glucose monitoring technology paired with an exceptional user-experience. Mr. Leach joined Dexcom in March 2004 to lead development of sensor electronics, which were part of the first generation Dexcom system. Jake has served in various roles within Dexcom, including Vice President of R&D, Senior Director of R&D, and Manager of Engineering. From 1996 to 2004, Jake held positions in research and development at MiniMed and subsequently Medtronic Diabetes, focusing on the development of glucose sensing systems. 34
Thomas Metz, PhD, Integrative Omics, Team Lead Scientist at Pacific Northwest National Laboratory, Richland, WA Throughout Dr. Metz’ career, he has been most interested in the study of disease and has approached this broad interest through the application of mass spectrometry-based techniques. Currently, Dr. Metz leads a group of scientists at Pacific Northwest National Laboratory whose focus is on development and applications of high throughput methods for various biological questions. He is the Principal Investigator of a number of grants and cores involving studies of disease, including type 1 diabetes mellitus.
Anne Peters, MD, Director, USC Clinical Diabetes Program, Professor of Clinical Medicine at Keck School of Medicine of USC, Los Angeles, CA Dr. Peters runs diabetes centers in Beverly Hills and in underserved East Los Angeles. In addition to her clinical work, she has been a Principal Investigator on multiple grants and has written over 200 abstracts and publications and 4 books. Her major interests involve translating research findings, from lifestyle interventions to technology, to people with diabetes throughout the socioeconomic spectrum. She was presented with the ADA “Outstanding Physician Clinician” award, the Bernardo Alberto Houssay Award from the National Minority Quality Forum, and has been listed on the “Best Doctors in America” continuously since 2001.
Marian Rewers, MD, PhD, Professor of Pediatrics and Medicine, Executive Director at the Barbara Davis Center for Childhood Diabetes, University of Colorado School of Medicine, Aurora, CO Dr. Rewers is a pediatric diabetologist and an internationally-recognized scientist. He has made important contributions to our understanding of the etiology of type 1 diabetes and its macrovascular complications by creating large prospective cohort studies: DAISY, CACTI, and an intentional consortium, “The Environmental Determinants of Diabetes in the Young (TEDDY).” His Autoimmunity Screening for Kids (ASK) program at the Barbara Davis Center is screening children for early diabetes and celiac disease to prevent or treat these diseases as soon as possible.
Lori Sussel, PhD., Director of Research at the Barbara Davis Center for Childhood Diabetes, University of Colorado School of Medicine, Aurora, CO Dr. Sussel’s research is focused on understanding how to generate and maintain functional insulin-producing beta cells. Paul Wadwa, MD, Associate Professor of Pediatrics at the Barbara Davis Center for Childhood Diabetes, University of Colorado School of Medicine, Aurora, CO Dr. Wadwa is the Medical Director of the pediatric clinic and Director of Telemedicine. His primary research focus is on development of the artificial pancreas. He is engaged as a site Principal Investigator and a coinvestigator for several NIH and industry-sponsored clinical research studies with the goal of advancing artificial pancreas technology. Imagine a disease that occupies your thoughts all day long, without a break and requires hundreds of life-threatening or life-saving decisions a day – THIS IS TYPE 1 DIABETES Over 40 years ago, when the Children’s Diabetes Foundation and the Barbara Davis Center were created, diabetes looked like a completely different disease. Home blood testing didn’t exist, synthetic insulins were unheard of, and you had to boil syringes in water before injecting large needles. Today, we have artificial pancreases, continuous glucose monitors, a choice between insulin pumps or injections, and so many other exciting medications and therapies that make diabetes management fit into the patient’s life, not the other way around. The Barbara Davis Center is a huge contributor to these advancements as they continually work on cutting-edge technology and medications. But sadly, we haven’t reached the finish line. Diabetes requires constant management and the physical, emotional, and social stress that diabetes can cause is ever-present in a diabetic and their family’s lives. The Children’s Diabetes Foundation and the Barbara Davis Center are fighting to improve patient lives every day by providing the best possible patient care, providing support groups, educating the public, and doing research to prevent diabetes, reduce its complications, and eventually find a cure.
The people who are affected by type 1 diabetes aren’t just patients. They are our heroes and we will help them every step of the way.

Diabetes Technology Research In the past few years, diabetes technology has made incredible strides and 2018 has created even more technology that will improve life for people with type 1 diabetes. The very first Artificial Pancreas (a system that combines an insulin pump and continuous glucose monitor that aims to keep blood sugars in target range by automatically calculating insulin doses) is now available for the masses, but multiple companies are now in the race with differing features that will improve diabetes care and lessen the burden of diabetes.
Artificial Pancreas
The Barbara Davis Center is doing research for multiple Artificial Pancreas systems currently in development. This gives them the unique opportunity to provide feedback to the manufacturers to ensure that patients are receiving the absolute best technology. The research done at the Barbara Davis Center also contributes to the FDA’s decision on whether or not to approve these future devices for consumers.
Continuous Glucose Monitors (CGM) A Continuous Glucose Monitor checks a patient’s sugar level every 5 minutes through a tiny sensor that stays on the body for several days. The device displays graphs to show how quickly blood sugar levels are rising or falling, and alarms when blood sugar levels are too high or low, a feature that is particularly helpful when the patient is sleeping. The device is truly remarkable and even allows a parent to see their child’s blood sugar when the child is at school or in a different state. This year, research is continuing to make these devices smaller, more accurate, and more user-friendly. As more companies develop systems with unique features, patients will be able to choose the product with features that best fit their needs.

BDCC Is One of Few labs in the World Making Insulin-Producing Beta Cells from Stem Cells Stem cell researchers at the Barbara Davis Center are now able to make insulin-producing beta cells derived from human stem cells on a large-scale. There are only a few labs in the world that are capable of this feat and the hope is to use these cells to treat type 1 diabetes patients. Several hurdles remain before cell replacement therapy can become a reality. Generated beta cells need an efficient delivery method, as well as effective protection to keep the patient’s immune system from destroying the cells. Dr. Holger A. Russ and his colleagues discovered that more than half of all beta cells die shortly after being transplanted. The lab is now working on strategies to improve the cell survival rate and protect them from autoimmune destruction to eventually use this cell therapy method to cure patients of diabetes. Dr. Holger A. Russ is currently an Assistant Professor at the Barbara Davis Center for Diabetes. His research focuses on the function of the human pancreatic beta cell, its interaction with the immune system, and how autoimmune type 1 diabetes develops. Dr. Russ and his team use this basic knowledge to develop novel strategies for cell therapy approaches to combat diabetes.
The Latino Program The Latino Program serves around 100 Latino patients and over 300 family members. Latino patients with type 1 face specific challenges to care related to cultural and language barriers, which puts them at high risk for poor diabetes control. The program spearheaded by Dr. Andrea Gerard Gonzalez, ensures that patients and their families are given care in a group of families with patients of similar ages and that all care and educational materials are given in their preferred language, which are adapted to be culturally appropriate. Almost all providers in the Latino Program are native speakers and are able to connect to patients through language and shared cultural values. 39
The Cost of Diabetes When a patient is diagnosed with type 1 diabetes, the start-up education and appointment fees total $1,400. Insulin is absolutely necessary for a type 1 to survive and can cost $300 per month for children and $600 per month for adults. Type 1s need to test their blood sugar 4 to 10 times a day. Test strips are needed to do this and can cost up to $70 per week. Insulin pumps allow a patient to get 1 needle every 2-3 days instead of multiple times a day and can give small doses to manage type 1 diabetes more minutely. The start-up costs for an insulin pump can be up to $6,500. Supplies for the insulin pump cost up to $1,200 per month. The Artificial Pancreas requires patients to start on an insulin pump and then upgrade to the automated version. This upgrade costs $2,500. Continuous Glucose Monitors (CGMs) test a patient’s blood sugar every 5 minutes and give an immense amount of detail to better control blood sugar levels. Supplies for a CGM can cost $500 for a three month supply. When blood sugars are too high, a patient may go into a dangerous physical state known as Diabetic Ketoacidosis. A hospital stay for this can easily cost $10,000. If blood sugars are too low, a patient may go unconscious, have a seizure, or in extreme cases, die. Glucagon kits are the last resort to save a patient when they are not able to chew or swallow carbs and sugars to raise their blood sugar. These kits cost $300 per year and need to be replaced every 12 months.

MEANWHILE, WE ARE SEARCHING FOR A CURE‌ Research at the Barbara Davis Center costs about $1,000 per hour for expert staff and state-of-the-art equipment. Stem cell research is looking incredibly promising in ways of a cure, but $35,000 is required to produce enough beta cells for one patient. $75,000 will support a post-doctoral fellow for one year for clinical care at the Barbara Davis Center. $100,000 is needed to establish a new, groundbreaking research project for one year. 41

he Barbara Davis Center for Diabetes (BDC) is one of the largest centers in the world specializing in type 1 diabetes research and care for children and adults. Clinicians, clinical researchers, and biomedical scientists work at the BDC to find the most effective treatment, prevention, and cure for type 1 diabetes. The center’s clinics provide state of the art care for over 7,000 children and adults with diabetes. Barbara Davis Center clinics offer extensive education and support for patients and their families, as well as specialized programs such as the Pregnancy and Diabetes Clinic, the Hispanic/Latino Diabetes Care Program, and a model Telemedicine program servicing residents in Wyoming and remote parts of Colorado. BDC research goals include investigation of the causes of type 1 diabetes, the early detection of autoimmunity, prevention, and early intervention. BDC clinical faculty members are developing new strategies and treatments for improved outcomes of care, including prevention of complications of both type 1 and type 2 diabetes. Investigators of the BDC receive more than $6 million in research funding per year from the Children’s Diabetes Foundation, and other federal, foundation, and industry sponsors. Research areas include: • Artificial Pancreas/Hybrid Closed Loop Systems: The evaluation of devices for continuous glucose monitoring and insulin pump therapy and their combination with computer algorithms designed to adjust insulin delivery • Finding the Cause of Diabetes: The investigation of environmental and immunogenetic causes of type 1 diabetes and development of tests to predict the disease • Finding a Cure: The development of novel ideas and discoveries in the immunology, genetics, and cell biology of diabetes
The BDC Research Division provides cytometry and islet preparation services for local clientele. The Molecular Biology Service Center provides basic molecular biology support, DNA sequencing and cell line authentication. The Autoantibody/HLA Service Center performs assays for islet autoantibodies and markers of other autoimmune disorders, including celiac and Addison’s disease. This laboratory serves as the core laboratory for numerous national and international trials for the prevention of type 1 diabetes. The BDC serves as a model for combining basic and clinical investigation and is an outstanding training environment for developing physician-scientists, clinicians, and basic science researchers. Faculty members provide laboratory and clinical research training opportunities for young investigators from around the world, including participants in the highly successful Pediatric Endocrinology Fellowship Program and Pediatric Endocrinologist Career Development Program. The annual Keystone Conference remains the Center’s flagship in the area of continuing medical education (CME) in Management of Diabetes, regularly selling out with over 500 participants. What is Diabetes?
iabetes is an autoimmune disease that afflicts 30 million people in the United States, about 1.25 million of whom have type 1 diabetes (also known as childhood, juvenile, or insulin-dependent diabetes). Approximately 40,000 people are diagnosed with type 1 diabetes every year and this number continues to rise. You have probably heard of insulin and glucose, but how do they work? After you eat a meal, food is broken down into glucose and nutrients, which are absorbed into the blood stream. This causes sugar levels in the blood to rise, which triggers the pancreas to make the hormone insulin and release it into the bloodstream. In people with diabetes, the body either can’t make or can’t respond to insulin properly. Because of this, people with type 1 diabetes take artificial insulin throughout the day to counteract high blood sugars. They take the medicine through multiple doses of insulin every day, either through injections or an insulin pump worn on the body. While these measures are certainly helpful for maintaining a steady blood sugar, they are not a

perfect science. There are many variables that go into diabetes management, such as diet, exercise, hormones, stress levels, and countless others. Another concern for diabetics is low blood sugar levels, or hypoglycemia. Symptoms of hypoglycemia include blurred vision, shaking, sweating, slurred speech, irritability, drowsiness, and acting incoherent or spacey. If the blood sugar is severely low, the person may become unconscious or have a seizure. These low blood sugars put a diabetic in immediate danger. Type 1 diabetes means being connected to multiple devices 24 hours a day, 7 days a week. Many people use an insulin pump, which is dispensing artificial insulin throughout the day, as well as a Continuous Glucose Monitor, which measures blood sugar levels every 5 minutes to show trends and reduce finger pricks. These devices are extremely helpful for diabetes management, but often require more needles and devices to be worn. While they usually improve blood sugar levels, these medical supplies come at a huge expense for the patient or family. Unfortunately, there is no cure for diabetes. Each child who is diagnosed with type 1 diabetes will have the disease for the rest of his or her life, unless we can find a cure. The risk of developing diabetes is higher than virtually all other childhood, chronic illnesses. It is estimated that one out of every three babies born today will have diabetes in their lifetime. For this reason, the Barbara Davis Center continues to improve diabetes care and search for a cure for diabetes.

It is not recommended to calibrate your CGM device when sensor or blood glucose values are changing rapidly, e.g., following a meal or physical exercise. If a control solution test is out of range, please note that the result may be transmitted to your pump when in the “Always” send mode.
